1.The interaction analysis of genetic and behavioral factors on the occurrence of acute coronary syndrome

Objective To explore the the interaction analysis of genetic and behavioral factors on the occurrence of acute coronary syndrome. Methods This study consisted of 134 subjects between June 2009 and Dec.2009 from affiliated hospital of Jining Medical University, All subjects underwented selective coronary angiography or coronary artery CT. Coronary artery disease based on the results of coronary angiography or coronary artery CT that at least one coronary artery diameter reduction of more than 50% and fulfilled the diagnostic criteria of ACC/AHA. 84 patients with acute coronary syndrome and 50 controls, and general informations, current disease history,past medical history, related be havioral factors were collected. DNA was extracted from acute coronary syndromepatients and healthy control subjects, stored in -20 ℃. The genotype of CFH Y402H was detected by polymerase chain reaction and restriction fragment length polymorphism(PCR-RFLP) methods in acute coronary syndrome patients and controls. PCR amplification products was verificated by electrophoresis on agarose gel. Deviation of genotype distribution form Hardy-Weinberg equilibrium was assessed using x2 test in each group. Univariate and multivariate conditional logistic analyses were conducted in the end. Results The results showed that smoking history (P=0.000, OR =4.894,OR 95% CI:2.537 ~9.441 ), alcohol drinking history(P= 0.008,OR =2.879,OR 95% CI: 1.499 ~ 5.528 ), hypertension (P = 0.000, OR = 4.222, OR 95% CI: 2.195 ~ 8.123), sports activities (P =0.002,OR =0.333, OR 95% CI:0.188 ~ 0.589 ), salt intake (P= 0.006, OR = 0.457, OR 95% CI:0.287 ~0.727 ), character(P = 0.000, OR = 0.385, OR 95% CI :0.247 ~ 0.600 ) stress of occupations (P = 0.015, OR =2.118, OR 95% CI: 1.278 ~ 3.511 ) were associated with acute coronary syndrome of Northern Chinese Han population. Smoking history(P = 0.010, OR = 6.084, OR 95% CI: 1.543 ~ 23.988), hypertension (P= 0.024, OR =2.821, OR 95% CI: 1.143 ~ 6.595 ), sports activities (P= 0.004, OR = 0.297, OR 95% CI:0.130 ~ 0.678 ), personality(P= 0.011, OR = 0.435, OR 95% CI:0.229 ~ 0.829 ) were significantly associated with acute coronary syndrome in multivariate conditional logistic analyses after adjusting other factors. Conclusions Smoking history,hypertension, personality are risk factors of acute coronary syndrome of Northern Chinese Han population. Butsports activities is protective factors.

3.Detection and susceptibility to antibiotics of Mycoplasma in genitourinary tract during 2000~2004

Objective To investigate infection rate and susceptibility to antimicrobial agents of Mycoplasma urealyticum and Mycoplasma hominis in the genitourinary tract, and to provide a reference for clinical diagnosis and therapy. Methods Genitourinary secretions were collected with swabs. They were cultured with the diagnostic kit of Mycoplasma (Biomerieux Company) to detect M. urealyticum and M. hominis. Meanwhile the susceptibility of Mycoplasma against 9 antimicrobial agents was tested with the same kit. According to the manual of the kit, the results were read. The data were statistically analyzed with WHONET5.1 and SPSS. Results A total of 1 008 samples were collected, and the positive rate was 77.5%. Among 781 positive cases of Mycoplasma, 572 were M. urealyticum(56.0%), 40 were M. hominis (4.0%), and 169 were M. urealyticum combined with M. hominis (16.7%). The susceptibility rate of M. urealyticum to Doxycycline, Josamycin, Ofloxacin, Erythromycin, Tetracycline, Ciprofloxacin, Azithromycin, Clamycin and Pristinamycin was 89.3%, 95.1%, 17.5%, 64.3%, 84.1%, 14.8%, 77.8%, 90.8% and 96.5%, respectively. The susceptibility rate of M. hominis to the above drugs was 86.0%, 78.9%, 23.2%, 0, 73.2%, 57.1%, 0, 7.7% and 83.9%, respectively. 4.Distribution and resistance of enteric pathogenic bacteria in Beijing area

Objective To monitor the distribution and resistance of enteric pathogenic bacteria in Beijing area to offer the data for guiding epidemiologic study and clinical treatment. Methods Enteric pathogenic bacteria were cultured and identified to spicies, group, and serotype with the biochemical and serologic test. Then, the susceptibility of bacterium to antimicrobial agents were tested. Results Enteric pathogenic bacteria infection occurred with male, children, and youth being prominant. It peaked in June and July. Shigellae spp and Vibrio spp were the main pathogenic bacteria of intestinal tract. There presented difference among the sensitive rates of differential spicies, or groups to antimicrobial agents. Conclusions There are many spicies of enteric pathogenic bacteria causing infective diarrhea in Beijing area. Their distributions are different in sex, age and season. The is resistance rate are different needing surveillance.
5.Application of PCR-RFLP technique on identification and genotyping of Brucella spp

Objective To amplify the 16S RNA fragments of 7 clinically isolated strains of Brucella spp. by PCR-RFLP technique, so as to provide experimental basis for the studies on diagnostics, genetics and epidemiology of Brucella spp. Methods According to the gene sequence of ATCC 25840 standard strain in GenBank, special primers for the 16S RNA conservative area in the Brucella spp. were designed. DNA extraction and PCR amplification of the 16S RNA fragments were performed with the 7 isolated strains. PCR products were then sequenced and RFLP analysis was conducted with appropriate restricted enzymes to study the homology and the mutation sites in those strains. Meanwhile, the clinical data of infected patients were retrospectively analyzed to evaluate the relationship between the clinical features and genotypes of Brucella infection. Results The amplified target fragments were about 1500bp in length and consistent with what was expected. The sequencing and homology analysis showed a 98.88% homology and 11 mutation sites among the 7 isolated strains. Four genotypes were identified by RFLP. Retrospective analysis of the clinical data indicated that no obvious relationship existed between the genotypes and the clinical features. Conclusions Amplifying 16S RNA fragments by PCR technique is a feasible method to make an early diagnosis of Brucella infection. The 7 clinically isolated strains are different in genotypes and 16S RNA fragment is a highly conservative fragment in bacterial genome with some mutations. The research provides evidence for the genetics and epidemiology of brucellosis.
6.Roles of Akt pathway and cell apoptosis in leptin-mediated chronic mor-phine antinociceptive tolerance

AIM:To explore the roles of Akt ( also called protein kinase B ) and active caspase-3 in the leptin-mediated chronic morphine antinociceptive tolerance in rats .METHODS: A model of chronic morphine antinociceptive tolerance was established in the SD rats .The protein levels of spinal Akt and cleaved caspase-3 were tested by Western blotting.The technique of immunohistochemical staining was used to detect the immunoreactivity positive cells of phospho -rylated ( p)-Akt and cleaved caspase-3 in the spinal cord .Double staining of immunohistochemistry was used to examine the cellular location of the p-Akt and cleaved caspase-3 positive cells.RESULTS: The chronic intrathecal injection of morphine (15 μg) for 7 d markedly upregulated the spinal protein levels of p-Akt and cleaved caspase-3 in the rats.Thirty min before injection of morphine , intrathecal injection of leptin antagonist (3μg) for 7 d significantly attenuated the upreg-ulation of the protein levels of p-Akt and cleaved caspase-3 induced by chronic morphine treatment .The p-Akt was exclu-sively observed in the spinal neurons .The cleaved caspase-3 was only localized with the spinal astrocytes .Intrathecal injec-ting the inhibitors of leptin , Akt and caspase-3 ameliorated the chronic antinociceptive tolerance .CONCLUSION: The spinal Akt pathway and active caspase-3 are involved in the leptin-mediated chronic morphine antinociceptive tolerance in rats.
7.Effect of simvastatin on angiotensin Ⅱ-stimulated secretion and proliferation of adrenocortical carcinoma H295R cells

Objective To investigate the potential effects of simvastatin on angiotensin Ⅱ-stimulated secretion and proliferation of adrenocortical carcinoma H295R cells.Methods The H295R cells were divided into control group, Angiotensin Ⅱgroup, simvastatin group and Angiotensin Ⅱ plus simvastatin group.Cortisol in medium was determined by chemiluminescent method , and aldosterone was determined by radioimmunoassay .The mRNA expression of 11 beta-hydroxylase ( CYP11B1 ) and aldosterone synthase ( CYP11B2 ) were examined by RT-qPCR.Cell proliferation was detected by MTS method.Results Compared with control group, angiotensin Ⅱincreased the secretion of cortisol and aldosterone, and the expression of CYP11B1 and CYP11B2.Simvastatin decreased cortisol secretion and CYP11B1 mRNA expression ( P<0.05 ) .Simvastatin also inhibited angiotensinⅡ-induced the secretion of cortisol and aldosterone , and the expression of CYP 11 B1 and CYP11 B2 compared with Angiotensin Ⅱgroup ( P<0.05 ) .Angiotensin Ⅱhad no effect on cell proliferation , while simvastatin significantly inhibited cell proliferation .The inhibitory effect of simvastatin on proliferation was enhanced when simvastatin was prescribed with angiotensin Ⅱ( P<0.05 ) .Conclusions Simvastatin inhibits angiotensin Ⅱ-induced secretion of cortisol and aldosterone in H295R cells.Simvastatin inhibits cell proliferation, which could be enhanced by angio-tensin Ⅱ.
8.Effects of Apigenin on Platelet Derived Growth Factor-induced Migration of Vascular Smooth Muscle Cells

Objective To investigate the effects of apigenin on the migration of vascular smooth muscle cells (VSMC) induced by platelet derived growth factor (PDGF)-BB and the possible molecular mechanism. Methods VSMCs were isolated from thoracic aortas of male Sprague-Dawley rats using enzyme digestion method. Migration of VSMCs was determined by transwell assay. Western blotting was carried out to evaluate phosphorylation of c-jun N-terminal kinase (JNK). Results Treatment with PDGF-BB (20 ng·mL-1 ) significantly promote VSMC migration,the number of migrated cells was 2. 46 times than that of control group. However,after 12. 5 μmol·L-1 apigenin pretreatment,the number of migrated cells was 46. 5% of the PDGF-BB group. Various dose of apigenin can significantly inhibit VSMC migration induced by PDGF-BB,12. 5 μmol · L-1 apigenin treatment significantly inhibited PDGF-BB phosphorylation of JNK. Conclusion Apigenin can suppress the migration of VSMC induced by PDGF-BB. These beneficial effects on VSMC were at least partly mediated by the inhibition of activity of JNK.
9.Levonorgestrel-releasing intrauterine system for treatment of dysmenorrhea associated with adenomyosis

Objective To evaluate the efficacy and safety of a levonorgestrel-releasing intrauterine system(LNG-IUS)for the treatment of dysmenorrhea associated with adenomyosis.Methods We recruited 48 women with moderate or severe dysmenorrhea associated with adenomyosis.All women were inserted of LNG-IUS into their uterine cavity from days 5-7 of their periods and maintained for 12 months.We compared the visual analogue scale(VAS)scores and verbal rating scale(VRS)scores of their dysmenorrhea and dyspareunia at baseline and 12 monthes follow-up.Results Forty-four women completed the study. There were significant differences between mean VAS and VRS scores changes of dysmenorrhea and dyspareunia at baseline and 12 monthes follow-up,those of dysmenorrhea dropping from 75?13 to 11?11 and 2.3?0.4 to 0.4?0.3,those of dyspareunia dropping from 54?19 to 4?4 and from 1.6?0.8 to 0.2?0.2 respectively.Overall 29 women(66%)were very satisfied or satisfied with the one-year treatment. Conclusion Insertion of LNG-IUS alleviates moderate or severe dysmenorrhea associated with adenomyosis remarkably.
10.Review analysis of reported cases of myocarditis induced by immune checkpoint inhibitors

Objective:To explore the clinical characteristics and influencing factors of myocarditis induced by immune checkpoint inhibitors (ICIs).Methods:Using programmed death receptor-1 (PD-1), nivolumab, pembrolizumab, programmed cell death receptor ligand-1 (PD-L1), atezolizumab, durvalumab, avelumab, cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), ipilimumab, tremelimumab as keywords respectively, we combined these words with myocarditis or the corresponding Chinese to search.Results:A total of 49 articles were reported, including 64 patients. Nivolumab was the most reported, followed by pembrolizumab and ipilimumab. The average age was (65.47±13.24)years, mainly elderly patients; 37 cases (57.81%) were male; the overall mortality rate was 31.25%(20/64). The clinical symptoms were diverse and nonspecific, with dyspnea being the most common (39/64, 60.94%). Heart biomarkers were elevated in 94.64%(53/56) of the patients. 35 patients (54.69%, 35/64) developed myocarditis after 1-2 doses and 17 patients died. 60 patients received steroids as initial treatment, and immunosuppressive therapies such as infliximab, intravenous immunoglobulin, antithymic globulin, and/or plasmapheresis were used in 25 patients, symptoms improved in 17 cases (68.00%).Conclusions:ICIs can cause myocarditis, with high mortality, and should be closely monitored and timely treatment. Steroids can be used as initial first-line therapy and immunosuppressants and/or plasmapheresis may improve clinical symptoms and survival rate.
