1.Early diagnosis and microsurgical treatment of cerebral arteriovenous malformation hemorrhage in childhood

Objective To investigate the diagnostic method,microsurgical operation time and effect of brain arteriovenous mal-formation(AVM)rupture hemorrhage in childhood.Methods 72 children (≤16 years old)of AVM hemorrhage from 2003 to 2013 were retrospectively analyzed.Results The CT diagnosis rate was 48.6%(35/72),the MRI diagnosis rate was 78.3%(47/60),56 cases were performed the early whole brain DSA under the condition permitting,its diagnosis rate was 98.2%(55/56).All the cases were performed the hematoma clearance plus the pertinent lesion resection.70 cases were re-examined by DSA/CTA/MRA during postoperative perioperation period.The lesion resection rate was 91.5%,the partial resection rate was 8.6%.60 cases(85.7%) were successfully followed up for 6 months to 6 years and assessed by the Glasgow outcome scale,5 points in 5 5 cases,4 points in 2 cases,3 points in 2 cases and 2 points in 1 cases.Conclusion DSA is the main detection measure for early diagnosing AVM hem-orrhage and the microsurgical operation in the acute stage can reduce the fatality and improve the prognosis.
2.A New Internal Quality Control Rules Design Tool in Clinical Laboratory-Westgard Sigma Rules

The right internal quality control rules are really important for improving the quality of patient results report in clinical laboratory.In order to look for faster and simpler tools that will help laboratories select the right internal quality con-trol rules for their own applications,recently Westgard introduced a newer tool “Westgard Sigma Rules”which is quicker and easier to use than previous tools.And it is very easy to figure out the right internal quality control rules by using West-gard Sigma Rules.This article introduced the characteristics and usage of original Westgard Rules and sigma metrics graph, and focusing on the application methods and practices of Westgard Sigma Rules which is the combination of original West-gard Rules and sigma metrics graph.Once the sigma quality of the tests and methods in laboratories are calculated,the West-gard Sigma Rules diagram can make it easy to select the right control rules and the right number of control measurements.
3.Effects of Buyang Huanwu Decoction on Contents of PCⅢ and Ⅳ-C in Blood Serum of Pulmonary Fibrosis Rats

Objective To discuss intervention effects of Buyang Huanwu decoction on pulmonary fibrosis (PF) rats. Methods Totally 144 healthy male SD rats were randomly divided into six groups:sham-operation group, model group, prednisone group, Buyang Huanwu decoction high-, medium-, low-dose groups, 24 rats in each group. Except for sham-operation group, the rest groups were given injection of BLM via tracheal intubation for the establishment of PF model, while sham-operation group was instilled the same amount of NS. From the second day, all the rats received a gavage by related medicine. Eight rats in each group were killed respectively on day 7, 14 and 28 after intratracheal instillation. The contents of PCⅢ and Ⅳ-C in blood serum of experimental rats were tested by radioimmanoassay method. Results Compared with the sham-operation group, the contents of PCⅢ and Ⅳ-C in blood serum of other groups increased gradually, and that of the model group was the most obvious (P<0.01). Compared with the model group, each medicine group had different decreases in the contents of PCⅢ and Ⅳ-C in blood serum (P<0.01). Compared with the prednisone group, the contents of PCⅢ and Ⅳ-C in blood serum of Buyang Huanwu decoction medium-dose group obviously decreased on the 14th and 28th day (P<0.05), as well as that of Buyang Huanwu decoction low-dose group on the 28th day (P<0.05). Conclusion Buyang Huanwu decoction could effectively reduce proliferation of collagen fibers and excess deposition of ECM in lung tissue of PF, and played an anti-fibrosis effect on lungs.
4.Quality Indicators in Laboratory Medicine:A Fundamental Tool for Quality and Patient Safety

Quality indicators (QIs)in laboratory medicine can measure how well a clinical laboratory meets the needs and re-quirements of users and the quality of all operational processes.The identification of reliable QIs is a crucial step in enabling users to quantify the quality of laboratory services.Different QIs and terminologies are currently used and,therefore,there is the need to harmonize proposed QIs.A model of quality indicators has been consensually developed by a group of clinical la-boratories according to a project launched by a working group of the International Federation of Clinical Chemistry and La-boratory Medicine.The model includes 57 QIs related to key processes (35 pre-,7 intra-and 15 post-examination phases)and 3 to support processes.The developed a model of quality indicators and the data collected provide evidence of the feasibility of the project to harmonize currently available QIs,but further efforts should be done to involve more clinical laboratories and to collect a more consistent amount of data.
5.External quality assessment schemes and standardization/harmonization of clinical laboratory measurement

Although the key role of External Quality Assessment schemes ( EQAS ) in the standardization/harmonization process has been clear from the start, it has never been totally implemented, mainly due to the lack of commutable materials, the difficulty target values and acceptable range.EQAS may also improve standardization/harmonization by proposing rules for the post-examination phase.Improvements in standardization and harmonization will really be able to work under the joint efforts of stakeholders.
6.Effect of type Ⅰ and type Ⅱ quenchers on virus inactivation and red cell cytotoxicity

Objective To research the effect of Hydroxyl radical(type Ⅰ mechanism)and singlet oxygen(type Ⅱ mechanism)on virus inactivation and red cell cytotoxicity induced by DMMB and M007 phototreatment.Methods Hydroxyl radical quenchers mannitol and glycerol,singlet oxygen quenchers sodium azide and tryptophan were employed to investigated their ability to inhibit the inactivation of bacteriophage Phi6 and red cell cytotoxicity by induced DMMB and M007 phototreatment.Results Singlet oxygen quenchers could inhibit the inactivation of bacteriophage Phi6 obviously, whereas hydroxyl quenchers hardly affect the inactivation of bacteriophage Phi6.Both type of quenchers could reduce the hemolysis caused by CMMB or M007 phototreatment.Conclusion Viral inactivation of DMMB and M007 phototreatment is mainly through type Ⅱ mechanism, both type I and type Ⅱ mechanisms contribute to red cell cytotoxicity.

FISH technology has become a routine technique in many clinical laboratories.It can be used for the examination of genetic aberrations that are too small to visualize by conventional cytogenetic assays,but too large to detect by standard DNA sequencing.Today,FISH applications are used not only in medical genetics,but also in many other disciplines including neuroscience,evolutionary biology,microbiology,pathology,toxicology,and reproductive genetics.As FISH technology keep developing,the requirement of the results of FISH is increasingly high.This paper described the quality management for FISH detailedly,aimed at improving the accuracy and reliability of the result of FISH,satisfying the requirements of clini-cians and patients and improving the healthcare quality and outcome of patients,hoping for providing some references for la-boratories who develop FISH technology.
9.Effect of bone xenograft extract on biological characteristics of bone marrow mesenchymal stem cells

BACKGROUND:How to preserve the biological properties and osteogenic activity of bone xenograft materials after antigen removal has become one of the hot spots. OBJECTIVE:To investigate the effect of bone xenograft extract on proliferation, migration and osteogenic differentiation of bone marrow mesenchymal stem cel s. METHODS:Bone marrow mesenchymal stem cel s from Sprague-Dawely rats were cultured respectively in bone xenograft extract (experimental group) and DMEM/F12 medium containing 10%fetal bovine serum (control group). MTT, Transwel chamber and flow cytometry were used to detect cel proliferative ability, migration ability and cel cycle changes. Passage 3 bone marrow mesenchymal stem cel s were selected in the two groups and cultured in osteogenic induction medium. At 7 and 14 days of osteogenic induction, alkaline phosphatase staining was done;at 21 days of osteogenic induction, alizarin red staining was performed to observe calcium nodule formation. RESULTS AND CONCLUSION:After 1, 3, 5, 7 days of culture, the cel proliferative activity and cel cycle had no significant difference between the two groups, but the number of migrating cel s was significantly lower in the control group than the experimental group (P<0.05). At 7 and 14 days of culture, the alkaline phosphatase level was significantly higher in the experimental group than the control group (P<0.05);at 21 days of culture, calcium nodes formed in both two groups, but the calcium nodule area was larger in the experimental group than the control group. These findings indicate that bone xenograft materials have no cytotoxicity but possess good cytocompatibility, which can promote the proliferation, migration and osteogenic differentiation of bone marrow mesenchymal stem cel s.
10.Effects of recipe for invigorating kidney on expression of estrogen receptors in ovariectomized rats

OBJECTIVE: To observe the effects of recipe for invigorating kidney on expression of estrogen receptors in ovariectomized rats. METHODS: The rats, which were given high-cholesterol diet after ovariectomy, were chosen as experimental subjects. After fed kidney-invigorating recipe for 16 weeks, the rats were killed and the levels of serum lipid and estradiol of these rats were examined. The expressions of estrogen receptor alpha and beta mRNAs in aorta, pituitary gland and hypothalamus, and the affinity of estrogen receptor-ligand were also detected. RESULTS: The kidney-invigorating recipe could increase the expression levels of estrogen receptor alpha and beta mRNAs in different parts such as aorta, pituitary gland and hypothalamus, and enhance the affinity of estrogen receptor-ligands. CONCLUSION: The recipe for invigorating kidney may meliorate atherosclerosis in ovariectomized rats by regulating the estrogen receptors.
