Objective To explore a simple, effective and stable method for the isolation and purification of Kupffer cells from rat liver, enabling further study on the structure and function of these cells in vitro. Methods After laparotomy, a catheter was inserted into the portal vein and secured with artery clamp. Then, the rat liver was perfused and digested with solution Ⅰ and solution Ⅱ containing 0.05% collagenase Ⅳ respectively. The cell suspension was centrifuged with isopycnic sedimentation in a two-step Percoll gradient to harvest Kupffer cells. The isolated Kupffer cells were purified by selective adherence after 30 min of cultivation, and identified by evaluation of phagocytosis of India ink and peroxidase staining with DAB through light and electron microscopy. Results It was verified that the viability of isolated Kupfffer cells was more than 90% through Trypan blue staining. Those Kupffer cells could attach to plastic quickly and phagocytose ink, and had the appearance of "fried eggs" in positive peroxidase staining with a purity of 95%. Under the light microscopy, the appearance of newly isolated Kupffer cells was round with uniform shape and size. After two days of culture, Kupffer cells appeared to distend with irregular or stellate shape. The typical features were observed in the transmission electron micrographs. There were numerous pseudopods and occasional cup-like indentations in the cell membrane of Kupffer cells. The cytoplasm contained numerous types of lysosomes and other phagocytotic vesicles. Conclusion The method for isolating and culturing Kupffer cells in this study is effective and stable, and the biological characters are preserved in the cultured cells.

Objective To compare and analyze the clinical effects and reliabilities of intermandibular flexible distraction and rigid internal fixation in treating mandibular condylar fractures. Methods Of 40 patients with mandibular condylar fractures, 21 cases were managed with intermandibular flexible distraction (external fixation group, Group A) and 19 with rigid titanium internal fixation (internal fixation group, Group B). There were 15 cases with high position fractures and 25 with low position ones. Results During 2-4 weeks postoperatively, X-rays confirmed the condylar fracture healing in all cases with no loosened mini-tinum plates in the Group B. There were 10 cases in the Group A and 14 cases in the Group B with the opening width over 30 mm, and 11 cases in the Group A and 5 in the Group B with the opening width less than 30 mm. Conclusions Both external and internal fixations can attain condylar fracture healing. However, compared with the internal fixation, the reduction of external fixation is less exact and the function recovery slower after operation, especially in the low position fractures. No irreversible damages on TMJ happen in all cases.

Objective To evaluate the clinical efficacy of acupuncture plus auricular point electroacupuncture and its effect on the quality of life (QOL).Method Ninety-eight patients with post-stroke depression were randomized into a treatment group and a control group, 49 cases in each group. The treatment group was intervened by acupuncture plus auricular point electroacupuncture, while the control group was by oral administration of Fluoxetine hydrochloride. The Hamilton Depression Scale (HAMD), Side-effect Rating Scale (SERS), and the World Health Organization Quality of Life Questionnaire-Brief Version (WHOQOL-BREF) were observed before and after intervention, and the clinical efficacies were also compared.Result The total effective rate was 89.1% in the treatment group versus 87.0% in the control group, and the between-group difference was statistically insignificant (P>0.05). Respectively after 2-week, 4-week, and 8-week treatment, the HAMD scores were significantly different from that before intervention in the treatment group (P<0.05). Respectively after 2-week, 4-week, and 8-week treatment, the HAMD scores were significantly different from that before intervention in the control group (P<0.05). There were no significant differences in comparing the HAMD scores between the two groups after 2-week, 4-week, and 8-week treatment (P>0.05). The SERS scores in the treatment group were significantly different from that in the control group after 4-week and 8-week treatment (P<0.05). In the treatment group, the SERS score after 8-week treatment was significantly different from that after 4-week treatment (P<0.05). After treatment, the scores of general health domain 1 and 4, physical health domain, psychological health domain, and environment domain from WHOQOL-BREF were significantly changed in both groups (P<0.05). After intervention, the physical health domain score of the treatment group was significantly different from that of the control group (P<0.05).Conclusion Acupuncture plus auricular point electroacupuncture is an effective approach in treating post-stroke depression, and it can improve the quality of life of the patients.

Objective To study the effect of preoperative selective arterial infusion chemotherapy (PSAIC) on large intestine cancer. Methods 63 patients with colorectal cancer underwent PSAIC with 5 fluorouracil,mitomycin and E adriamycin; the changes of clinical manifestation and pathology were observed and analyzed. Results (1)clinical manifestation:abdominalgia relieved in 18 patients and abdominal distention relieved in 16 patients.Improvement of hematochezia was found in 7 patients.In addition, of 13 patients with partial ileus, the clinical symptoms relieved in 9 patients. (2) pathology: there were karyopyknosis,karyorrhexis,coagulation and necrosis of cytoplasm in cancer cells. infiltration of inflammatory cells, edema and fibroelastosis in mesenchyne of cancer tissue. Proliferous intima and thrombus were also observed in the vessels. Most of these changes were moderate,and marked changes could also be seen in necrosis of cytoplasm and vessel intima proliferation. Conclusion PSAIC can control the focus of primary disease and the micrometastatic foci as well as improve the clinical symptoms, such as intestinal obstruction, and hematochezia, so PASIC is helpful to subsequent operation,and can improve the survival rate of patients with colorectal cancer.

ObjectiveTo research in the essence of the caring experience of caregiver for children with eye enucleation, to find out the caring experience and caregiver's needs, to give them physical and mental nursing in order to make them cooperate with us and implement effective treatment to children.Methods12 caregivers for children with eye enucleation were given semi-structured interviews. The materials were analyzed, putted in order, and themes were got.ResultsThrough deep interview of 12 caregivers, five titles were summarized:overburdened psychology, limited schedule, requirement of health information, overload economic burden, to return and rebuild.ConclusionsThe results could help nurses to give more correct health education and psychological care to caregivers. Then caregivers can cooperate with nurses to give effective treatment to children, thus life quality of children with eye enucleation will be improved a lot.

Objective To investigate the characteristic and rehabilitation of polyglot aphasia.Methods A case whose native languages are both Chinese and Korean,and can also speak fluent Japanese,suffering aphasia after a traffic accident was reported.Results and Conclusion His communication ability mainly on understanding and expression improved after a few months rehabilitation.The rehabilitation in different languages improved on different level.

Objective To summarize the clinical features,treatment and prognosis of bullous systemic lupus erythematosus (BSLE).Methods Clinical data were collected from 6 cases of BSLE between May 2009 and September 2015,and were analyzed retrospectively.Results Among the 6 patients,2 were male and 4 were female.The mean age of onset was 34 years.All the 6 patients presented with tense blisters and bullae arising on an erythematous base or normal skin,which were arranged in an annular pattern in 3 patients.The SLE Disease Activity Index (SLEDAI) score was ＞ 4 in all the 6 patients.Histopathological examination showed subepidermal blisters or fissures in all the patients.Direct immunofluorescence (DIF) revealed continuous linear deposition of immunoglobulin G (IgG),IgM and IgA in all the patients,linear deposition of C3 in 4 patients,and linear deposition of Clq in 2 patients at the basement membrane zone (BMZ).All the 6 patients were treated with oral glucocorticoids and hydroxychloroquine,and 2 patients were additionally treated with cyclophosphamide.During the follow-up period,adverse reactions to different extents were observed in the 6 patients.Conclusions BSLE mainly occurs in young and middle-aged people.Histopathologically,subepidermal blisters or fissures can be observed with linear deposition of IgG,IgM,IgA,or C3 along the basement membrane zone on DIF.

Objective To investigate the association between HLA-DRB1 alleles and pemphigus vulgaris (PV) in a Han population in Sichuan.Methods Polymerase chain reaction with sequencespecific primer (PCR-SSP) method was used for low-resolution and high-resolution typing of HLA-DRB1 alleles in 19 patients of Han nationality with PV and 25 healthy controls in Sichuan.Allele frequencies were calculated,and differences in the allele frequency between the above two groups were compared by using chisquare test.Results Totally,9 kinds of DRB 1 low-resolution alleles and 19 kinds of DRB 1 high-resolution alleles were identified in the PV patients and healthy controls.Frequencies of the DRB1* 14 allele (39.47％[15/38] vs.8.00％[4/50],x2 =17.43,P ＜ 0.05) and DRB1*1405 allele (15.79％[6/38] vs.2.00％[1/50],x2 =4.25,P ＜ 0.05) were significantly higher in the PV patients than in the healthy controls.Conclusion The HLA-DRB1*14 allele may be a common susceptibility gene for PV in the Han population in Sichuan,and the HLA-DRB1* 1405 allele may be most closely associated with PV.

Objective To evaluate the value of low concentration of isotonic contrast agent combined with 80 kV and adaptive statistical itera-tive reconstruction (ASiR)technique for bronchial artery with CT angiography (CTA).Methods 40 patients (18.5 kg/m2 ≤ body mass index(BMI)≤25 kg/m2 )with lung cancer were randomly divided into two groups and performed enhanced chest CT examina-tion.20 patients (low concentration group)were scanned with Visipaque 270mg I/mL,scanning protocol of (80 kV,260 mA)and other 20 patients (high concentration group)were scanned with Ultravist 370 mg I/mL,scanning protocol of (120 kV,1 50 mA). The CT value,signal noise (SD),signal-to-noise ratio (SNR),contrast-to-noise ratio (CNR)and effective dose (ED)of the two groups were compared and analyzed statistically.Differences between the two groups were determined with t test(P <0.05).Results The mean CT value of the low concentration group (31 7.33 HU±70.44 HU)was higher than that of the high concentration group (256.39 HU±37.38 HU)(P <0.05).SD,ED and dose length product (DLP)of the low concentration group (SD:7.55 ±1.51 HU;ED:1.74±0.14 mSv;DLP:124.94±9.9 mGy/cm)were lower than those of the high concentration group (SD:9.36±1.26 HU;ED:3.20±0.35 mSv;DLP:228.82±25.13 mGy/cm)(P <0.05).CNR and SNR of the low concentration group (CNR:59.71± 17.36;SNR:44.01±14.71)were higher than those of the high concentration group (CNR:37.61±6.16;SNR:27.75±4.72)(P <0.05).The subjective scores of the two groups were (3.73±0.66)and (3.81±0.59),respectively,with no statistical difference.Conclusion For the patients with normal body mass index ,it can reduce 47% radiation dose and 27% iodine dosage using low concentration of isotonic contrast agent combined with 80kV and ASiR technique for bronchial artery CTA,while maintaining similar excellent image quality.
