Objective:To study the effect of retinoic acid (RA) and basic fibroblast growth factor (bFGF) on neural stem cells’(NSCs) proliferation and differentiation from new born rats’ pallium. Method: NSCs were isolated from the brains of new born Sprague-Dawley rats’ pallium, and the features of cells were characterized by immunofluorescence staining. The effects of different culture medium on survival and proliferation of cells were determined by MTT assay. The effects of bFGF and RA on differentiation of NSCs were observed. Results: MTT assay indicated the proliferation of cells in bFGF group, bFGF+RA group and RA group was continually increased, highest in bFGF group. The percentage of neurons differentiated from NSCs in RA group was 2 or 3 times that in bFGF group and control group . Conclusion: bFGF is important to the proliferation and long-term living of NSCs. It can prohibit the differentiation of NSCs. RA can counteract the effects of bFGF and also promote NSCs to differentiate into neurons in vitro.

Objective To assess the therapeutic effect of radioactive seed 125I brachytherapy combined with GP chemotherapy regimen (gemcitabine 1000 mg/m2, cisplatin 75 mg/m2) for inoperabale stage Ⅲ non-small cell lung cancer (NSCLC). Methods Thirty-nine documented inoperable stage Ⅲ NSCLC patients, enrolled between January 2005 and June 2008 for the study group, were treated with the combination of 125I brachytherapy and GP regimen. The brachytherapy methods were conducted according to TPS and each patient was treated under those patients were treated with standard GP regimen. Chest CT scans were performed every three months post-procedurally, until disease progression or recurrence. The follow-up time was up to twenty four months after treatment. In the control group, equal amount of Ⅲ stage NSCLC patients were treated with standard GP regimen alone. Chi-square test and survival analysis with Kaplan-Meier and Log-rank were used to compare the differences of recent (3 months after therapy)efficiency, survival rate, survival time between two groups. Results The re-cent effective rates of the study group (71.8%, 28/39) and control group (61.5%, 24/39) were not statistical different (χ2=0.93, P>0.05), yet the tumor CR rates in two groups showed significant disparity (χ2=4.48, P<0.05). The one-year survival rates of the study group and the control were 79.5% (31/39), 66.7%(26/39) respectively, with no significant difference (χ2=1.57, P>0.05). However, singificant differences (χ2=4.07, 4.63,both P<0.05) were found in 2-year survival rate and median survival time, with 41.0% (16/39) vs 23.1% (9/39) and 18.9±2.7 months vs 14.2±0.7 months. Conclusions 125I brachytherapy combined with GP regimen chemotherapy could be an effective treatment method and could improve the tumor CR rate and survival rate for patients with inoperable stage Ⅲ NSCLC.

A series of [1,3]dioxolo[4,5-f]isoindolone derivatives were designed, synthesized and evaluated as inhibitors of acetylcholinesterases (AChE). Furthermore, their effects on memory impairment of mice induced by scopolamine were investigated with step-through test. The results suggested that most of the target compounds exhibited potential inhibition on AChE with IC50 values at micromolar range. Compounds I1 (IC50 value of 0.086 μmol · L(-1)) and I2 (IC50 value of 0.080 μmol · L(-1)) showed the strongest AChE inhibitory activity, which are equipotent to donepezil (IC50 value of 0.094 μmol · L(-1)). Moreover, compounds I1-I4 could improve the memory impairment induced by scopolamine in mice.

10.Alpha-synuclein interacted proteins: the relevance with the pathogenesis of Parkinson's disease.

Journal of Zhejiang University. Medical sciences 2008;37(5):524-530
Parkinson's disease (PD) is characterized pathologically by the relatively preferential loss of dopaminergic neurons with resultant depletion of striatal dopamine and presence of Lewy bodies mainly composed by alpha-synuclein (alpha-SYN) in the remaining neurons in the substantia nigra. A lot of evidence suggests that the aggregation of alpha-SYN play an essential role in the pathogenesis of PD and formation of Lewy body. Increasing findings have implicated that some proteins, including parkin, synphilin-1,14-3-3, agrin and tau, interact with alpha-SYN and are involved in the abnormal aggregation of alpha-SYN.
