1. Surgical treatment of obstructive jaundice induced by biliary invasion in patients with gallbladder cancer: An analysis of outcome in 48 cases

Objective: To analyze the outcomes of surgical treatment of obstructive jaundice induced by biliary invasion in patients with gallbladder carcinoma. Methods: We retrospectively analyzed the clinical data of 48 patients with gallbladder cancer and biliary invasion-induced obstructive jaundice, who were treated in our hospital during January 2004 to December 2008. Results: Thirty-six patients who received surgical treatment had a median survival time of (17.39±3.98) months, and 12 patients received non-surgical treatment had a median survival time (3.75±0.51) months, with significant difference found between the two groups (P<0.01). Fifteen patients underwent radical resection, 7 underwent R1 resection, and 14 underwent R2 resection, with their median survival time being (30.93±7.42) months, (13.57±6.70) months, and (5.00±0.67) months, respectively; there were significant difference between the three groups (P<0.01). Conclusion: The prognosis of gallbladder cancer with obstructive jaundice is poor; surgical treatment can partly improve the prognosis of patients with obstructive jaundice-induced by gallbladder invasion. Radical curative resection, sometime with cholecystectomy, partial hepatectomy, or bile duct resection, should be performed for these patients.
2.Modification by wheat germ agglutinin delays the ocular elimination of liposome.

The purpose of this study is to explore the feasibility of wheat germ agglutinin (WGA) modified liposome as a vehicle for ophthalmic administration. Liposome loaded with 5-carboxyfluorescein (FAM) was prepared by lipid film hydration method. WGA was thiolated and then conjugated to the surface of the liposome via polyethylene glycol linker to constitute the WGA-modified and FAM-loaded liposome (WGA-LS/FAM). The amount of thiol groups on each WGA molecule was determined, and the bioactivity of WGA was estimated after it was modified to the surface of liposome. The physical and chemical features of the WGA-modified liposome were characterized and the ocular bioadhesive performance was evaluated in rats. The result showed that each thiolated WGA molecule was conjugated with 1.32 thiol groups. WGA-LS/FAM had a mean size of (97.40 +/- 1.39) nm, with a polydispersity index of 0.23 +/- 0.01. The entrapment efficacy of FAM was about (2.95 +/- 0.21)%, and only 4% of FAM leaked out of the liposome in 24 h. Erythrocyte agglutination test indicated that after modification WGA preserved the binding activity to glycoprotein. The in vivo ocular elimination of WGA-LS/FAM fitted first-order kinetics, and the elimination rate was significantly slower than that of the unmodified liposome, demonstrating WGA-modified liposome is bioadhesive and suitable for ophthalmic administration.

3.Selenium dioxide inducing apoptosis of cervical cancer cells by regulating apoptosis-related proteins

Objective To investigate the inducing effects of selenium dioxide(SeO2 ) on the apoptosis in human cervical carcino-ma cell line Hela and its influence on the expression of apoptosis-related proteins caspase-3 and P53 .Methods Hela cells were trea-ted with different concentrations of SeO2 for 24 h in vitro ;the morphological changes of Hela cells were observed by the optical mi-croscope;the influence of SeO2 on the cell proliferation and vitality was examined by the MTT assay ;the flow cytometry was em-ployed to detect the cell apoptosis rate ;the expressions of caspase-3 and P53 proteins in Hela cells were determined by the Western blot analysis .Results Under the optical microscopy ,SeO2 generated the obvious influence on the cell growth morphology ,a large number of cells became rounded and shrunken ,and lost the normal form ,while the adherence cell number was evidently decreased and the proliferation was slowed down ;the MTT results showed that SeO2 markedly inhibited the cell proliferation and viability in a dose-dependent manner ,in which ,the cell apoptosis rates induced by the 0 ,1 .875 ,3 .750 ,7 .500 ,15 .000 and 30 .000 μmol/L con-centrations of SeO2 were 3 .12% ,30 .56% ,33 .42% ,37 .50% ,45 .43% and 69 .38% respectively ,which revealing the obviously in-creasing trend;the Western blot assay revealed that SeO2 could up-regulate the caspase-3 and P53 levels ,and reached the peak value at the concentration of 7 .500μmol/L .Conclusion SeO2 could induce the cervical cancer cell apoptosis possibly by up-regulating the expressions of caspase-3 and p53 in Hela cells .

5.Correlation Study on FTO Expression and Insulin Secretion Level in Pregnant Women

Objective To evaluate the correlation of maternal FTO expression and insulin secretion levels in pregnant women.Methods In outpatient of Zhuhai Municipal Maternal and Child Healthcare Hospital,60 cases of pregnant women with gestational diabetes mellitus (GDM) and 60 cases of healthy pregnant women were recruited in this study from March 2016 to August 2016.Each pregnant woman was taken two tubes of venous blood.Lymphocytes was isolated from one tube of anticoagulated whole blood.The expression of FTO mRNA in lymphocytes was investigated using RT-PCR.Serum was isolated from another tube of non-anticoagulant whole blood.Its FTO protein was investigated using ELISA,insulin was investigated using chemiluminescence method,and GLU,CHO,TG,HDL-C and LDL-C were investigated by automatic biochemical analyzer.The correlation of the biochemical indicators were analyzed.Results The expression level of FTO mRNA,FTO protein,fasting glucose data,OGTT at 1 h and 2h glucose levels in the case group were 0.17 % ± 0.10 %,61.32 ± 25.23 pg/ml,4.71 ±0.54 mmol/L,10.70±1.36 mmol/L and 8.97 ± 1.60 mmol/L respectively.The values of these biochemical indicators in control group were 0.11%±0.07%,51.47±22.97 pg/ml,4.41± 0.28 mmol/L,8.05±1.04 mmol/L and 6.56±0.75 mmol/L respectively.The differences were statistically significant (t =3.876,2.236,3.817,11.964 and 10.578,P<0.05).The insulin levels were 6.83±9.76 mU/L in the case group,and 13.15±13.99 mU/L in control group.The differ ences was statistically significant too(t=-2.869,P<0.05).FTO expression level was positively correlated with FTO protein,OGTT 1h glucose and OGTT 2h glucose levels (r=0.232,0.292,0.242,all P<0.05),and it was negatively correlated with insulin levels(r=-0.185,P<0.05).There was no correlation between FTO and fasting glucose expression levels (P >0.05).Conclusion The expression of FTO level in pregnant women with GDM was higher than that in healthy pregnant women.The relative expression of FTO mRNA was negatively correlated with insulin.And the regulation of glucose metabolism might be effected by insulin.

7.Effect of Helicobacter pylori lipopolysaccharide on expression of Gli and Ptch-1 proteins in sonic hedgehog signaling pathway of gastric mucosa GES-1 cells.

OBJECTIVETo investigate the effect of Helicobacter Pylori lipopolysaccharide (Hp-LPS) on expression of Gli and Ptch-1 proteins in sonic hedgehog (Shh) signaling pathway of gastric mucosa GES-1 cells.
METHODSThe LPS was extracted from Hp by hot phenol water method, and then the concentration of LPS was detected by the kinetic turbidimetric assay. GES-1 cells were stimulated by different concentrations of Hp-LPS (0, 1, 10, 20, 30 and 40 μg/ml). The inhibition rates of cell growth were measured by MTT assay after treated with Hp-LPS for 24 h. The expression of Gli and Ptch-1 proteins were determined by Western Blot.
RESULTSMTT assay showed that the inhibition rates of GES-1 cell growth after treatment by different concentrations of Hp-LPS (1, 10, 20, 30 and 40μg/ml) were 25.8% ± 2.7%, 34.2% ± 3.1 %, 46.3% 3.4%, 60.8% ± 2.1% and 82.9% ± 2.8% respectively (r=0.985, P<0.001). Western blot showed that the expressions of Gli and Ptch-1 proteins were decreased after Hp-LPS treatment (0, 1, 10, 20, 30 and 40 μg/ml): the relative expression values of Gli were 1.286 ± 0.180, 0.963 ± 0.067, 0.850 ± 0.085, 0.566 ± 0.058, 0.549 ± 0.056 and 0.377 ± 0.047, respectively (r=-0.945, P<0.001); those of Ptch-1 were 1.688 ± 0.088, 1.466 ± 0.061, 1.170 ± 0.065, 1.042 ± 0.064, 0.648 ± 0.057 and 0.482 ± 0.074, respectively (r=-0.985, P<0.001).
CONCLUSIONHp-LPS can decrease the related protein expression of Shh signaling pathway, which indicates that Hp may interfere with the function of Shh signaling pathway in gastric mucosa via the effect of its LPS.

8.Changes in distribution and antimicrobial resistance of pathogens causing bloodstream infection in 2012-2015

Objective To investigate the changes in distribution and antimicrobial resistance of pathogens causing bloodstream infection in a hospital in 2012-2015.Methods Pathogenic bacteria isolated from blood culture in a hospital microbiology laboratory between January 2012 and December 2015 were collected and divided into 2012-2013 group and 2014-2015 group,distribution characteristics,constitute,and antimicrobial susceptibility of two groups were compared.Results Gram-negative bacteria were the main pathogens isolated during two periods,ac-counting for 54.96% and 54.66% respectively,there was no significant differences in pathogen distribution between two groups (P >0.05).Gram-positive cocci had a high sensitivity to vancomycin and linezolid,resistant rates were both 0;resistance rates of coagulase negative staphylococci to oxacillin were all >80%,resistance to penicillin was also >90%;Enterobacteriaceae was highly sensitive to imipenem and meropenem,but resistance strains had ap-peared;Acinetobacter baumannii was sensitive to minocycline during two periods,resistance rates were 35.90% and 34.55% respectively,resistance rates to other antimicrobial agents were also high (>75%).Conclusion The iso-lation rate of drug-resistant pathogenic strains causing bloodstream infection is high,monitoring on bacterial resistance is helpful for guiding rational use of antimicrobial agents in clinic.
9.Protective effect of Danxuetong injection against testicular injury after testis torsion/detorsion in rats.

OBJECTIVETo investigate the protective effect of Danxuetong injection (DXT, a combination of Danshen and Xueshuantong injections) against testicular ischemia-reperfusion injury following testis torsion/detorsion in rats.
METHODSThirty-two 4-week-old healthy male SD rats were randomly divided into four groups of equal number: sham operation, normal saline, single DXT injection, and successive DXT injection. The rat models of testicular ischemia-reperfusion injury were established by 2-hour 720-degree torsion/detorsion of the unilateral testis. At 6 weeks after modeling, the rats were killed and their testes were harvested for measure- ment of testicular coefficients, sperm counts, sperm motility, and the levels of total anti-oxidative capacity (T-AOC) , superoxide dismutase (SOD) , nitric oxide synthase (NOS) , and malondialdehyde ( MDA) in the testis tissue.
RESULTSCompared with the rats of the normal saline group, those of the single DXT injection and successive DXT injection groups showed significant increases in the testicular coefficient (0.11 ± 0.03 vs 0.35 ± 0.04 and 0.40 ± 0.06, P < 0.05), sperm count ([0.46 ± 0.10] vs [1.44 ± 0.50] and [3.00 ± 1.28] x10(9)/ml, P < 0.05), sperm motility ([13.63 ± 14.04] vs [39.63 ± 5.04] and [76.31 ± 3.67]%, P < 0.05), the activity of SOD (72.76 ± 5.58 vs 116.25 ± 8.83 and 133.20 ± 13.84, P < 0.05), and the level of T-AOC (5.58 ± 1.07 vs 13.34 ± 5.81 and 19.21 ± 5.69, P < 0.05), but a remarkable decrease in the content of MDA (42.38 ± 8.94 vs 20.94 ± 5.65 and 15.02 ± 1.03, P < 0. 05) in the injured testes.
CONCLUSIONDXT can effectively rid the testis tissue of oxygen free radicals, improve sperm count and motility by antioxidation, and protect the testis tissue of prepubertal rats against testicular ischemia-reperfusion injury after testis torsion/detorsion. It also has a protective effect on the contralateral testis, and successive injection has a better effect than single injection of DXT.

10.Study on vector-specific and foreign gene-speciflc immune responses induced by rAd5 and rAAV2/1 vaccines

Objective To compare the foreign gene-specific and vector-specific immune responses in BALB/c mice immunized with rAd5 or rAAV2/1 expressing the same gene.Methods BALB/c mice were immunized with rAd5-gag or rAAV2/1-gag once,HIV-1 Gag-specific and vector- specific cellular immune responses were analyzed by Elispot assay,HIV-1 P24-specifc IgG and vector-specific IgG were tested by ELISA assay.Results Mice immunized with rAd5-gag induced potent Gag-specific cellular immune responses and that were significantly higher than Ad5-specfic cellular responses,while rAAV2/1-gag elicited weak Gag-specific and AAV2/1-specific cellular responses.Both P24-specific and Ad5-specific IgG titers induced by rAd5-gag were high and in similar level.Higher level of P24-specific IgG was found in mice inoculated with rAAV2/1-gag than rAd5-gag.And the P24-specific IgG titers were higher than the vectorspecific IgG titers in mice immunized with rAAV2/1.Conclusion rAd5 could elicit strong foreign genespecific cellular and humoral immune responses,weak vector-specific cellular responses and strong vectorspecific antibodies,rAAV2/1 could induce potent foreign gene-specific antibodies that were much higher than vector-specific IgG,while both foreign gene-specific and vector-specific cellular responses were very low.
