1.Effect of Ji Desheng Sheyao on coagulation, liver,kidney function in bee sting injury patients with severe toxic effect

Objective To analysis the the effect of Ji Desheng Sheyao on coagulation,liver, kidney function in bee sting injury patients with severe toxic effect.Methods 42 bee sting injury patients with severe toxic effect were collected.Corresponding treatment and nursing measures were given respectively, after the treatment, the coagulation function,liver, kidney function and nursing effect were detected in all patients.Results After treatment compared with control group,there was no significant difference in the coagulation function of the patients in the experimental group;the serum levels of ALT,AST and TBIL were lower in the experimental group (P<0.05); the serum levels of BUN and Scr were lower in the experimental group(P<0.05);the incidence of complications was lower, and the nursing satisfaction was higher in the experimental group(P<0.05).Conclusions Ji Desheng Sheyao and good nursing measures can significantly reduce oxidative stress in patients with severe toxic effect , improve liver and kidney function, reduce the incidence of complications, improve patient care satisfaction.
2.Effect of XinBao pill on electrophysiological properties of human HCN4 channel

Objective To explore the basic medical mechanism of XinBao pill on electrophysiological characteristics of hyperpolarization-activated cyclic nucleotide-gated channel 4 (HCN4), and illustrate the mechanism of its therapeutical effect on bradycardia.Methods Human HCN4 mRNA was injected into the Xenopus laevis oocytes, after incubated for 2 ~3 days, channel current properties of HCN4 perfused with 40 mg/L XinBao Pill were observed by double electrode voltage clamp technique.Results At-90mV test potential, compared with control group (no XinBao pill), HCN4 channel peak current and tail current in 40 mg/L XinBao pill group had obvious changes, and V1/2 from ( -103.61 ±3.57)mV to ( -106.42 ±5.33)mV in XinBao pill group, from( -81.11 ±4.26)mV to( -86.36 ±7.44)mV in control group.The values of k from (15.15 ±2.23)mV to (17.33 ±3.58) mV in XinBao pill group, from(11.78 ±0.85)mV to(12.39 ±1.51)mV in control group(n=10).At test potential -90 mV, 40 mg/L XinBao pill perfusion fluid decreased the instantaneous current of(0.15 ±0.24)%, the EC50 was (30.8 ±4.8)mg/L (n=8).At test potential-140 mV~-100 mV level, 40 mg/L XinBao pill group increased the channel activation time constant compared with control group[(226.73 ±31.36)ms vs(143.67 ± 21.44)ms;-140 mV,n=10,P<0.05].40 mg/L XinBao pill group increased the channel deactivation time constant compared with control group [(1293.53 ±95.02)ms vs (647.12 ±61.35)ms;-140 mV,n=10,P<0.05].Conclusion The XinBao pill enhances the instantaneous current of HCN4 in a concentration-dependent manner, and extents channel activation and deactivation processes.
3.Cluster Analysis on Traditional Chinese Medicine Syndrome of Metaphase and Advanced Lung Cancer

objective To analyse the traditional Chinese medicine syndromes of lung cancer by the way of variable cluster analysis. Methods 176 cases of lung cancer were collected and their symptoms were recorded. Then these patients’ clinical data were analyzed by variable cluster analysis. Results The Chinese medicine syndromes of these patients could be divided into 5 types, and the variable proportion is 50.94%. Conclusion TCM syndromes of metaphase and advanced lung cancer patients include both deficiency and excessive syndrome, mainly deficiency syndrome with. And, the way of variable cluster analysis could help TCM syndrome differentiation of lung cancer patients.
5.Study on the mechanism of primary tumor treatment promoting tumor metastasis

Tumor metastasis is a primary cause of leading to death of tumor patients,and difficult to control.The relationship between primary tumor(PT)and metastasis tumor(MT)is extremely complicated. Considerable animal experiments and clinical data confirm that treatment of PT encourages the growth of MT, but the mechanism is not still fully clear.Some studies show that the density depression of antiangiogenesis factor and immune escape of tumor cell are main reasons for MT accelerated growth.This article reviews the animal experiment,clinical observation and occurrence mechanism of primary tumor treatment promoting tu- mor metastasis.
7.Roles of Akt pathway and cell apoptosis in leptin-mediated chronic mor-phine antinociceptive tolerance

AIM:To explore the roles of Akt ( also called protein kinase B ) and active caspase-3 in the leptin-mediated chronic morphine antinociceptive tolerance in rats .METHODS: A model of chronic morphine antinociceptive tolerance was established in the SD rats .The protein levels of spinal Akt and cleaved caspase-3 were tested by Western blotting.The technique of immunohistochemical staining was used to detect the immunoreactivity positive cells of phospho -rylated ( p)-Akt and cleaved caspase-3 in the spinal cord .Double staining of immunohistochemistry was used to examine the cellular location of the p-Akt and cleaved caspase-3 positive cells.RESULTS: The chronic intrathecal injection of morphine (15 μg) for 7 d markedly upregulated the spinal protein levels of p-Akt and cleaved caspase-3 in the rats.Thirty min before injection of morphine , intrathecal injection of leptin antagonist (3μg) for 7 d significantly attenuated the upreg-ulation of the protein levels of p-Akt and cleaved caspase-3 induced by chronic morphine treatment .The p-Akt was exclu-sively observed in the spinal neurons .The cleaved caspase-3 was only localized with the spinal astrocytes .Intrathecal injec-ting the inhibitors of leptin , Akt and caspase-3 ameliorated the chronic antinociceptive tolerance .CONCLUSION: The spinal Akt pathway and active caspase-3 are involved in the leptin-mediated chronic morphine antinociceptive tolerance in rats.
8.Osteogenetic ability of mouse spermatogonial stem cells cultured in vitro

BACKGROUND:Spermatogonial stem cells are a kind of adult stem cells, which have self-renewal and differentiation potential, and can be differentiated into specific cells in vitro, suggesting that the spermatogonial stem cells may be possibly differentiated into osteoblasts. But the related research has not been reported. OBJECTIVE:To observe the biological characterization and osteogenic process of mouse spermatogonial stem cells cultured in vitro. METHODS:Spermatogonial stem cells were obtained from the testicle of mice aged 15-20 days, and were cultured on the feeder layer from bone marrow stroma cells in vitro. When cultured for 3 days, the cells were cultured in the conditioned medium (experimental group) and basic medium (control group). The cells proliferation capability and osteogenic property were examined by phase-contrast microscope, alkaline phosphatase activity and type I col agen immunofluorescence staining. RESULTS AND CONCLUSION:Spermatogonial stem cells proliferated faster in the experiment group than in the control group. cells grew rapidly in colony-like shape in the conditioned medium at 3-6 days, the three-dimensional feeling enhanced, cellmass and clusters continued to increase in size, the extracellular matrix was increased in number and the cytoplasmic bridge was not obvious. After culture for 15 days, cells in the two groups were positive for alkaline phosphatase staining that the cytoplasmic membrane was dyed black. Under the fluorescent microscope, green fluorescence was visible in the experimental group, suggesting the cells in the experimental group was positive for type I col agen, but negative in the control group, which is similar with the biological characteristics of osteoblasts. These findings indicate that spermatogonial stem cells possess the osteogenic capability under induction conditions, which are expected to provide seed cells for bone tissue engineering.
9.Survey and analysis of knowledge, attitude and practice of physical restraint on critical parents in secondary hospitals and tertiary hospitals in Hubei province

Objective To investigate the situation of knowledge,attitude and practice of physical restraint from nurses for critical parents in secondary hospitals and tertiary hospitals in Hubei province.Methods 300 nurses from four tertiary hospitals and four secondary hospitals were investigated for the situation of knowledge,attitude and practice of physical restraint by questionnaires.Results It showed that nurses had a good knowledge of physical restraint,(11.02±1.96),their attitude of physical restraint tended to be neutral,(34.99±3.52),the practice of physical restraint turned out to be at meddle-up level,(51.90±6.67).There was a statistically significant difference in the scores of physical restraint knowledge and practice between nurses who receive physical restraint train or not after they got a job,besides,there was also statistically significant difference in the scores of physical restraint knowledge between intensive department and non-intensive department,tertiary hospitals and secondary hospitals,however there was no statistically significant difference in the scores of physical restraint attitude and practice between them.Conclusions Compared with non-intensive department and secondary hospitals,the nurse managers of intensive department and tertiary hospitals should strengthen the training and education of physical restraint of clinical nurses,guide the nurses to build the positive attitude of physical restraint,to reduce the use of preventive physical restraint and then to promote their quality of practice on physical restraint.
