1.The effects of HtrA on the expression and biological activity of GTFs of Streptococcus mutans isolated from children with high cario-susceptibility

Objective:To compare the expression and biological activity of glucosyl transferases (GTFs)of Streptococcus mutans (S.mutans)under normal outside environment between high temperature requirment serine proteinase A(HtrA)-deficient strains and high virulent strains isolated from children with high cario-susceptibility.Methods:The HtrA-deficient strains and high virulent strains of S.mutans were obtained by preliminary study.The strains were reanimated and incubated in BHI medium to exponential phase at tenth hour.The expression of gtfB,gtfC and gtfD were detected by real-time RT-PCR.The biological activity of the GTFs were detected by entong sulfuric acid method and Western Blot.Results:The expression of gtfs and GTFs in the HtrA-deficient strains was higher than those of high virulent strains,but the biological activity of the GTFs was lower.Conclusion:The HtrA gene plays an important regulatory role in the process of the GTFs expression of S.mutans isolated from children with high cario-susceptibility.
2.Effect of Cucumber Mosaic Virus-encoded 2b Protein on Photosynthesis and Chloroplast Structure of The Host Plant

The 2b protein encoded by Cucumber mosaic virus(CMV)plays an important pathogenicity role in many solanaceous hosts,but mechanism of inducing disease is still unknown.In order to investigate virulence of the 2b protein on Nicotiana glutinosa plants,in terms of chloroplast structure and photosynthesis,a mutant Fny-CMV?2bpro,which cannot express the 2b protein,was achieved by introducing mutant sites in the 2b gene of Fny-CMV.N.glutinosa seedlings were inoculated with wild-type Fny-CMV and the mutant Fny-CMV?2bpro,and were analyzed for symptom expression,chlorophyll content,photosynthetic rate,and ultra-structural alteration of chloroplast.Up to 30 days post inoculation,wild-type Fny-CMV caused symptoms of severe mosaic,leaf deformation,and stunting,reduced photosynthetic rate and chlorophyll content,and altered the ultra-structure and morphological characters of the chloroplasts.However,host seedlings inoculated with the mutant Fny-CMV?2bpro expressed only slight mosaic symptom.Their photosynthetic rates and chlorophyll contents were not significantly different from those of the mock-inoculated plants,and the ultra-structure and morphological characters of their chloroplasts appeared to be normal.The observed low photosynthetic rates and chlorophyll contents were related to the breakage of the chloroplast morphology and ultra-structure.Results of Northern blotting showed that the virulence of 2b protein was associated with high accumulation level of CMV progeny RNAs in systemic leaves.Non-expression of the 2b protein reduced the accumulation levels of its genomic RNAs 1 and 2.The level of subgenomic RNA4,encoding CP protein,was found to be significantly decreased.
3. T-lymphocyte subsets in peripheral blood of children with hand, foot and mouth disease: a Meta analysis

Objective:
To evaluate the changes of T-lymphocyte subsets in peripheral blood of children with hand, foot and mouth disease(HFMD).
Methods:
CNKI, Wanfang, VIP, CBM, PubMed, and Springerlink were searched for articles published from January 2014 to October 2017 with "HFMD, lymphocyte subsets, T-lymphocyte subsets" . Two reviewers selected the studies and extracted the data independently. We performed data analysis using RevMan 5.3 and Stata 14.0 software.
Results:
Twenty-three studies were included in this meta-analysis. Mild vs. severe: The mean difference (MD) and 95% CI of CD3+ CD4+ T cells, CD3+ CD8 + T cells, CD3+ T cells and CD4/CD8 were 7.27 (5.91, 8.63), 3.40 (0.68, 6.12), 11.56 (8.59, 14.53), 0.24 (0.14, 0.34), higher than the severe group. Mild vs. control: The MD and 95% CI of CD3+ CD4+ T cells, CD3+ CD8+ T cells, CD3+ T cells and CD4/CD8 were -5.62 (-7.16, -4.07), -1.79 (-2.79, -0.79) -6.85 (-9.63, -4.07), -0.27 (-0.48, -0.05) respectively, lower than those in healthy controls. Severe vs. Ccontrol: The MD and 95% CI of CD3+ CD4+ T cells, CD3+ CD8+ T cells, CD3+ T cells and CD4/CD8 were -12.03 (-14.99, -9.07), -5.45 (-7.47, -3.43) -18.52 (-24.92, -12.12), -0.46 (-0.71, -0.20) respectively, lower than those in healthy controls.
Conclusions
T-lymphocyte subsets in children with HFMD were related to their condition of disease and declined as the disease worsened.
4.Effect of preoperative thyroid dysfunction on the 30-day postoperative mortality and complications in elderly patients with hip fracture

Objective:To investigate the influence of preoperative thyroid dysfunction on the 30-day postoperative mortality and complications in elderly patients with hip fracture.Methods:A retrospective analysis was conducted of the 349 elderly patients with hip fracture who had been admitted to Department of Orthopedic Trauma, Beijing Luhe Hospital Affiliated to Capital Medical University from January 2018 to December 2019. They were 108 males and 241 females, with an average age of 76.3 years (from 60 to 104 years). There were 190 femoral intertrochanteric fractures and 159 femoral neck fractures. By the preoperative level of thyroid function, the patients were divided into a normal function group of 290 cases and a dysfunction group of 59 cases. The 2 groups were compared in terms of hospital stay, mortality and incidence of complications within 30 days postoperation.Results:In this cohort, the rate of 30-day postoperative mortality was 3.4%(12/349) and the incidence of 30-day postoperative complications 14.6%(51/349). The 2 groups were comparable because there was no significant difference between them in the preoperative general data except for the preoperative comorbidity of coronary heart disease ( P>0.05). In the dysfunction group, the hospital stay averaged (10.2±6.9) d, the rate of 30-d postoperative mortality 1.7%(1/59) and the incidence of 30-day postoperative complications 16.9%(10/59), which were insignificantly different from those in the normal function group [(10.7±7.5) d, 3.8%(11/290) and 14.1%(41/290), respectively] ( P> 0.05). Conclusion:Since preoperative thyroid dysfunction does not affect the 30-day postoperative mortality and postoperative complications in the elderly patients with hip fracture but no definite thyroid disease, routine thyroid function screening is not recommended for them.
5.Clinical application of low-dose spiral CT for orthodontics

Objective To determine the effect of reducing the value of mA or kV on the image quality and the radiation dose of the patients undergoing low-dose spiral CT for orthodontics.Methods Thirty patients were divided into three groups,each group has 10 patients.They were group 1(80 kV and 200 mA),group 2 (120 kV and 80 mA),group 3(120 kV and 200 mA)The volume CT dose index(CTDI)was recorded and the average dose-length produce(DLP)was calculated in three groups,respectively.Image quality of three groups were compared and scored by two radiologists,and the results were statistically analysed.Results The CTDI among three groups in diagnostic image quality.Conclusions Low-dose spiral CT for orthodontics,especially the low-kV scan,may decrease the radiation exposure and guarantee the image quality.
6.Treatment research and new progress of early-onset scoliosis

BACKGROUND:Early-onset scoliosis is a kind of disease that seriously affects the growth of children’s spine and development of cardiorespiratory function. The treatment of the disease has always been the focus of many clinical researchers.OBJECTIVE:To analyze the therapy for early-onset scoliosis and explore the spinal fusion, spinal non-fusion, conventional growth rod technology and magnetic control ed growth rod technology of early-onset scoliosis. METHODS:We retrieved PubMed, CENTRAL, EMbase, the ISI Web of Knowledge Databases, VIP, CNKI, CBM and Wanfang Database for related studies published from inception of the database to March 2016. The key words were“scoliosis, growing rod, complications”. The included 54 studies were analyzed and discussed. RESULTS AND CONCLUSION:For these children of early-onset scoliosis, we should not only maintain the correction of spine deformities, but also protect the ability of spine growth, keeping the normal cardiopulmonary function. In addition to conventional (non-surgical) treatment, there are surgical treatment (such as spinal fusion and growing rod technique) and magnetical y control ed growing rod, a new technology for the treatment of early-onset scoliosis. A comprehensive understanding of the effect of surgical treatment on the spine growth and cardiopulmonary function of children with early-onset scoliosis wil help to prevent the occurrence of related complications, so as to obtain a better therapeutic effect.
7.Death patterns of patients in Department of Respiratory Care Unit of General Hospi-tal of PLA

Objective To analyze retrospectively the death pattern, risk factors, and death time of 253 patients at the Respiratory Care Unit of General Hospital of PLA in order to improve care quality and reduce mortality.Methods The information of patients was extracted from the hospital information system ( HIS) , and then classified and calculated accord-ing to different time points.Results Between November and next March,the mortality rate was higher than in other months (P<0.05), accounting for 19.5%.Mortality of those admitted between 8∶01 and 9∶00 or between 23∶01 and 24∶00 was higher than at other times(P<0.05), accounting for 41.7%and 50.0%, respectively.There was statistically significant difference(P<0.01) in mortality between days of the week,with the highest on Saturday, accounting for 43.1%.Mortality on non-work days was higher than on workday(P<0.01), accounting for 38.3% and 13.2%, respectively.Mortality at off-hour was higher than at office time(8∶00-11∶30 and 14∶30-18∶00 on workday) (P<0.01), accounting for 31.3%and 5.2%, respectively.Logistic regression analysis showed that age, month of admission, and the hour of discharge were associated with the outcome.Conclusion The high mortality between November and next March may be related to the higher incidence of respiratory diseases in winter, air pollution and cold weather.High mortality is also significantly associ-ated with the care quality of the medical staff.
8.Role and mechanism of autophagy in the arsenic trioxide-induced death of Burkitt lymphoma Raji cells

Aim To investigate the role of autophagy and its mechanism in Raji cell death induced by arse-nic trioxide. Methods Transmission electron micros-copy ( SEM) and MDC fluorescence staining were used to observe autophagy. MTT colorimetry was employed to assay the cellular proliferating activity. Cell apopto-sis and cell cycle analysis were performed using FITC-Annexin-V/PI double staining and flow cytometry ( FCM) . The expressions of LC3 and the conversion of LC3-I to LC3-II were measured by western bloting. The expression of bcl-2 mRNA and p53 mRNA were detected by reverse transcription-polymerase chain re-action ( RT-PCR ) . Results Arsenic trioxide could obviously inhibit the proliferation of Raji cells, arrest the cells at G2/M phase and induce apoptosis. Mean-while, arsenic trioxide markedly inhibited the expres-sion of bcl-2 mRNA and enhanced the expression of p53 mRNA in Raji cells. Arsenic trioxide also induced autophagy synchronously which paralleled with the in-duction of apoptosis in Raji cells, and 3-MA, an auto-phagy inhibitor, was able to reverse the arsenic triox-ide-activated autophagic activity, up-regulate bcl-2, down-regulated p53 expression and suppress the lethal effect of arsenic trioxide on Raji cells to reduce their sensitivity to arsenic trioxide. In contrast, the Rapamy-cin, an autophagy inducer, possessed the completely opposite effects on Raji cells compared with 3-MA. Conclusions The apoptosis and autophagic cell death are coexistent in arsenic trioxide-triggered death of Raji lymphoma cells, and Bcl-2 and p53 may play a key regulating role in this process.

Treatment of calcaneal fractures has always been the focus of many clinical researchers.The goals of traditional surgical treatment are not only to restore the integrity of calcaneal articular surface but also to reconstruct the anatomy of the calcaneus.More importantly,we need to reduce postoperative soft tissue swelling and incidence of postoperative complications.In recent years,scholars have reported satisfactory clinical efficacy and prognosis resulting from a sinus tarsal approach for treatment of calcaneal fractures.This paper reviews the latest research progress concerning the sinus tarsal approach for treatment of calcaneal fractures at home and abroad,intending to provide helpful information for the clinical surgeons.

AIM: To investigate the correlation between renal transforming growth factor-?1 (TGF-?1) expression and cellular apoptosis, and effect of tea polyphenols (TP) on TGF-?1 expression and apoptosis. METHODS: Four group animals in a rat model of CsA-induced chronic nephrotoxicity were respectively treated by vehicle (olive oil, 0.1 mL?kg~(-1)?d~(-1), sc), TP (80 mg?kg~(-1)?d~(-1), ig), CsA (15 mg?kg~(-1)?d~(-1), sc) and TP (80 mg?kg~(-1)?d~(-1), ig) plus CsA (15 mg?kg~(-1)?d~(-1), sc). At the end of 28th day of treatment, renal creatinine clearance and tissue pathology were analyzed. The TGF-?1 expression was detected by immunohistochemistry and Western blotting.
