Chinese Journal of Experimental Ophthalmology 2013;(1):49-54
Background Vitronectin is a glycoprotein that has a variety of functions. Its expression was markedly higher in the retina of oxygen induced mice, which was confirmed in our animal model, and also increased in human umbilical vein endothelial cells (人 UVECs) that were cultured in high glucose. However, there was no evidence that showed vitronectin was involved in retinal neovascularization. Objective This study was to observe the influence of vitronectin on cytoskeleton remodeling, cell migration and blood vessel formation in 人 UVECs conditioned by high glucose. Methods 人 UVECs were cultured in high glucose and the expression of vitronectin was knocked down using RNA interference technology. The experiments were divided into the high glucose group (人 UVECs were conditioned with DMEM medium that contained 50 mmol/L glucose),negative interference group (人 UVECs were transfected with control siRNA in advance, and then were conditioned with DMEM medium that contained 50 mmol/L glucose) and positive interference group (HUVEC were transfected with vitronectin siRNA in advance, and then were conditioned with DMEM medium that contained 50 mmol/L glucose). The protein expression of vitronectin was measured by Western blot, and the microfilament cytoskeleton of 人 UVECs was examined by immunofluorescence cytochemical staining followed by fluorescence microscopy.Cell migration ability in a scratch wound assay and blood vessel formation ability in a matrigel assay of 人 UVECs were evaluated. The general differences were analysed by One-Way ANOVA ;further contrasts of the two groups were analysed by the LSD-t test. Results The differences in vitronectin expression of the three groups were not obvious at 0 hour (F=1.064,P>0.05).After 24 hours, vitronectin expression was highest in the high glucose group, lower in the negative interference group, and the lowest in the positive interference group, and the differences were significant (F =15.519,P<0.05).After 48 hours, vitronectin expression of the three groups displayed the same pattern, and the differences were also significant (F=37.521,P<0.05).Immunofluorescence showed that the cytoskeleton structure was most obvious in the high glucose group, moderate in the negative interference group, and was the least obvious in the positive interference group, after both 24 hours and 48 hours. In the scratch wound assay, the cell migration ability of the high glucose group was the highest, lower in the negative interference group, and the lowest in the positive interference group after 24 hours, and the differences were significant (F=90.685,P<0.05).After 48 hours, the cell migration abilities of the three groups displayed the same pattern, and the differences were also significant (F=67.880,P<0.05).In the matrigel assay, after 6 hours, the number of blood vessels formed in the high glucose group was more than that in the negative interference group, and the least amount was found in the positive interference group. The differences among of them were significant (F =86.653,P<0.05).The number of blood vessel formed in the positive interference group was also the lowest after 12 hours, and the differences were also significant (F=18.992,P<0.05).Conclusions Vitronectin can bring about cytoskeleton remodeling, increase in cell migration, and enhancement of blood vessel formation in 人 UVECs conditioned in high glucose. It may be one of the important influence factors of diabetic retinopathy.

OBJECTIVETo discuss operative methods and effects for Müller-Weiss disease.
METHODSFrom March 2005 to May 2011, 15 patients were operated. There were 2 males and 13 females, with an average age of 51.8 years (range, 26 to 62 years). The preoperative American Orthopaedic Foot and Ankle Society (AOFAS) ankle-hindfoot score was (42 ± 13) points. According to the Maceira Staging system, 1 foot was grade 2, 6 feet were grade 3, 4 feet were grade 4, and 4 feet were grade 5. The technique consisted of arthrodesis of the talonaviculocuneiform joints with plate, arthrodesis of the talonavicular joint and arthrorisis of naviculocuneiform joint with plate, or triple arthrodesis with screws, according to the preoperative evaluation.
RESULTSTen feet were treated with arthrodesis of the talonaviculocuneiform joints, 3 feet were treated with arthrodesis of the talonavicular joint and arthrorisis of naviculocuneiform joint and 2 feet were treated with triple arthrodesis. Two patients were lost to follow-up. The average follow-up after operation was 19.2 months (range, 9 to 38 months). All feet were solid fusion and the average duration of union was 13.3 weeks (range, 12-16 weeks). The AOFAS ankle-hindfoot score at the last follow-up was (83 ± 6) points. Two feet were excellent, 10 feet were good, and 1 foot was fair. The length of feet was (14.2 ± 1.0) cm before surgery and (15.7 ± 0.9) cm at the last follow-up (t = 11.570, P < 0.05). The Meary's angle was -6.1° ± 13.1°before surgery and 1.1° ± 3.0° at the last follow-up (t = 2.248, P < 0.05). The talocalcaneal angle was 4.5° ± 2.2° before surgery and 18.0° ± 4.0° at the last follow-up (t = 11.700, P < 0.05). One foot had breakage of the plate and screws at arthrorisis of naviculocuneiform joint and none had complications related to the incision.
CONCLUSIONOperations for Müller-Weiss disease, according to concrete conditions using different therapeutic program, may achieve a satisfactory outcome.

Chinese Journal of Surgery 2011;49(8):737-740
OBJECTIVESTo provide a cumulative data about the complications of second or third generation ankle prostheses in the literature, and to provide a summary high-grade complications associated with implant failure.
METHODSA comprehensive search for all relevant articles published in English from January 1995 to December 2010 was conducted. Two reviewers evaluated each study to determine whether it was eligible for inclusion and collected the data of interest. Meta-analytic pooling of results across studies was performed for the complications and failure rate.
RESULTSThirty-five primary studies with 4395 implants were identified. The three highest complications of total ankle arthroplasty were aseptic loosening (12.51%), intra-operative bone fracture (11.97%) and bony impingement (11.27%). The three high-grade complications associated with implant failure were aseptic loosening (45.00%), infection (33.00%) and malalignment (29.00%). The pooled mean failure rate was 10.98% (95%CI: 8.80% - 13.16%), and the pooled mean failure rate of STAR implant was 14.20% (95%CI: 10.64% - 17.76%).
CONCLUSIONSIt is found that aseptic loosening, infection and malalignment are high-grade complications associated with implant failure in total ankle arthroplasty. The orthopaedic surgeons should be more careful in the operation, and the patients should coordinate with the post-operative rehabilitation plan.

OBJECTIVETo investigate the placement of a long tube into the small intestine under fluoroscopic guidance and to evaluate its decompression effect on early postoperative small bowel obstruction (EPSBO).
METHODSFifty-four patients with EPSBO requiring decompression between April 2010 and July 2014 were enrolled in the study. Insertion of a long tube was guided by fluoroscopy. We first used the guide wire to pass the pylorus and then used the 10 Fr feeding tube as an exchangeable tube to put the superstiff wire into the duodenum. Finally the long tube could be passed over the guide wire through the pylorus into the intestine. The total procedure time, the radiation exposure time, and the incidence of complications were evaluated.
RESULTSThe long tubes passed into the jejunum on initial insertion for all patients, so the success rate of this technique was 100%. The long tube was inserted into ileum in 18 patients. The mean total procedure time was 34.4 ± 8.6 minutes, and the mean radiation exposure time 18.9 ± 6.8 minutes. A total of 47 patients (87%) experienced full recovery following long-tube decompression and without the need for surgical intervention.
CONCLUSIONSUsing the wire-exchange technique, it is easy to place a long tube into the small bowel under fluoroscopic guidance. This decompression method is safe and effective for management of EPSBO.

Microbiology 2008;0(08):-
The population of bacterial physiological groups in tomato with different resistant to Ralstonia solanacearum was studied. The results suggested that endophytic bacterial communities and population in tomato variety changed with different resistant cultivars, different stages of tomato and seasons. It was con-ducted that the amount of ammoniation bacteria was the highest among the seven physiological bacterial groups. There were more ammoniation bacteria in high resistant tomato cultivars than that in high suscepti-ble cultivars. It may indicate that ammoniation bacteria played a key role in the occurrence of tomato bacte-rial wilt. In addition, the total amount of physiological bacteria in resistant cultivars was more than that in susceptible cultivars in different stages of tomato, and the tendency of changing displayed fluctuation. The average level of quantities of the ammoniation bacteria, nitrifiers bacteria, erobic nitogenfixing bacteria and desulphate reducer bacteria in summer were higher than that in winter, while the population of the sulphate reduced bacteria in winter was higher than that in summer. Furthermore, the amount of anaerobic bacteria was the least among them.

Objective To study collagen changes in dermis of hairless mice that were exposed to ultraviolet. Methods The hairless mice was irradiated under UVA, UVB and the combination of the two for 20 weeks, total dose of UVA was 222J/cm~2, and that of UVB was 5.9J/cm~2. After irradiation, the dorsal skin's collagens of animals were analysed by computer imaging analysis system, histopathologic examination, specific stains and electorn microscopy. Results The hairless mice exposed to ultraviolet A were unchanged in dermis collagen. The hairless mice was irradiated under UVB and the both UVA and UVB, and the content of collagen was decreased with less affinity for collagen staining. These findings were supported by electron microscopy, which showed fraying, thickened, and proliferating collagen, coalesced into extensive denaturalization. The ratio of types Ⅲ/Ⅰ+Ⅲ collagen was significantly increased. Conclusion The qualitative and quantitative changes of the collagen in the ultraviolet irradiated skin of hairless mice are related to ultraviolet B but not to UVA. UVB is a key factor of skin collagen damage in UV-irradiation.

Objective To promote the use of chemical peeling in facial rejuvenation with the phenol and croton oil peeling agents to the UVA/B-irradiated skin of hairless mice, and to provide the experimental evidence for the clinical application of the treatment of irradiated skin. Methods Sixty BALB/C hairless mice were photo-aged by use of chronic ultraviolet A and ultraviolet B irradiation for 20 weeks. After irradiation the animals were randomly divided into two groups:untreated (10 mice) and treated (50 mice). The phenol and croton oil chemical peeling agents were applied to the dorsal skin of treated animal group while it was full anesthetized. Punch biopsies were taken at 7, 14, 30, 60, and 90 days after peel for histological analysis. At 60 days after irradiation, the skin wrinkling of animals were analyzed by macroscopy, cleavage line amplification, and computer imaging analysis system. Results The treated areas of irradiated skin recovered rejuvenation and exhibited a unique connective tissue layer composed of fine collagen fibers beneath the epidermis. Conclusion The mixture of phenol-croton oil may reverses the visible stigmata of photoaging skin. Our results will be of great help to promote the use of chemical peeling in facial rejuvenation.
