Objective To study the application value of preoperative medical adhesive locating in performing thoracoscopy for small pulmonary nodules (SPN).Methods The clinical data of 41 patients with solitary SPN,who received thoracoscopy with the help of preoperative locating procedure,were retrospective analyzed.The preoperative locating methods included medical adhesive locating (n=22,medical adhesive group) and Hook-wire locating (n=19,Hook-wire group).The locating effects,complications,the time of engagement stage and the time used for pulmonary wedge resection were determined,and the results were compared between the two groups.Results The locating of SPN was successfully accomplished in all patients of both groups.The incidences of pneumothorax,pulmonary hemorrhage and chest pain in the medical adhesive group were 18.2％,9.1％ and 4.5％ respectively,which were lower than those in the Hook-wire group;among them the difference in the incidence of pulmonary hemorrhage between the two groups was statistically significant (P＜0.01).No-complication rate in the medical adhesive group (63.6％) was higher than that in the Hook-wire group (21.1％),and the difference was statistically significant (P＜0.05).The time of engagement stage for patients in the medical adhesive group was (16.32±8.83) hours,which was longer than (3.29±4.21) hours in the Hook-wire group,the difference between the two groups was statistically significant (P＜0.01).The time used for puhnonary wedge resection in the medical adhesive group and in the Hook-wire group was (21.14±7.01) min and (18.58±5.22) min respectively,the difference between the two groups was not statistically significant (P＞0.05).Conclusion Preoperative medical adhesive locating for the performance of thoracoscopy for SPN is safe and effective,it carries less complications when compared with Hook-wirelocating method and can obtain a longer interwal period,therefore,this technique has high application valuein clinical practice.

AIM: The autoantibodies against ?_1-adrenergic receptor that was found in patients with malignant hypertension, primary hypertension and refractory hypertension has the agonist activity liked the NE, and may play a role in hypertension. In this paper, the effects of this antibody on vascular smooth muscle cell (VSMC) proliferation and its mechanism were to be studied. METHODS: The cultured rat VSMC proliferation induced by the antibodies against ?_1- adrenergic receptor that was purified by the immune affinity chromatography, was measured by the BrdU cell proliferation assay and cell cycle distribution. The expression of c-jun and c-fos were determined by RT-PCR and Western blotting. RESULTS: Compared to the normal IgG, the antibodies against ?_1-adrenergic receptor promoted the VSMC proliferation and increased the mRNA and protein expression of the c-jun significantly. The role was similar to the norepinephrine, and all was blocked by prazosin, while the mRNA and protein expression of c-fos were not affected by the antibodies. CONCLUSION: The antibodies against ?_1-adrenergic receptor promote the rat VSMC proliferation, and increase the expression of c-jun, which maybe play a role in the vascular remodeling in hypertension.

OBJECTIVETo evaluate the clinical efficacy of combined treatment with lamivudine and famciclovir on chronic hepatitis B virus (HBV) infection.

METHODSNinety patients with chronic HBV infection were divided into 3 groups. Group one had 28 patients and was treated with combination of lamivudine (0.1 g/d, PO) and famciclovir (1.5 g/d,PO) for 24 weeks. Group two and three had 30 and 32 cases, respectively, and were treated with lamivudine 100 mg/day PO and famciclovir (1.5 g/d,PO) alone. All the patients had positive markers of HBsAg, HBeAg and anti-HBcAg in serum assayed by ELISA and of HBV DNA tested by PCR.

RESULTSThree strategies of treatment had no different effects on the change of patients' ALT levels. The serum HBV DNA became negative after treatment in 89.3% (25/28) of patients treated with combination of lamivudine and famciclovir, 66.7% (20/30) of patients treated with lamivudine, and 40.6% (13/32) of patients treated with famciclovir. The rate of serum HBeAg loss in 3 groups were 28.6% (8/28), 23.3% (7/30) and 21.9% (7/32), respectively.

CONCLUSIONSThe combination treatment of lamivudine and famciclovir for chronic HBV infection is safer than and superior to that of either drug alone.

OBJECTIVETo study the clinical characteristics and preventive measures of liver failure with nosocomial septicemia.

METHODSRetrospective analysis of nosocomial septicemia seen between 2001 and 2002 was carried out in our hospital.

RESULTSIncidence of nosocomial septicemia was 0.61%, mortality was 14.29%, the main pathogen was Escherichia coli, the drug resistance occurred in most pathogens to the commonly used antibiotics.

CONCLUSIONIn order to reduce nosocomial septicemia, antibiotics should be used rationally, should be paid attention to bacterial culture and antibiotic sensitivity, and preventive measures should be taken.

T cell immune receptor with Ig and ITIM domains (TIGIT), a promising new target in cancer immunotherapy, plays a critical role in limiting adaptive and innate immunity against tumors. The extracellular domain of human TIGIT was used to immune BALB/c mice, and a new anti-human TIGIT chimeric antibody (c7D3) was developed. The mice in this study were used in accordance with the international guidelines for the care and use of laboratory animals, and the animal study was approved by the Institutional Animal Ethics Committee of AbMax Biotechnology. The biological activity of c7D3 was studied. The results showed that c7D3 exhibited high affinity for TIGIT and effectively inhibited the interaction between TIGIT and its ligands. Cell-based assays indicated that c7D3 induced strong luciferase signaling in TIGIT/CD155 signaling reporter assay and enhanced cytokine secretion in a T cell stimulation assay. The data showed that c7D3 has high binding affinity and excellent blocking bioactivity, supporting the further advancement for therapeutic application.

Objective To construct a eukaryofic expression vector for A PP695 gene carrying green fluorescence protein (GFP) gene. Methods RT-PCR was used to amplify the full-length APP695 cDNA. The PCR products and the eukaryotic expression vector pIRES2-EGFP were digested by restriction endonueleases, and the digested APP695 gene was inserted into the digested eukaryotic expression vector. The positive recombinants were identified by PCR analysis, Nhe Ⅰ and Hind Ⅲ digestion and sequence analysis. Results The 2088-bp DNA fragment was amplified by PCR from the plasmid pCB6, and an identical DNA fragment was also amplified from the recombinants. The products of double restriction enzyme digestion were A PP695 gene with a 5.3-kb DNA fragment. Sequence analysis confirmed successful insertion of A PP695 gene into pIRES2-EGFP vector. Conclusion The eukaryotie expression vector pIRES2/APP695-EGFP has been successfully constructed.

OBJECTIVETo discuss the changes of lymphocyte subsets in HCV children with different genotypes during treatment with pegylated interferon alfa-2b and ribavirin.

METHODSThe genotype of 45 HCV infected children were identified by real time PCR. The lymphocyte subsets were dynamically detected by BD FACSCalibur flow cytometer with four color MultiTEST IMK Kit during the treatment.

RESULTSFor the children with 1b genotype, after 24 weeks, the CD4+ T cells were higher than pre-treatment (P < 0.05). For the children with 2a genotype, after 12 weeks and after 24 weeks, the CD3+ T cells and CD4+ T cells significantly increased while the NK cells decreased than pre-treatment (P < 0.05).

CONCLUSIONSThe lymphocyte subsets of HCV children with 2a genotype were different from 1b genotype during trentment with pegylated interferon alfa-2b and ribavirin.

In order to explore the effect of abendazole sulfoxide-loaded thermo-sensitive hydrogel (ABZSO/PLGA-PEG-PL-GA) on alveolar hydatid cysts,the viable protoscoleces incubated in vitro with ABZSO/PLGA-PEG-PLGA (20,40,80 μg/mL),ABZSO (40 μg/mL),PLGA-PEG-PLGA and the blank control,respectively,and the viable protoscoleces were counted.The morphological of the alveolar hydatid cysts or protoscoleces in each group were observed by scanning electron microscop(SEM).After the rats infected with alveolar hydatid cysts having been treated,the sizes of the alveolar hydatid cysts were observed.When the alveolar hydatid cysts incubated in different drug group in 18 days,the percentage of viable protoscoleces in ABZSO/PLGA-PEG-PLGA (40μg/mL) group was lower than that of ABZSO (40 μg/mL) group.Results of SEM showed that the protoscoleces in the ABZSO/PLGA-PEG-PLGA (80 μg/mL) group were seriously damaged.The in vivo study demonstrated the anti-alveolar hydatid cysts efficacy of ABZSO/PLGA-PEG-PLGA (4 mg/mL) group was better than the other groups.The anti-alveolar hydatid cysts efficacy of ABZSO/PLGA-PEG-PLGA is evident.
