1.Isolation,cultivation,purification and proliferation of different gestational age human embryonic neural stem cells

Objective:To explore the proliferation regularity of different gestational age human embryonic neural stem cells after them were isolated, cultivated and purified successfully.Methods:3~18 weeks human embryo were obtained under asepsis condition, and divided into 3 groups according to gestation age: A group(3~6w), B group(7~11w), C group(12~18w).The each group of NSCs were isolated and purified after primary culture, and identified by single-cell clone culture and immunocytochemistry.(ICC)To study NSCs growth feature by counting numbers and diameters of neurospheres. Results:The cells could survive and proliferate in vitro which were isolated from 3~6 weeks human embryo pallium and neurocanal or 7~18 weeks human fetus pallium. Identification of the cells showed that they had the typical feature of NSC biology in many aspects,such as growth pattern,morphosis,function feature and surface marker. The numbers and diameters of neurospheres from 3~6w gestation age group are much higher than that from 7~18w gestation age group(P
2.Effect of batroxobin on endothelin and calcitonin gene-related peptide in patients with acute cerebral infarction

Objective To investigate the effect of batroxobin on the plasma levels of endothelin (ET) and calcitonin gene-related peptide(CGRP) in patients with acute cerebral infarction(ACI). Methods 60 patients with ACI were randomly divided into therapy group(n = 30) and control group(n = 30),500 ml of 5% GS plus 1.0g ramme of Troxerutin was intravenously dripped par day and ticlopidine 250mg/day in the control group. Based on the control group therapy,Batroxobin was intravenously dripped with 10U on the 1st day and 5U on the 3rd and 5th day for seven days. The plasma levels of ET and CGRP were detected pre and post treatment by radioimmunoassay. Results Plasma levels of ET and CGRP in batroxobin therapy group post treatment ( 81.25±27.38 ) ; ( 44. 13±13.30) ng/L were significantly different from those pre treatment(109. 83±38.24) ; (24. 73±13.59) ng/L (all P<0. 01 ) ,and were also significantly different from those post treatment in the control group(98.07±30. 39) ; (37.02±12. 41 )ng/L (all P<0. 01 ). Conclusion Batroxobin can accommodate the equilibration between ET and CGRP, and protect brain cell and blood vessel endothelial cell.
3.EFFECT OF CORYDALIS AMABILIS MIGO TOTAL ALKALOID ON FUNCTION OF PLATELETS IN RATS

Corydalis amabilis Migo total alkaloid (COAMTA) given in vitro or in vivo caused an inhibition on ADP-induced blood platelet aggregation. In vitro, COAMTA 15.6, 31.3 & 62.5 ?g/ml PRP showed their inhibitory rates of 8.2, 32.0, & 36.8%. In vivo, at 10, 20, 40 min after iv COAMTA ( 0.6 mg/kg ) , the rates of inhibition were 40, 62.8, 24%.The thrombi formed in vitro were measured also. The results showed that COAMTA given in vitro or in vivo reduced the length and weight of thrombus, at same time, reduced rat platelet adhesion.A carotid-jugular extracorporeal shunt was made in the rats. A 6cm length of silk thread was placed in the shunt. 15 min after the restoration of blood flow in the shunt, the Wet weight of thrombus developed on the thread was measured.The thrombus weight was significantly reduced when COAMTA was given iv 0.3, & 0.6 mg/kg, im 0.9 mg/kg & po 1.5 mg/kg. The thrombus weight of control group was 26.1?0.7, but that of the medicated group was 8.9?1.7, 18.0?1.3, 16.4?0,6 a & 19.7?0.7 mg. The inhibitory rates were 66.1, 31.3, 37.2, & 28.4%.

Corydalis amabilis Migo total alkaloids (COAMTA) possessed antihypertensive and vaso-dilatative action in animal experiments. Its effects on the physiological properties in isolated guinea pig papillary muscles & atrium preparations were described in this papar.10mg/L COAMTA markedly prolonged the functional refractory period(FRP) and inhibited epinephrine induced automaticity. Suppression of normal & abnormal automaticity and prolongation of FRP may contribute to the effectiveness of COAMTA on arrhythmia.
5.The analysis of factors related to adherence to the diagnosis and treatment of gout patients after discharge

Objective To explore factors affecting diagnosis and treatment compliance in patients with gout leaving hospital,in order to provide a reference for late-stage clinical treatment.Methods 390 patients with gout treated in our hospital were selected from August 2012 to February 2016 for the study,all patients were followed up by telephone,their diagnosis and treatment compliance after discharge was investigated,and the factors affecting diagnosis and treatment compliance in gout patients after discharge investigated were,as well.Results The mean score of treatment adherence of 390 cases of gout patients after discharge was(3.93± 1.78) points.There were 117(30.00%) patients of which their score were ≥6 points for compliers,and 273 (70.00%) patients who do not comply with scores <6 points.Disease duration,education level,frequency of gout attacks,marital status and payment of medical expenses were closely related to diagnosis and treatment adherence(P<0.05).Longer duration,low educational level,payment of medical expenses at their own expense were independent risk factors for poor adherence (P<0.05).Conclusion We should strengthen the monitoring and follow-up of patients with gout who are with long duration of disease,low educational level,and pay medical expenses from their own pocke tinorder to improve patient diagnosis and treatment adherence,provide references for late-stage clinical treatment.
6.Influence of Ulcer Stage on Helicobacter pylori Eradication Rate

Background: Helicobacter pylori (Hp) infection is associated with a variety of gastrointestinal diseases.Studies have shown that Hp eradication rate may be affected by the disease type and stage of disease progression.Aims: To investigate the influence of endoscopic ulcer stage on Hp eradication rate.Methods: A total of 423 peptic ulcer (PU) patients with Hp infection from January 2015 to July 2016 at the People's Hospital of Xinjiang Uygur Autonomous Region were enrolled.Of them,133 were gastric ulcer (GU) and 255 were duodenal ulcer (DU).A 14-day bismuth quadruple therapy was given for Hp eradication,and the eradication rates were analyzed with the endoscopic ulcer stage.Univariate analysis was conducted to identify factors influencing the eradication rate.Results: There was no significant difference in Hp eradication rates between GU and DU groups (82.7% vs.83.9%,P>0.05).The eradication rates according to endoscopic ulcer stage were significantly different in GU group (active stage vs.inactive stage: 89.0% vs.75.0%,P<0.05);but in DU group,the eradication rate in active stage was similar to that in inactive stage (85.6% vs.83.0%,P>0.05).Univariate analysis revealed that ulcer stage was correlated with the success rate of eradication;the eradication rate in active stage was significantly higher than that in inactive stage (87.8% vs.80.3%,P<0.05).Conclusions: Endoscopic ulcer stage is correlated with the Hp eradication rate in PU,especially GU patients.Hp eradication therapy performed in patients with active PU could yield a higher eradication rate.

8.Optimization on Formulation of Controlled-onset Extended-release Tablets of Nicorandil by Central Composite Design-response Surface Method

Objective To optimize the formulation of controlled-onset extended-release tablets of nicorandilby central composite design-response surface method. Methods The tablets containing nicorandil were prepared bydry-compression coating technique.The influence factors included the amount of HPMC, the amount of EC, and the ratio of lactose/MCC in coating film. The evaluation parameter was Q4 h and Q5 h . The possibly optimal formulation was predicted by response surface method. Results The OD was simulated using second-order polynomial equation and the r2was 0.970 1.Optimal prescription was as follows:HPMC 89.96 mg, EC 46.21 mg, Lactose/MCC 1.87.Bias between the observed and predicted values of the OD was within 0.43%. Conclusion The controlled-onset extended-release tablets of nicorandil can release drug quickly in vitro at the predetermined time.Central composite design-response surface method can be used to optimize the formulation and the model developed in this study was proved highly predictable.
9.Cryptotanshinone effects on the proliferation and apoptosis of renal carcinoma stem cells

Abstract BACKGROUND:Previous studies have found that cryptotanshinone represses multiple tumors, but little is reported on its effect on renal carcinoma. OBJECTIVE:To explore the effect of cryptotanshinone on the proliferation and apoptosis of the renal carcinoma stem cels. METHODS:CD133+ renal carcinoma stem cels were separated from OS-RC-2 cels by immunomagnetic bead separation. Effects of 0, 0.2, 1, 5 mg/L cryptotanshinone on the proliferation and apoptosis of CD133+ renal carcinoma stem cels were detected by MTT and flow cytometry, respectively. Expression levels of Ki67, Bcl-2, Caspase-3 and p-Caspase-3 protein were detected by western blot assay. RESULTS AND CONCLUSION:After magnetic cel sorting, the percentage of CD133+ cels was increased from 6.32% to 82.73%, and there was a significant difference before and after cel sorting (P < 0.001). Cryptotanshinone could repress the proliferation of CD133+ renal carcinoma stem cels and promote cel apoptosis in a dose-dependent manner. The protein expression levels of Ki67 and Bcl-2 in the 5 mg/L cryptotanshinone group were significantly decreased compared with the control group, while the protein expression level of p-Caspase-3 protein was significantly increased. In addition, there was no difference in the protein expression of Caspase-3 between cryptotanshinone and control group. These findings indicate that cryptotanshinone may be a potent anticancer drug for the treatment of renal carcinoma by inhibiting expression of Ki67 and Bcl-2 and promoting protein expression of p-Caspase-3.
