1.Magnetic resonance imaging study of twisting or untwisting Taixi acupoint(KI3) on brain function

OBJECTIVE:To identify the influence of twisting or untwisting Taixi(K13) on the brain function by observing functional mag-netic resonance imaging.METHODS:Twelve healthy young cases were enrolled in this research.A block design including three blocks which baseline and stimulation appear alternately was adapted.In stimulation phase,the needle was twisted manually,or not twisted.Scanned images were analyzed using SPM2.RESULTS:The right superior temporal gyrus(BA22),left medial frontal gyrus(BA46),followed by right and left postcentral gyrus of parietal lobe(BA2,BA3),left inferior frontal gyrus(BA45) and left inferior parietal lobule(BA40) were evoked by acu-puncture at Taixi,but there was no activeation area after untwisting acupoint.CONCLUSION:The different areas evoked by twisting or untwisting Taixi acupoint are closely related with the internal organs as well as the course of the channels and collaterals.
2.Effect of diabetic neuropathy on renal 1-? hydroxylase and bone mineral density in old type 2 diabetic rats

Objective To investigate the change of renal 1-alpha hydroxylase and its effect on bone mineral density in the old rats with type 2 diabetic nephropathy. Methods Forty Wistar rats of 18 months old were randomly divided into normal control (N), diabetes (D), diabetes treated with vitamin D_ 3 (T1) and diabetes treated with 1-?(OH)D_ 3 (T2) groups, respectively. Ten rats in each group. Dual energy X-ray absorption (DEXA) was used to determine bone mineral density (BMD) of lumbar spines and femoral bone. 24 h urinary protein excretion, serum 25(OH)D_ 3 and 1,25(OH)_ 2 D_ 3 were measured by radioimmunoassay. Results Compared with controls, 24 h urinary protein excretion increased remarkably D, T1, T2 groups, while BMD greatly decreased, much lower in D group and T1 group than T2 group (P0.05). 5.Study on the mechanism of primary tumor treatment promoting tumor metastasis

Tumor metastasis is a primary cause of leading to death of tumor patients,and difficult to control.The relationship between primary tumor(PT)and metastasis tumor(MT)is extremely complicated. Considerable animal experiments and clinical data confirm that treatment of PT encourages the growth of MT, but the mechanism is not still fully clear.Some studies show that the density depression of antiangiogenesis factor and immune escape of tumor cell are main reasons for MT accelerated growth.This article reviews the animal experiment,clinical observation and occurrence mechanism of primary tumor treatment promoting tu- mor metastasis.
6.Exenatide Weaks Tumor Angiogenesis within Endometrial Cancer Ishikawa Xenografts in Nude Mice

[Objective] To investigate the role of exenatide on angiogenesis in endometrial cancer Ishikawa xenografts in nude mice.[Method] We used the subcutaneous human endometrial cancer cell Ishikawa xenografts in nude mice model,and divided them into control group and exenatide-treated group.The harvested tumors were preserved for immunohistochemistry staining and western blot analysis.[Results] The micro-vessel density (CD31 positive) in exenatide-treated group was (13.2 ± 1.4)/400 power field,less than that in control group [(25.9 ± 5.8)/400 power field] (P < 0.01).The expression level of VEGF was significantly lower in exenatide-treated group than that in control group (P < 0.05).The mean density of tumor associated macrophages (F4/80 positive) is (31.4 ± 3.4)% in exenatide-treated group and (72.1 ± 4.2)% in control group with significant difference (P < 0.01).[Conclusion] Exenatide weaks tumor angiogenesis within endometrial cancer Ishikawa xenografts in nude mice.
7. HPLC fingerprint and simultaneous determination of nine components in Taohong Siwu Decoction of classical prescriptions

Objective: To establish chemical fingerprint and multi-components determination of 15 batches of Taohong Siwu Decoction (TSD), and provide reference for the improvement of its quality control. Methods: The separation was performed on Thermo Hypersil Gold C18 column (250 mm × 4.6 mm, 5 μm) for gradient elution with methanol-0.1% phosphoric acid aqueous solution, flow rate 1.0 mL/min, column temperature 30 ℃, and detection wavelength 225 nm. The HPLC fingerprint was established and evaluated by the similarity evaluation system of TCM (version 2012A), and the difference of chemical information between 15 batches of different samples was evaluated by cluster analysis. Furthermore, the content of the nine active components in the sample was determined by HPLC multi-component wavelength switching method, with the partial least squares-discriminant analysis (PLS- DA) by SIMCA 14.1 software to find significant components of the quality between the batches. Results: The HPLC fingerprint of 15 batches of TSD was established. The similarity was greater than 0.96, and 35 common peaks were identified as gallic acid, chlorogenic acid, amygdalin, albiflorin, hydroxysafflor yellow A, paeoniflorin, ferulic acid, senkyunolide I, benzoylpaeoniflorin and ligustilide (corresponding to peaks 2, 8, 9, 13, 14, 15, 16, 25, 31, and 32). The linearity relationships of gallic acid, 5-hydroxymethylfurfural, chlorogenic acid, albiflorin, hydroxysafflor yellow A, paeoniflorin, ferulic acid, verbascoside, and senkyunolide I (r ≥ 0.999 6) were good. The results of content determination respectively were 187.5-344.4, 6.2-154.8, 413.2-459.2, 507.5-923.5, 873.8-1 202.0, 2 122.3-2 782.9, 59.2-121.3, 6.4-26.9, and 38.9-79.6 μg/g, respectively, including higher content of paeoniflorin, hydroxysafflor yellow A, and albiflorin. Furthermore, 15 batches of samples from different origins were classified into three categories. Using PLS-DA analysis, the content determination result showed that paeoniflorin, albiflorin, hydroxysafflor yellow A, and 5-hydroxymethylfurfural were the four components that affected the quality of different batches of TSD. Conclusion: HPLC fingerprint combined with multi-components determination is suitable for quality control and evaluation of TSD preparation.
8.Correlation between RAR-βgene methylation and p53 gene mutation in bronchoalveolar lavage fluid in non-small-cell lung cancer

Objective To investigate the clinical significance and correlation between RAR-βgene methylation and p53 gene mutation in bronchoalveolar lavage fluid(BALF)in non-small-cell lung cancer.Methods BALF samples from 85 lung cancer pa-tients(lung cancer group)and 70 cases(benign lung diseases group)with benign lung diseases were collected.RAR-βgene methyla-tion in BALF samples was detected by methylation-specific PCR (MSP),and p53 gene mutation was detected by PCR and DNA se-quencing method.Results The rate of RAR-βmethylation and p53 mutation in BALF in lung cancer were 49.4% and 36.5%,re-spectively.Both were higher than in benign lung diseases group(P <0.01).RAR-βmethylation rate(32.5%)of patients with TNM stages(Ⅰ+Ⅱ)(32.5%)was higher than the p53 mutation rate(12.5%)over the same stages (P <0.05).RAR-βmethylation rate and p53 mutation rate of patients with stages(Ⅲ+Ⅳ)were higher than those with stages(Ⅰ+Ⅱ)(P <0.01).p53 mutation rate in lung cancer patients with RAR-βmethylation was higher than those with unmethylated(P <0.01).RAR-βmethylation rate of lung cancer patients with p53 mutation was higher than those without p53 mutation(P <0.01).Conclusion Detection of RAR-βmethyl-ation and p53 mutation in BALF contribute to the diagnosis of lung cancer.
9.Effect of phacoemulsification on corneal endothelium of cataract with lupus nephritis

To evaluate the influence of phacoemulsification on corneal endothelium of senile cataract with lupus nephritis (LN).
●METHODS:This clinical trial involved 40 cataract patients with lupus nephritis (40 eyes), and 50 cases (50 eyes) without lupus nephritis. All of them underwent phacoemulsification+lOL implantation. The parameters of corneal endothelial cell including central corneal endothelium cell density ( CED ), average area of endothelial cell ( AVE) and coefficient of variation ( CV) were recorded by corneal endothelial microscope pre -operation and at one month after operation. The data were analyzed by SPSS 13. 0 statistical software.
● RESULTS: Both LN group and control group, the morphology of coneal endothelial was statistical significant differences between pre - operation and 1mo postoperation. The CED was lower, AVE and CV were higher ( P < 0. 05, respectively). A significant decrease in CED was seen in the LN group than did in the control group (P < 0. 05). Compared to control group,the post -operative AVE and CV in LN group was significantly increased (P<0. 05).
● CONCLUSlON: The corneal endothelial cell in lupus nephritis patients is more fragile. Safe and reliable operation should be selected for these patients.
10.INVESTIGATION OF BONE METABOLISM IN GRAVES' DISEASE

Serum Ca, P, AKP, plasma iPTH, cAMP, urinary Hyp and BMC were measured in 31 cases of Graves' disease. The results showed that about 66.67% of the patients (including 21 controlled and 10 uncontrolled cases) had a decreased BMC, while serum Ca, P, plasma iPTH, cAMP were within the normal range. The data also showed that about 54.6% of the patients had increased serum AKP, but it would not exceed 25 U if no hepatic disease existed. There were positive correlations between serum T, and urinary Hyp, and between urinary Hyp and serum AKP, but a negative correlation between urinary Hyp and BMC. It was suggested that serum AKP and urinary Hyp could be considered as the indices of negative balance of bone metabolism in Graves' disease, and probably it was induced by excessive thyroid hormone directly.
