1.Stem cells in the skin regeneration and wound healing:problem and perspective

BACKGROUND:Present treatments for chronic skin wounds have certain limitations, and adult stem cels play a potentialpart in cutaneous repair and regeneration.
OBJECTIVE:To review effects of stem cels in skin regeneration and wound healing.
METHODS:The first author retrieved CNKI and Medline databases by computer for relevant articles published from 2000 to 2010. Thekeywords were “epidermal stem cels, hair folicle stem cels, stem cels, transplantation, dermal stem cels” in Chinese and in English, respectively. Then totaly 489 papers were obtained after initial survey, and according to the inclusion criteria, 30articles were selected for review.
RESULTS AND CONCLUSION:Epidermal stem cels and other adult stem celshave beenapplied to treat wounds and other skin diseases. Epidermal stem celsarethe crucialcelsource of skin development, repairandremodeling.Epidermal stem celsarealwaysina resting statein vivo.Unless,skin injure or culturein vitro, celdivision and proliferation wilbe significantly fastened. The stability of the epidermis mainly dependson the asymmetric divisionof a subpopulation, in which two daughter celsare produced, including one with characteristics of stem cels, and the other differentiated into transient amplifying cels that wil be differentiated intopost mitotic celsafter a series of cel divisions (3-5 times). Afterwards, those post mitotic celsaredeveloped into terminal differentiation cels onthe basal layer, finaly detachfromthe epidermisasdander. In addition, it is unclear whether epidermal factors are related to apoptosis, migration and differentiation in the process of wound repairandeven under physiological conditions. Therefore, application ofstem celsinwound healing requiresa further discussion.
5.Determination of Ephedrine Hydrochloride in Zhike Pingchuan Tangjiang by HPLC

Objective To establish the method for detemining the content of ephedrine hydrochloride in Zhike Pingchuan Tangjiang by HPLC. Methods Diamonsil ODS1 C_(18) Column was used with acetonitrile -0.1% phosphoric solution (with 0.1% triehylamine) (3 : 97) as the mobile phase, the detection wavelength as 205 nm, and flow rate was 1.0 mL/min. Results The calibration curve was linear at the range of 0.12~ 0.96 ?g for ephedrine hydrochloride and linear equation was Y= 109759X+3792.8, r=0.9998. The average recovery was 98.4% and RSD was 0.87% (n =5). Conclusion This method was simple, accurate and proper, with good reproducibility. It can be used for quantitative analysis of ephedrine hydrochloride in Zhike Pingchuan Tangjiang.
6.Clinical Observation of Shenmai Injection Combined with Trimetazidinee Tablet in the Treatment of Hypothy-roid Heart Disease

OBJECTIVE:To observe the clinical efficacy and safety of Shenmai injection combined with Trimetazidinee tablet in the treatment of hypothyroid heart disease. METHODS:64 patients with hypothyroid heart disease were randomly divided into control group and observation group. Patients in 2 groups were conventional treatment. Based on the treatment,control group was treated with Trimetazidine tablets 20 mg,orally,3 times a day;based on the treatment in control group,observation group was ad-ditionally treated with Shenmai injection 40 ml by intravenous infusion,once a day. The treatment course was 2 months. The clinic data was observed,including clinical efficacy,and T3,T4,TSH,LVEF,LVEDD and LVESD before and after treatment,pericar-dial effusion depth and incidence of adverse reactions. RESULTS:The total effective rate in observation group was significantly higher than control group(P<0.05). After treatment,T3 and T4 in 2 groups were significantly higher than before,TSH and LVESD were significantly lower than before(P<0.05),however,the difference was not statistically significant between 2 groups (P>0.05);LVEF in 2 groups was significantly higher than before,observation group was higher than control group,LVEDD and pericardial effusion depth were significantly lower than before,observation group was lower than control group(P<0.05). There were no obrious adverse reactions during treatment. CONCLUSIONS:Based on the conventional treatment,Shenmai injection com-bined with trimetazidinee has better efficacy than only trimetazidinee in the treatment of hypothyroid heart disease,with similar safety.
7.The progress of epigenetics in the development and progression of non-alcoholic fatty liver disease

Non-alcoholic fatty liver disease(NAFLD)has become a global epidemic nowadays.Although the path-ogenesis of NAFLD remains to be uncovered,it is highly correlated with the changing of environment as other metabolic diseases.Epigenetics is the study on the differences of gene expression not caused by the changes in DNA sequencing.The epigenetics is considered to link the environment factors and the onset and development of NAFLD.As an increasing number of researches on epigenetics have emerged in recent years,our understanding of how epigenetic factors take part in the pathogenesis of NAFLD has been improved.This article reviews the recent studies on important epigenetic factors contributing to the progression of NAFLD including the DNA methylation,modification of histones and non-coding RNAs.It may give us a hint to discover novel diagnosis methods and drug targets for NAFLD.

9.Effects of berberine on proliferation, migration and adhesion of lung adenocarcinoma cell line A549

Background and purpose: Reports showed that berbefine not only had an effect ofdetoxification and inflammation but also could induce the cell to apoptosis. Berberine may also inhibit the migration and metastasis of tumor. To investigate the effects and mechanism of berberine on migration and metastasis of A549 cells, the proliferation, mobility and adhesion of A549 cells were observed after incubation with berberine. Methods: The proliferation was determined by MTT. Wound healing assay and adhesion test were used to observe the mobility and adhesion rate. The mRNA and protein expressions of MMP2 and MMP9 were assessed by semi-quantitive RT-PCR and gelatin zymography, respectively. Results: Berberine could significantly inhibit the proliferation ofA549 within a certain range of treating time and dose (P<0.05). The mRNA and protein expression of MMP2 and MMP9 gene were decreased slightly by the treatment ofberberine (P<0.05), especially at dose of 100 μg/ml. Conclusion: Berberine can inhibit the migration and adhesion abilities of A549 cells, probably by down-regulating the expression and activation of MMP2 and MMP9.
10.Advances in cardiovascular effects of tanshinone II(A).

Cardiovascular diseases, like coronary heart disease and myocardial infarction, are the most common cause of death worldwide. Chinese medicines have demonstrated rich cardioprotective activities for clinical applications. Salvia miltiorrhiza, a very important component of traditional Chinese medicine, can promote blood circulation and relieve blood stasis. Salvia miltiorrhiza is widely used in treatment of cardiovascular and cerebrovascular disease such as coronary heart disease and cerebral infarction ( CI). Tanshinone II(A), the major lipophilic components extracted from the root of S. miltiorrhiza, possesses anti-atherosclerosis, anti-cardiac hypertrophy, anti-oxidant, anti-arrhythmia and so on. This paper discusses current research status of tanshinone II(A) in cardioprotective effects.
