1.Experimental investigation of the pathological changes in subchondral cocortical bone in steroid-induced osteonecrosis

[Objective]To investigate the morphological changes that take place in the subchondral cocortical bone in steroid-induced osteonecrosis and analyze the reasons leading to humeral head collapse in juvenile rabbits.[Method]Five-six month-old female rabbits were separated by two groups.A modified version of the methods was used to replicate steroid enhanced osteonecrosis anminal humeral head models with Shwartzman reaction in group A,and group B served as the single control.Each humeral head was obtained 10 weeks after the drugs injection.Subchondral cortical bone was observed,and the number of haversian canals was counted.The microcirculatory changes were also detected with scanning electron microscope.[Result]In group A,the Haversian canals in' subchondral area almost disappeared;the subchondral cortical bone disappeared with its arch,dome and bridge structures.Microcirculatory stasis happened in the subchondral vessels.Some humeral heads collapse were observed.While in group B,subchondral cortical bone is integrity and continuity,forming arch,dome and brige structures with subtrabecular bone.[Conclusion]The disappearance of the subchondral cortical bone is a major reason leading humeral head collapse,and ischemia is the critical reason of it.
2.Effects of Simvastatin on the proliferation and osteoblastic differentiation of human periodontal ligament cells

Objective: To study the effects of Simvastatin on the proliferation and osteoblastic differentiation of human periodontal ligament (PDL) cells. Methods: The fourth passage human PDLs were cultured in conditioned mineralization medium with different concentrations of Simvastatin. Proliferation ability was detected by MTT method. Alkaline phosphatase (ALP) activity was measured with 4-nitrophenyl phosphate hexahydrate (PNPP) assay. Results: Proliferation and osteoblastic differentiation of human PDLs were increased in all treatment groups with different concentrations of Simvastatin(10-9, 10-8, 10-7, 10-6 mol/L). Compared with control group, significant statistical difference was found in 10-8 mol/L, 10-7 mol/L and 10-6 mol/L groups(P<0.05). The maximum effect was observed at the concentration of 10-7 mol/L. Conclusion: Optimal concentration of Simvastatin can improve the proliferation and osteoblastic differentiation of human PDLs.
3.Human papillomavirus genotypes in male patients attending the STD clinic in Zhenjiang area.

OBJECTIVETo investigate the status of human papillomavirus ( HPV) infection and its genotypes in male patients in Zhenjiang area.
METHODSUsing PCR and reverse dot blot hybridization, we determined the genotypes of HPV DNA in 245 male patients at our Clinic of Dermatology and STD.
RESULTSThe total rate of HPV infection was 43.67% (107/245), and 18 subtypes were detected. Among the 107 HPV-positive cases, low-risk, high-risk, and combined high- and low-risk infections accounted for 39.25% (42/107), 38.32% (41/107), and 22.43% (24/107), respectively. The most notable low-risk HPV types were HPV6 and HPV11, and the most notable high-risk HPV types were HPV16, HPV52, and HPV58. The rates of single infection and multi-infection were 53.27% (57/107) and 46.73% (50/107), respectively. One case had the most types, infected with 8 genotypes. No statistically significant differences were observed in the total rate of HPV infection among different age groups (Χ2 = 7.999, P > 0.05).
CONCLUSIONThe dominant subtypes of HPV infection in male patients in Zhenjiang area were HPV6, HPV11, and HPV16. The most common subtypes were HPV6 and HPV11 in low-risk infection, and HPV16, HPV52, and HPV58 in high-risk infection.

4.A Semi-quantitative Serological Method to Assess the Potency of Inactivated Rabies Vaccine for Veterinary Use

Potency is one of the most important indexes of inactivated vaccines.A number of methods have been established to assay the potency,of which the NIH test and single-dose mouse protection test are the prescribed methods.Here,we report a method to semi-quantitatively assay the potency of an inactivated rabies vaccine,which uses fewer animals and takes less time to complete.Depending on the quality requirements of a vaccine(e.g.minimum potency),a rabies reference vaccine is,for example,diluted to the minimum potency,and 50 μL of the dilution is taken to inoculate 10 mice.The same amount of the test rabies vaccine is inoculated into another 10 mice.After two weeks,all mice are bled and serum samples are assayed for viral neutralizing antibody by the fluorescent antibody virus neutralization(FAVN) test.By comparing the median and interquartile range of antibody titers of the reference vaccine with those of the test vaccine,the test vaccine potency can be semi-quantitatively judged as to whether it is in accord with the required quality.The reliability of this method was also confirmed in dogs.The procedure can be recommended for batch potency testing during inactivated rabies vaccine production.
5.Bone marrow mesenchymal stem cells inhibit inflammatory response of microglial cells

Objective To investigate the inhibitory effect of bone marrow mesenchymal stem cells ( BMMSC) on the inflammatory response of microglial cells .Methods The samples were divided into four groups .Group I:microglial (BV-2) cells were cultured in DMEM (high glucose).Group II: BV-2 cells were cultured in DMEM containing lipopo-lysaccharide (LPS).Group III:BV-2cells and BMMSCs were co-cultured in DMEM.Group IV: BMMSCs were cultured in DMEM containing LPS .The growth state and ultrastructure of BV-2 cells were observed and the changes of TNF-αex-pression were detected .Results Different cell densities of BV-2 cells were observed under the optical microscope in an or-der from high level to low level:group I >group III >group II.The expressions of TNF-αwere:group Ⅱ >groupⅢ>group Ⅰ.Ultrastructural observation of BV-2 cells showed that there were a large number of swollen mitochondria and endoplasmic reticulum , some of them showed vacuolization .No BV-2 cells with multiple hucleoli were found in the group II indicating the absence of active cell growth .At the same time, cytolysis was observed only in the group II .The growth of BV-2 cells in the group III was better than that in the group II .Conclusions BMMSCs can inhibit inflammatory response of microglial cells, therefore, play a neuroprotective role.
6.Effect of IGF-1 and bFGF on differentiation of human dental pulp stem cells in vitro

Objective To investigate the effect of insulin-like growth factor-1(IGF-1) and basic fibroblast growth factor (bFGF) on the differentiation of human dental pulp stem cells in vitro. Methods Mono-cell suspension was separated from the human adult dental pulp with collagenase Ⅰ and dispase digestion. Colony-forming efficiency of cells was calculated. Cells were observed under an invert microscope. Dentin sialoprotein (DSP), dentin matrix protein 1 (DMP-1) and dentin sialophosphoprotein (DSPP) mRNA of the 3rd passage cells induced with IGF-1 and bFGF in vitro were detected by immunohistochemistry and RT-PCR. Results Clonogenic cells were obtained from dental pulp and colony-forming efficiency of cells was 2.6-3.5 colonies/10 4 cells. Human specific DSP, DMP-1 protein and DSPP mRNA expressed in colony-forming cells after 7 d of induction with IGF-1 and bFGF in vitro. Conclusion Adult stem cells might exist in human dental pulp. Dental pulp stem cells could differentiate into odontoblast under the induction of IGF-1 and bFGF.
7.Correlative Study of Scan Beginning Time with Ejection Fraction of Heart in Angiography of Brain by Spiral CT and Its Clinical Application

Objective To find up a suitable beginning scan time so that to assure the image quantity during CTA of brain.Methods 30volunteers were checked by heart colour sonography first,then CTA at earlier stage was done,when the density in basilar artery high up to120 HU,that time was regarded the time as beginning scan time,the relative curve between beginning scan time and ejection fraction(EF) was drew,according to this relative curve,34 patients were examined by CTA,the displaying situation of vessel was evaluated comparatively with DSA in part of patients.Results According to this method,the successful rate of CTA in patient’s group was 100%,the trunk of brain vessel and its 1~3 level branch were showed in 100%,the 4~5 level branch of middle cerebal artery showed in 94.1%(32/34).Conclusion According to EF of heart to determine the beginning scan time of CTA is practical,that the image quantity of CTA can be assured.
8.Animal Host of Schistosoma japonicum and Transmission of Schistosomiasis in Poyang Lake Region

The article clarified, according to the recent research results which have been done in Poyang Lake region of Jiangxi province, bovines are the most important animal hosts of Schistosoma japonicum and infected bovines are main infectious sources of Schistosomiasis in Poyang Lake region. Spring (March to June) is the " cross transmission stage" between definitive host and intermediate host, that is, spring is not only the susceptible season of infection for livestock and human, but also is the susceptible season of infection for snails. In flood season (July to August), the transmission of Schistosomiasis between livestock and snails belong to " low level phase" . Autumn (September to Octber) is the second seasonal peak of cercariae and is the susceptible season of infection for livestock and human. Winter(November to Feburary in next year) is the non- susceptible season of infection. Most of the susceptible zones of infection for livestock, human and snails are the marshlands near the endemic villages.
