2.Effects of estrogen on the expression of stromal cell-drived factor- 1 in human matrix fibroblasts and breast cancer cell lines

ObjectiveStromal cell-derived factor -1 (SDF-1 ) is closely related to the biological characteristics of breast cancer. We aimed to explore whether estrogen affected breast cancer by SDF-1. MethodsThe breast cancer cell line MCF-7 and MRC5 were chosen, and divided into three groups: the control group, the estrogen group and the estrogen + estrogen receptor blocker group. Each group was cultured with different physiological concentrations of 17-β estrogen at certain time, and the same alcohol concentration of 17-β estradiol at different time points, and then the enzyme-linked immunosorbent assay (ELISA) was used to measure the concentration of SDF-1 in culture medium, and the semi-quantitative reverse transcriptionpolymerase chain reaction (RT -PCR) was used to detect the expression of SDF-1 mRNA in each group.ResultsSDF-1 can be detected in the culture medium of both MCF-7 and MRC5 cell lines. All different concentrations of 17-β estradiol may increase the secretion of SDF-1 in MCF-7 cells. When adding 17-β estradiol to the concentration of 107mol/L, the secretion of SDF-1 reached the peak in 2 hours, which was 6 times and 2.7 times that of control group ( P < 0.01 ). The effect could be ehminated by pure estrogen receptor ICI182,780. In addition, the mRNA expression of SDF-1 was consistent with the SDF-1 protein levels-l07 mol/L group. The expression of SDF-1 mRNA was higher than both that of the control group and the blocking group in 2 hours (P < 0.05 ). ConclusionsIn some breast cancer cell lines, physiological concentrations of estrogen can increase the secretion of SDF- 1, and this effect is mainly achieved through the estrogen receptor. Estrogen can influence the biological characteristics of breast cancer by SDF-1.
3.Influence of left-thoracic radiotherapy on heart function and quality of life in patients with breast cancer after breast-conserving surgery

Objective: To observe the influence of left-thoracic radiotherapy on heart function and quality of life in patients with breast cancer after breast-conserving surgery. Methods: A total of 35 patients with breast cancer, who recently received breast-conserving surgery and left-thoracic radiotherapy, were continuously selected as radiotherapy group. They received routine echocardiography (ECG), tissue Doppler imaging (TDI) and evaluation of Chinese questionnaire of quality of life in patients with cardiovascular diseases (CQQC) before and after radiotherapy. The results were compared with those of 31 patients (control group), who received radical mastectomy without breast radiotherapy in the same period. Results: There were no significant difference in left heart function indexes by ECG and TDI, all dimensional and total scores of CQQC between two groups before radiotherapy (P>0.05 all). After five-week treatment, compared with evaluation before radiotherapy and control group after treatment, there were significant decrease in E [(0.90±0.32) cm/s vs. (0.90±0.38) cm/s vs. (0.86±0.29) cm/s], E/A [(1.28±0.46) vs. (1.28±0.45) vs. (1.20±0.38)], Em [(10.12±1.93) cm/s vs. (9.95±1.64) cm/s vs. (7.51±1.12) cm/s], Em/Am [(1.25±0.26) vs. (1.24±0.23) vs. (0.90±0.15)] (P<0.05 all), all dimensional scores (P<0.05~<0.01)and total score [(67.36±7.87) scores vs. (63.75±7.26) scores vs. (52.83±6.40) scores] (P<0.01)of CQQC in radiotherapy group. Conclusion: There are often decrease in left heart function and quality of life in patients with breast cancer undergoing left-thoracic radiotherapy after breast-conserving surgery, so it’s necessary to perform targeted rehabilitative treatment.
4. Identification of Saigae Tataricae Cornu in Chinese materia medica preparation using DNA barcoding

Objective: To discuss the feasibility of using partial sequence of mitochondrial cytochrome C oxidase subunit I (COI) gene as DNA barcoding to identify Saigae Tataricae Cornu (STC) in Chinese materia medica (CMM) preparation. Methods: The DNA barcoding identification was constructed in STC samples from eight different manufactories. The genome DNA was extracted by DNA extraction kit from adequately ground samples. The COI nucleotide sequences were PCR amplified with the Primers LCO1490 and HC02198 or specified primer 0703 and sequenced bi-directionally. The BLAST comparison was made in GeneBank, and the phylogenetic trees were constructed with the homologisation tree (DNAMAN) and Neighbor-Joining (NJ, MEGA 5.2) method. Saiga tatarica (gb|JN632700.1) and STC were selected as reference medicinal materials and the genetic distance was calculated using Kimura-2-parameter. Results: The lengths of partial mitochondrial COI gene collected from STC in eight samples of CMM preparation were about 658 bp. The phylogenetic tree showed that the samples of CMM preparation and reference substance assembled distinctly, and were completely separated from the outgroup. The intraspecific genetic distances of these samples ranged in 0-0.064 with an average of 0.020, and the interspecific genetic distances ranged in 0.167-0.195. Conclusion: The mitochondrial COI gene is a valid DNA barcoding gene for STC identification in CMM preparation. But other organs in antelope also contain the same gene. In order to ensure the accuracy of the results, other texst methods also need to be used cooperatively.
5.Comparative Analysis of Limb Dysfunction in Different Surgical Breast Cancer

Objective To compare the incidence of limb dysfunction, sensory disturbance and lymphedema after different treatment methods in breast cancer patients, and evaluate the clinically relevant factors of limb dysfunction. Methods According to the different processing operation of axillary lymph nodes, 235 patients with breast cancer during 2005 to 2012 were divided into axillary lymph node dissection group in Mastoscopy (group A, n=120) and routine axillary lymph node dissection group (group B, n=115). The flexion, extension, abduction, adduction activity of the ipsilateral shoulder were compared with the contralateral, and the circumference of the up-and-down 15 cm of both upper limbs olecroanon were measured. The incidence of limb dysfunction of the patients was followed up in 6 months, 1 year and 2 years. Results There was no significant difference in limb activity and sensory disturbance between 2 groups 6 months, 1 year and 2
years after operation (P>0.05), the incidence of limb lymphedema was higher in group B than in group A (P<0.05). Radiotherapy was related with limb lymphedema in both groups (P<0.05). Conclusion Compared with the conventional axillary lymph node dissection, endoscopic axillary lymph node dissection can reduce the incidence of limb lymphedema after operation, but there was no significant difference in the limb activity and sensory disturbance between 2 groups. Postoperative radiotherapy is a risk to increase the occurrence of limb lymphedema.

8.Relationship of serum 25-hydroxyvitamin D with bone turnover biomarkers in pregnant women

[Summary] A total of 215 pregnant women at mid-gestation were enrolled, and their serum 25-hydroxyvitamin D levels were measured. Of them, 38 (17. 7%) were classified as cases with vitamin D sufficiency, 70 (32. 5%) as insufficiency, and 107 (49. 8%) as deficiency. Compared with the sufficient group, parathyroid hormone level increased and osteocalcin level decreased significantly in the deficient group. Therefore, vitamin D deficiency is highly prevalent among pregnant women and is associated with their potential disorder of bone turnover.
9.Influence of icaritin in proliferation and apoptosis of NB4 leukemia cells and its mechanism

of NB4 cells at S phase was lower than those in control group (P <0.05),and the number of NB4 cells at G1 phase was increased (P <0.05).The expression levels of Bcl-2 in ICA groups were significantly lower than that in control group (P < 0.05 ), while the expression levels of Bax were higher than that in control group (P < 0.05 ). Conclusion ICA can induce the apoptosis of NB4 cells via inhibiting the expression level of Bcl-2 and up-regulating the expression level of Bax.
10.Chemical constituents of Crotalaria ferruginea

Objective To study their chemical components of xianglingcao(Crotalaria ferruginea) and identify their chemical structures.Methods The compounds were isolated by chromatography and their structures were identified by spectral analysis and compared with the published data.Results Ten compounds were isolated and identified as vomifolilol(Ⅰ),genistein(Ⅱ),p-hydroxybenzoic acid(Ⅲ),5,7-dihydroxy-4-methoxy-flavone-7-O-?-D-glycopyranoside(Ⅳ),octadecoic acid(Ⅴ),octacosanol(Ⅵ),?-stitosterol(Ⅶ),?-daucosterol(Ⅷ),stigmasterol(Ⅸ),and ?5,22 stigmasterol-3-O-?-D-glycopyranoside(Ⅹ).Conclusion The ten compounds are obtained from xianglingcao for the first time.
