1.Study Advances in Anti-diarrhea with Single Chinese Herb and Its Effective Components

Objective: To introduce recent research progress in anti-diarrhea with single Chinese herb and its effective compo-nents. Methods:The references on anti-diarrhea with single Chinese herb and its effective components in recent years were reviewed and analyzed. Results:The researches on anti-diarrhea activity and possible mechanism of single Chinese herbs, such as Terminalia chebula Retz, Fructus mume, Radix Puerariae and Pulsatilla chinensis ( Bunge) Regel, were reviewed. Conclusion: At present, the researches on anti-diarrhoea with single Chinese herb and its effective components have made some progress, while further and wider researches are still needed to explore the underlying mechanisms.
2.Early postnatal overnutrition results in insulin resistance in adulthood

Overnutrition during the early postnatal life,a critical time window for growth and development,may induce metabolic syndrome later in life,including overweight/obesity and insulin resistance.The important target organs of insulin,such as liver,adipose tissue,skeletal muscle,and central nervous systems show insulin resistance.The involved mechanisms include abnormality of insulin signal pathway,increment of free fatty acid and some adipocytokines,oxidative stress,maladjustment of orexigenic and anorexigenic neuron,modifications of the hypothalamic-pituitary-adrenal glucocorticoid axis as well as epigenetic,etc.Hence,overnutrition should be avoided during the early postnatal life,so as to decrease the risk of developing long-term insulin resistance.
3.Isolation and Purification of Kupffer Cells from Rat Liver

Objective To explore a simple, effective and stable method for the isolation and purification of Kupffer cells from rat liver, enabling further study on the structure and function of these cells in vitro. Methods After laparotomy, a catheter was inserted into the portal vein and secured with artery clamp. Then, the rat liver was perfused and digested with solution Ⅰ and solution Ⅱ containing 0.05% collagenase Ⅳ respectively. The cell suspension was centrifuged with isopycnic sedimentation in a two-step Percoll gradient to harvest Kupffer cells. The isolated Kupffer cells were purified by selective adherence after 30 min of cultivation, and identified by evaluation of phagocytosis of India ink and peroxidase staining with DAB through light and electron microscopy. Results It was verified that the viability of isolated Kupfffer cells was more than 90% through Trypan blue staining. Those Kupffer cells could attach to plastic quickly and phagocytose ink, and had the appearance of "fried eggs" in positive peroxidase staining with a purity of 95%. Under the light microscopy, the appearance of newly isolated Kupffer cells was round with uniform shape and size. After two days of culture, Kupffer cells appeared to distend with irregular or stellate shape. The typical features were observed in the transmission electron micrographs. There were numerous pseudopods and occasional cup-like indentations in the cell membrane of Kupffer cells. The cytoplasm contained numerous types of lysosomes and other phagocytotic vesicles. Conclusion The method for isolating and culturing Kupffer cells in this study is effective and stable, and the biological characters are preserved in the cultured cells.

5.Analysis of serum uric acid level in pregnant women with pregnancy induced hypertension

Objective To investigate the relationship between serum uric acid level and the severity of pregnancy induced hypertension (PIH) in pregnant women with hypertension.MethodsFrom January 2016 to July 2016 in our hospital for treatment of 100 cases of patients with pregnancy induced hypertension, as PIH group, divided into severe hypertension group (33 cases), moderate PIH group (33 cases) and mild PIH group (34 cases), then select 100 cases of normal pregnant women normal pregnancy group.The serum uric acid levels were measured and the results were analyzed.ResultsThe average serum uric acid levels in PIH group (359.75+89.23) mol/L was significantly higher than the control group (188.24+54.54) mol/L in PIH group, the average level of serum uric acid in severe, moderate and mild degree is decreasing, respectively (449.32+81.11), (352.89+76.41), (288.76+59.58) mol/L, a statistically significant difference between the groups (P<0.05).ConclusionThe detection of serum uric acid level in pregnant women can effectively determine the degree of pregnancy induced hypertension in pregnant.

8.HPLC fingerprints of tibetan medicinal herb "songdi" (Saxifraga umbellulata var. pectinata).

The research was carried out to establish HPLC fingerprints of Tibetan medicinal herb "Songdi" (Saxifraga umbellulata var. pectinata), and to provide reference for identification an quality control of it. It was performed on an Amethyst-C18-P (4.6 mm x 250 mm, 5 microm) column with the mobile phase of methanol-0.4% formic acid in a linear gradient mode at a flow rate of 1.0 mL x min(-1). The column temperature was 30 degrees C, and the detection wavelength was set at 254 nm. The software for chromatographic fingerprint was applied to analyse the pattern analysis, the common peaks and similarity. Cluster analysis was done based on the common peaks data of 33 samples from different plant species and sources by SPSS software. Ten common chromatographic peaks were identified by fingerprint, showing a low similarity in constituent and variety. Flavonoids and saponins were the principal components. The number and area of peaks were affected by the collection sources and method. The high similarity are showed by the samples derived from the same area with high accuracy and high purity. The method is so simple, exclusive, stable and high repeatable that it can provide reference for identification and quality assessment of "Songdi" (S. umbellulata var. pectinata).

9.Quality Indicators in Laboratory Medicine:A Fundamental Tool for Quality and Patient Safety

Quality indicators (QIs)in laboratory medicine can measure how well a clinical laboratory meets the needs and re-quirements of users and the quality of all operational processes.The identification of reliable QIs is a crucial step in enabling users to quantify the quality of laboratory services.Different QIs and terminologies are currently used and,therefore,there is the need to harmonize proposed QIs.A model of quality indicators has been consensually developed by a group of clinical la-boratories according to a project launched by a working group of the International Federation of Clinical Chemistry and La-boratory Medicine.The model includes 57 QIs related to key processes (35 pre-,7 intra-and 15 post-examination phases)and 3 to support processes.The developed a model of quality indicators and the data collected provide evidence of the feasibility of the project to harmonize currently available QIs,but further efforts should be done to involve more clinical laboratories and to collect a more consistent amount of data.
10.Detection techniques for calreticulin gene mutation and the clinical application

The mutation status of calreticulin gene (CALR) is helpful for the diagnosis of JAK2 / MPL mutation-negative myeloproliferative neoplasms (MPN), and is closely related to the MPN progression and prognosis.Currently, Sanger sequencing, PCR-fragment analysis, high resolution melting, Taqman RQPCRand the next generation sequencing have been reported to be used to detecting the CALR gene mutations.A proper method for CALR mutation detection and a right laboratory diagnostic procedure according to the MPN-related molecular markers will facilitate the rapid and effective diagnosis and treatment of MPN.
