1.Application of proteomics in study of hepatocellular carcinoma

Hepatocellular carcinoma (HCC)is one of the most fatal malignant tumors worldwide.As an important part of cutting-edge re-search fields,proteomics has been widely used in the studies of related diseases and has currently become a crucial experimental approach to research on HCC.Significantly expressed proteins can be identified as potential biomarkers for early diagnosis and targets for therapeutic drugs for HCC.Moreover,they can be used for prediction of the recurrence and prognosis of HCC,as well as for investigation of pathogene-sis of the disease.The proteomic results from worldwide clinical studies of HCC are summarized,and it is suggested that the clinical applica-tion of results of basic research on HCC proteomics will bring great benefit to the diagnosis and treatment of HCC.
3.Effects of hot-NaOH pretreatment on Jerusalem artichoke stalk composition and subsequent enzymatic hydrolysis.

In order to explore the possibility of Jerusalem artichoke stalk for bioenergy conversion, we analyzed the main composition of whole stalk, pitch, and core of the stalk. Meanwhile, these parts were pretreated with different NaOH concentrations at 121 degrees C. Afterwards, enzymatic hydrolysis was performed to evaluate the pretreatment efficiency. Jerusalem artichoke stalk was characterized by relatively high lignin content (32.0%) compared with traditional crop stalks. The total carbohydrate content was close to that of crop stalks, but with higher cellulose content (40.5%) and lower hemicellulose (19.6%) than those of traditional crop stalks. After pretreatment, the lignin content in the whole stalk, pitch, and core decreased by 13.1%-13.4%, 8.3%-13.5%, and 19.9%-27.2%, respectively, compared with the unpretreated substrates. The hemicellulose content in the whole stalk, pitch, and core decreased 87.8%-96.9%, 87.6%-95.0%, and 74.0%-90.2%, respectively. Correspondingly, the cellulose content in the pretreated whole stalk, pitch, and core increased by 56.5%-60.2%, 52.2%-55.4%, and 62.7%-73.2%, respectively. Moreover, increase of NaOH concentration for pretreatment could improve the enzymatic hydrolysis of the whole stalk and pitch by 2.3-2.6 folds and 10.3-18.5 folds, respectively. The hydrolysis of pretreated stalk core decreased significantly as 2.0 mol/L NaOH was employed, although the increased NaOH concentration can also improve its hydrolysis performance. Based on these results, hot-NaOH can be regarded as an option for Jerusalem artichoke stalk pretreatment. Increasing NaOH concentration was beneficial to hemicellulose and lignin removal, and consequently improved sugar conversion. However, the potential decrease of sugar conversion of the pretreated core by higher NaOH concentration suggested further optimization on the pretreatment conditions should be performed.

4.Recombinant adeno-associated virus encoding secreted forms of human acidic fibroblast growth factor transfecting endothelial progenitor cells

Objective To investigate the feasibility of recombinant adeno-associated virus encoding secrected forms of human acidic fibroblast growth factor transfecting endothelial progenitor cells.Methods sp-haFGF was obtained through combining signal peptide sequence of FGF-4 with native aFGF gene by PCR.sp-haFGF was cloned into AAV vector plasmid pAAV-IRES-hrGFP.Recombinant AAV encoding sp-haFGF was packaged through co-transfecting HEK293 cells with plasmid sp-haFGF-pAAV-IRES-hrGFP,pAAV-RC and pHelper.Ex vivo cultured EPCs were infected with concentrated rAAV.Expression of green fluorescent protein(GFP) in infected EPC was observed by fluorescence microscope and expression of sp-haFGF in EPCs was verified by RT-PCR and western blot analysis.Results Recombinant AAV encoding sp-haFGF was obtained.After EPCs being infected with rAAV,green fluorescence was found in about 20~30% EPCs,gene of(560 bp) was generated from EPCs by RT-PCR method,and(sp-haFGF) protein was detected in infected cells.Conclusion EPC was efficiently infected by rAAV encoding(sp-haFGF) and(haFGF) was expressed by EPCs,which establishes a basis for therapeutic angiogenesis by transgenic EPCs transplantation.
5.Investigation of infants/toddlers wheezing affected by the hand foot and mouth disease

Objective To investigate the relationship between the severity of infant/toddlers wheezing and hand,foot and mouth disease (HFMD) infection.Methods We selected infant/toddlers wheezing with HFMD cases in respiratory department of our hospital from January 2010 to January 2012,and 30 cases of enterovirus 71 (EV71) were selected as the experimental group and 30 cases of coxsackie virus A type 16 (CA16)as the control group,respectively,according to HFMD pathogen infection.Wheezing cases by severity were divided into intermittent stares with mild persistent group (A) and moderate and severe persistent group (B).HFMD by severity were divided into ordinary and severe cases group.Wheezing severity before and after HFMD,relationship between HFMD severity and wheezing severity and the content of serum cytokines such as interferon-γ(IFN-γ) and interleukin-4 (IL-4) were compared between EV71 group and CA16 group,and statistical analysis was performed.Results (1) Wheezing severity(A,B) corresponding cases:EV71 group(25,5),(5,25),and CA16 group(24,6),(15,15) before and after HFMD.The case-constituent ratio in EV71 and CA16 same group was compared before and after infection,and the differences were statistically significant(P < 0.05).The differences in EV71 group and CA16 group were not statistically significant (x2 =0.11,P > 0.05)before infection,and were statistically significant(x2 =7.50,P <0.05) after infection.(2) Severe cases,ordinary group and severe wheezing (A,B) corresponding cases:(17,1),(32,10) before infection,and (2,16),(18,24) after infection.The upgrade cases constituent ratio of wheezing severity after infection were compared between severe and ordinary cases group,and the difference was statistically significant (x2 =5.88,P < 0.05).(3) The content of serum cytokines as IFN-γand IL-4 and IFN-γ/IL-4 ratio were compared in EV7l group and CA16 group before and after infection,and the differences were statistically significant (P < 0.001) ; and the differences compared in EV71 group and CA16 group were not statistically significant(P >0.05) before infection and statistically significant(P < 0.05) after infection.Conclusions Infant/toddlers wheezing with HFMD and wheezing severity of severe disease and EV7l infections were exacerbated trend in the short term after infection.The infection of EV71 may promote Th1/Th2 drift.
6.Influence of bone mesenchymal stem cells on expression of transforming growth factor-beta Ⅲ receptor during CD117~+ cardiac stem cells differentiation

BACKGROUND:Bone mesenchymal stem cells(BMSCs) can accelerate vascularization of myocardial cell,relieve myocardial remodeling,and improve heart function.However,whether its paracrine action can facilitate the differentiation of CD117+ cardiac stem cells(CSCs) by effecting transforming growth factor-? Ⅲ(TGF-? Ⅲ) receptor expression is still unknown.OBJECTIVE:To study the influence of BMSCs on the expression of TGF-? Ⅲ receptor in the process of differentiation of CD117+ CSCs.DESIGN,TIME AND SETTING:In vitro cytology contract observation.The experiment was performed at the Xinhua Hospital from February 2008 to February 2009.MATERIALS:Neonatal rats,weighing 5-8 g,were used to prepare CSCs.Sprague Dawley rats,4 weeks old,weighing 200-250 g,were prepared for BMSCs.METHODS:CD117+CSCs were obtained from neonatal rats by explant cultures and magnetic bead cell sorting.Immunohistochemical experiment was performed to identify specificity of CSCs.CD117+CSCs suspension were synchronization without serum for 24 hours,washed by PBS for 3 times,and then cultured with DMEM/F12,followed by inoculation into 6-well plate.The cells were divided into the control and co-culture groups.In the control group,CSCs were induced differentiation by cardiac spheres,and Transwell chamber were used in the co-culture group.MAIN OUTCOME MEASURES:The growth of CSCs was observed by inverted microscope.The changes of cardiac Troponin T(cTnT),connexin43,TGF-? Ⅲ receptor,smad2 and phospho-smad2 expressions were detected by western blot at days 1,3,5 and 7 after culture.RESULTS:①CSCs isolated by explants cultures and MACS expressed CD117,which grew in disperse with smaller body.② The expression of cTnT and connexin43 were significantly higher in the co-culture group than that of the control group at days 5 and 7(P
7.Quality Analysis of Three Components in Shuanghuanglian Powder for Injection Before and After Ultrasonic Atomization

Objective To establish a new RP-HPLC method for simultaneous determination of chlorogenic acid,forsythin, and baicalin in shuanghuanglian powder for injection after ultrasonic atomization. Methods Hypersil ODS2 C18(250 mmí4. 6 mm,5 μm) was used as the chromatographic column. The mobile phase was methanol-0. 2% phosphate acid solution (4060). Flow rate was 1. 0 mL·min-1 . Sample volume was 5μL. Column temperature was 30℃. Detection wavelength was 324 nm at 0-10 min and 277 nm at 10-25 min. Results Contents of chlorogenic acid,forsythin, and baicalin had good linear relationship with the respective peak area (r≥0. 999 7) within the scope of the sample volume. The RSD was <2% for precision, reproducibility, and stability. Recovery rate was 98. 50%-101. 12% (n=6). Conclusion The method is rapid, accurate and reproducible, with high resolution. It can determine the content of three kinds of components at the same time. The three components in shuanghuanglian powder for injection did not change significantly before and after ultrasonic atomization.
8.Changes of Plasma Brain Natriuretic Peptide Concentration Induced Therapy by Daunorubicin and Its Clinical Significance in Children with Acute Leukemia

Objective To explore the clinical significance of the brain natriuretic peptide(BNP) in evaluating the cardiotoxicity caused by daunorubicin(DNR) through studying the changes of the plasma BNP levels in children with acute leukemia who accepted the chemotherapy with DNR.Methods Thirty-one children with acute lymphoblastic leukemia(ALL) admitted in the year of 2002-2004 underwent the chemotherapy in DVLP project.The plasma level of BNP was measured by enzyme linked immunosorbent assay(ELISA) and the left ventricular end diastolic diameter(LVEDD) by color Doppler respectively before and after the administration of DNR.Simultaneously,electrocardiography(ECG) and cardiac muscle enzymes(LDH1,CK-MB) were measured as routine.Results The plasma level of BNP increased from(3.97?2.41) ng/L to(18.25?7.63) ng/L(P
9.Application of bispectral index during sedation in the patients in ICU

ObjectiveTo evaluate the value of bispectral index(BIS) monitoring during sedation in the ICU.Methods60 patients in ICU were randomly divided into three groups. By transfusing propofol and midazolam with injecting pump, the BIS of groups Ⅰ,Ⅱ and Ⅲ were controlled within 75~85, 65~75 and 55~65, respectively. ResultsCompared with the pre-sedation, there was no remarkable change in the patients of groups Ⅰ and Ⅱ after sedation(P>0.05)while there was remarkable change in the patients of group Ⅲ(P<0.05).The average score of Ramsay in groups Ⅰ, Ⅱ and Ⅲ were 2.2, 3.4 and 4.6 while the dose of propofol were (9.54±2.43) μg/kg·min, (12.69±3.12) μg/kg·min, (14.18±2.91) μg/kg·min and the dose of midazolam were (0.23±0.09) μg/kg·min, (0.25±0.07) μg/kg·min, (0.28±0.11) μg/kg·min, respectively.ConclusionThe application of BIS can make good judgement in the sedation, which showed different choices to different needs to obtain optimistical sedation effect.
10.Effect of Continuous Blood Purification on Multiple Organ Dysfunction Syndrome:33 Cases Report

ObjectiveTo study the effect of continuous blood purification (CBP) on the multiple organ dysfunction syndrome(MODS). MethodsContinuous blood purification were used in 33 patients with MODS. Results Among 33 patients,24 mended and 8 died. The scores of Acute Physiology And Chronic Health Evaluation (APACHE Ⅱ) and MODS obviously declined(P<0.05), as well as the BUN and blood Cr (P<0.05), but the bilirubin did not (P>0.05). The haemodynamic variables were stabilized during CBP and no obvious side-effect related to CBP was found. ConclusionThe therapy of continuous blood purification can improve the prognosis to the patients with multiple organ dysfunction syndrome. The patients were able to bear at the quality better.
