1.Expression and significance of Dkk-1 and Bcl-2 protein in sinonasal squamous cell carcinoma

Objective To explore the expression and significance of Dickkopf-1 (Dkk-1) and cell lymphoma/leukemia-2 (Bcl-2) protein in sinonasal squamoucell carcinoma(SNSCC) .MethodThe immunohistochemical SP method and Western blomethod were adopted to determine the expression of Dkk-1 and Bcl-2 in 30 specimenof SNSC(SNSCgroup) ,38 specimenof sinonasal inverted papillomas(SNIP group) and 20 specimenof middle turbinate mucosa(control group) .ResultThe expression of DKK-1 protein in the SNSCgroup wasignificantly down-regulated compared with the SNIP group and the control group ,while the expression of Bcl-2 protein in the SNSCgroup wasignificantly up-regulated compared with the SNIP group and the control group;in the SNSCgroup ,the positive rateof DKK-1 protein and Bcl-2 protein in the high and middle differentiation group and the low differentiation group were 100 .00% ,68 .75% ,33 .33% and 50 .00% ,62 .50% ,100 .00% respectively ,the differencewere statistically significan(P<0 .05) .Conclusion Dkk-1 protein may play an importanpromoting role in the developmenand pro-gression procesof SNSC,the expression of Dkk-1 protein hanegative correlation with the expression of Bcl-2 protein in SNSC,which may become new targespoof SNSCgene therapy .
2.Eosinophilic cystitis (report of 2 cases and review of the literature)

Objective To evaluate the clinical presentations,pathologic characteristics,and the diagnosis and treatment of eosinophilic cystitis.Methods Two cases of eosinophilic cystitis were analyzed,and the relevant literature was reviewed.Case 1,a 63-year-old man,complained of voiding urgency and frequency,dysuria,intermittent hematuria,and lower abdominal pain.Cystoscopic examination showed that an about 5cm?3cm abnormal focus was 0.2-0.4 cm above bladder mucosa at the fundus of the bladder.Case 2,a 42-year-old woman,complained of voiding urgency and frequency,dysuria,and suprapubic pain with intermittent hematuria.Cystoscopic examination showed that about 4 cm?3 cm?2 cm abnormal foci of cauliflower-like were 0.3-0.4 cm above bladder mucosa at the fundus and neck of the bladder.Imaging examinations(Bultrasound,CT and IVU) showed diffuse thickening of bladder wall and tumor-like lesions.They both underwent transurethral resection of the lesions and were treated with oral steroids and antibiotics.Results Postoperative pathology confirmed eosinophilic cystitis in both cases.The follow-up was 6 months.The voiding symptoms disappeared,and imaging examinations showed no space-occupying lesions of the bladder in them.Up to now,there has no evidence of relapse.Conclusions Eosinophilic cystitis is a very rare tumor-like inflammatory disease.It is easily misdiagnosed as bladder neoplasm.Cystoscopy and biopsy are gold standard for the diagnosis.Current treatment modalities include transurethral resection of the bladder lesion along with nonspecific medical therapy.
3.Protective effects of trigonella foenum graecum L. on acute cerebral ischemia

AIM To study the protective effects of trigonella foenum graecum(TFGs) on acute cerebral ischemia. METHODS Acute incomplete ischemia was induced by ligaturing bilateral carotid arterise and cutting heads in mice, the survival time and asthmatic time were observed; the coagulation timewas measured by sheet glass method,and the blood viscosity was also assayed; the platelet aggregation induced by collagen were studied by turbdimetry in vitro. RESULTS TFGs prolonged survival time, coagulation time and asthmatic time significantly and inhibited platelet aggregation ratio in rabbit,and decreased the blood viscosity. CONCLUSION TFGs has the protective effects on acute cerebral ischemia.
4.Study of pectin-adriamycin conjugate to cardiac toxicity in rats

Aim To study the effect of pectin-adriamy-cin conjugate ( PAC) on cardiac toxicity .Methods 50 female SD rats were randomly divided into 5 groups with 10 animals in each group .Adriamycin ( ADM ) group received 3 mg? kg -1 , ip, every other day for 6 times.PAC group received ADM equivalent 1.5,3 and 6 mg? kg -1 , ip, every other day for 6 times.Control group received normal saline parallel to ADM .Rats were sacrificed and the echocardiogram , cardiac en-zymes , the oxidative stress levels in myocardial cells and histopathological changes after 48 h administration were detected.S180 ascites tumor bearing mice models were established to investigate the antitumor activity of PAC.Results The survival rate of ADM group was 50% and that of PAC each group was 100%.PAC could significantly increase body weight ,heart index and immune index and increase HR ,EF,FS,reduce LVIDd, LVIDs.PAC could also significantly increase the AST , LDH, CK, CK-MB level in serum .GSH-Px and SOD activities of PAC group were significantly increased and MDA contents were reduced , and histopathological changes decreased .PAC could effectively inhibit the growth of tumor cells and extend the survival period of mice.Conclusion PAC induces a significant reduc-tion in cardiotoxicity by increasing survival rate , im-mune and cardiac function , improving cardiac en-zymes ,oxidative stress and myocardial cell injury , and also PAC has obvious antitumor effect .

6.Changes of gray matter volume in patients with bipolar disorder and suicidal ideation: a voxel based morphometric analysis

Objective The study aims to analyze the changes in gray matter among Bipolar Disorder (BD) with suicidal ideation, those without suicidal ideation and health control (HC) and to explore the altered brain imaging changes of BD patients with suicidal ideation. Method The study recruited 28 BD with suicidal ideation, 40 BD without suicidal ideation and 46 HCs who undergone the high-resolution structural magnetic resonance imaging. Thus, the data of all participants were analyzed by voxel-based morphometry (VBM). Results Significantly decreased gray matter volume was detected in the right superior temporal gyrus, right middle temporal gyrus, left caudate nucleus, left lenticular putamen, left middle occipital gyrus and left occipital gyrus in patients with BD compared to HCs. However, there was no significant difference in gray matter volume between BD with suicidal ideation and those without suicidal ideation. There were significant differences in gray matter volume in the right postcentral, the right superior parietal gyrus and the right precuneus among BD with and without suicidal ideation and HCs. The gray matter volumes of BD with suicidal ideation were significantly lower than that of the other two groups. Conclusion Gray matter volume abnormalities in parietal lobe play an important role in the brain structural alternation of BD with suicidal ideation.

8.ALA-PDT alleviates graft-versus-host disease in mice following allogenic bone marrow transplantation.

OBJECTIVEThis study investigated the hypothesis that 5-aminolevulinic acid-mediated photodynamic therapy (ALA-PDT) might alleviate acute graft-versus-host disease (GVHD) following allogenic bone marrow transplantation (allo-BMT) in mice.
METHODSAcute GVHD model following allo-BMT was established in 40 recipient BALB/C mice. Fifty C57BL/6J mice were used as donors and another 10 BALB/C mice as blank control without any intervention. Recipients received a lethal dose of 8.5 Gy (60)Co radiation for 10 minutes before transplantation and then were randomly divided into four groups of 10 mice (A-D). Group A was injected with normal saline injection and served as controls. Group B received pure donor bone marrow and spleen cell infusion. Group C received donor bone marrow and mixed donor-recipient spleen cell infusion. Group D was administered with an infusion of donor bone marrow cells and mixed donor-recipient spleen cells treated with ALA-PDT. The 28th day survival rate, incidence of acute GVHD and hematological and pathological changes after transplantation were examined.
RESULTSAll the mice from the Blank control group survived. The survival rates for Groups A-D on the 28th day were 0, 0, 10% and 60% respectively. Group D showed a significantly higher survival rate than the other three groups (P < 0.01). Most of the mice in Groups B and C developed GVHD but only two developed in Group D. Moreover Group D had less severe hematological and pathological changes when compared with Groups B and C.
CONCLUSIONSALA-PDT significantly alleviated GVHD and increased the 28th day survival rate for allo-BMT mice. ALA-PDT may be a promising therapy for GVHD following allo-BMT. Future studies should focus on the underlying mechanism of its therapeutic effect.

9.Expression of integrin?_3 and integrin ?_1 in breast cancer and its clinical significance

Objective To investigate the expression of inregrin?_3 and integrin?_1 in breast cancer and its bio- logical significance.Methods Immunohistochemical assay was used to determine the expression of integrin?_3 and integrin?_1 in the breast cancer(32 cases).Results In normal breast tissue,the positive expression rates of integrin?_3 and integrin?_1 were 0 % and 25 %.In the breast cancer tissue,the positive expression rates of integrin?_3 and inte- grin?_1 were 36 % and 81%.Conclusion The integrin?_3 and integrin?_1 are close associated with the biological sig- nificance of breast cancer.To examine its expression is useful to evaluate the aggressive degree,metastatic potential and prognosis in patients with breast cancer.
10.The effects of compound CX09040 on the inhibition of PTP1B and protection of pancreatic β cells.

To investigate the effects of 2-(4-methoxycarbonyl-2-tetradecyloxyphenyl)carbamoylbenzoic acid (CX09040) on protecting pancreatic β cells, the β cell dysfunction model mice were induced by injection of alloxan into the caudal vein of ICR mice, and were treated with compound CX09040. Liraglutide was used as the positive control drug. The amount and the size of islets observed in pathological sections were calculated to evaluate the β cell mass; the glucose stimulated insulin secretion (GSIS) test was applied to estimate the β cell secretary function; the oral glucose tolerance test (OGTT) was taken to observe the glucose metabolism in mice; the expressions of protein in pancreas were detected by Western blotting. The effects on the target protein tyrosine phosphatase 1B (PTP1B) were assessed by the PTP1B activities of both recombinant protein and the intracellular enzyme, and by the PTP1B expression in the pancreas of mice, separately. As the results, with the treatment of CX09040 in alloxan-induced β cell dysfunction mice, the islet amount (P<0.05) and size (P<0.05) increased significantly, the changes of serum insulin in GSIS (P<0.01) and the values of acute insulin response (AIR, P<0.01) were enhanced, compared to those in model group; the impaired glucose tolerance was also ameliorated by CX09040 with the decrease of the values of area under curve (AUC, P<0.01). The activation of the signaling pathways related to β cell proliferation was enhanced by increasing the levels of p-Akt/Akt (P<0.01), p-FoxO1/FoxOl (P<0.001) and PDX-1 (P<0.01). The effects of CX09040 on PTP1B were observed by inhibiting the recombinant hPTP1B activity with IC50 value of 2.78x 10(-7) mol.L-1, reducing the intracellular PTP1B activity of 72.8% (P<0.001), suppressing the PTP1B expression (P<0.001) and up-regulating p-IRβ/IRβ (P<0.01) in pancreas of the β cell dysfunction mice, separately. In conclusion, compound CX09040 showed significant protection effects against the dysfunction of β cell of mice by enlarging the pancreatic β cell mass and increasing the glucose-induced insulin secretion; its major mechanism may be the inhibition on target PTP1B and the succedent up-regulation of β cell proliferation.
