2.Effects of delayed remote limb ischemic preconditioning on myocardial injury in patients undergoing cardiac valve replacement under cardiopulmonary bypass

Objective To evaluate the effects of delayed remote limb ischemic preconditioning (DRLIP) on myocardial injury in patients undergoing cardiac valve replacement under cardiopulmonary bypass (CPB).Methods A total of 41 patients of both sexes,aged 55-70 yr,weighing 50-75 kg,scheduled for elective cardiac valve replacement under CPB,of ASA physical status Ⅱ or Ⅲ (NYHA Ⅱ or Ⅲ),were randomly divided into 2 groups using a random number table:control group (group C,n =20) and group DRLIP (n =21).In group DRLIP,the patients underwent three 5-min cycles of unilateral lower limb ischemia,induced by a manual cuff-inflator placed on the left thigh and inflated to 200 mmHg starting from 24 h prior to surgery.Blood samples were taken from the central vein before aortic clamping,at 6 h of reperfusion and at 24 h after surgery for determination of plasma cardiac troponin Ⅰ concentrations.Before aortic clamping and at the end of CPB,myocardial tissues were obtained from the right auricle for determination of the expression of caspase-3 and cell apoptosis.Apoptotic index was calculated.The recovery of spontaneous heart beats was recorded.Results Compared with group C,plasma troponin Ⅰ concentrations were significantly decreased at 6 h of reperfusion,and the expression of caspase-3 was downregulated,apoptotic index was decreased at the end of CPB,and no significant change was found in the incidence of recovery of spontaneous heart beats in group DRLIP.Conclusion DRLIP can protect myocardium against injury in the patients undergoing cardiac valve replacement under CPB,and inhibition of cell apoptosis is involved in the mechanism.
3.Targetting microbubbles of tumor lymphangiogenesis and ultrasound contrast imaging in vivo

Objective To optimizing preparate for target microbubbles of tumor lymphangiogenesis which load anti-VEGFR-3 and anti-Podoplanin and to evaluate the imaging of this microbubbles Methods The biotinylated anti-VEGFR-3 and anti-Podoplanin were conjugated to biotinylated microbubbles through biotin-avidin its technology of preparation and progress were optimized The proportion of reagents were adjusted The targeted microbubbles were evaluated by immunofluorescence method and its effect was tested in vivo Results High adhesion rate target microbubbles were successfully achieved which average particle size was 0 99 μm and the average antibody combined rate was 81 3% Confirmed by flow cytometry and fluorescence microscope it can clearly show the lymphangiogenesis and the metastasis of lymph nodes in vivo Time-intensity curve showed more microbubbles more intensity more stay and better image Conclusions These target microbubbles have a small particle size and high adhesion rate and a better contrast imaging It is a more value ultrasound target contrast agents of lymph vessel with tumor and sentinel node.
5.Advances in poly (N-isopropylacrylamide) based platforms for cell culture.

Poly (N-isopropylacrylamide) (PNIPAAm), a temperature-responsive polymer, can be potentially applied to replace enzymes or cell scrapers to recover attached cells. Taking full advantage of this unique function of PNIPAAm, cells can be protected from enzymatic hydrolysis and mechanical treatment, thereby to provide ideal seed cells with high quality for biomedical fields. In this review we describe the method to facilitate cell effective adhesion and rapid detachment on thermoresponsive two dimensional surfaces, including selecting special substrate, introducing hydrophilic group, adjusting reactant ratio, controlling polymer thickness/density, providing appropriate external force, so as to effectively improve adherent cell adaptability to thermoresponsive surfaces, depress the risk of bacterial contamination and reduce the effect of low-temperature treatment on the cells. The three dimensional cell culture systems involved in temperature-sensitive microcarriers, scaffolds and gels were briefly discussed. The application based on the platforms for cell culture was also presented.

6.Dynamic detection of T cell subsets and CD3+HLA-DR+T cells in peripheral blood with hand allograft recipients

Objective To study dynamic changes of T cell subsets and CD3+HLA-DR+T cells in peripheral blood of composite tissue transplantation-hand allograft recipients. Methods The levels of CD3+、CD3+CD4+、CD3+CD8+、CD3+HLA-DR+T cells and ratio of CD4/CD8 in peripheral blood of hand allograft recipients in different periods were examined by using flow cytometry.The recipients before transplantation were as the control groups.Results The first day after transplantation,the levels of CD3+、CD3+CD4+、CD3+CD8+、CD3+HLA-DR+T cells and ratio of CD4/CD8 all began to descend.The 3th to 5th day after transplantation,the levels were the lowest.The 8th day,the levels of CD3+、CD3+CD4+、CD3+CD8+、CD3+HLA-DR+T cells and ratio of CD4/CD8 were eventually rised and go to stable 15th day after transplantation.But the levels of CD3+、CD3+CD4+and value of CD4/CD8 were still lower than those of the contols,and the levels of CD3+CD8+、CD3+HLA-DR+T cells were higher distinctly.Conclusion Dynamic changes of T cell subsets and active T cells in peripheral blood of composite tissue transplantation-hand allograft recipients were accordant with that of renal allograft recipients with stable period and with stable clinical state of the hand transplantation recipients.
7.Isolation of Carbapenems-resistant Gram-negative Bacillus and Analysis of Producing Metallo-β-lactamase

OBJECTIVE:To provide reference for rational use of antibiotics in the clinic of our hospital. METHODS:Drug re-sistance of Gram-negative bacillus in the inpatients of our hospital were analyzed retrospectively during May 2013-Dec. 2015 as well as the situation of producing metallo-β-lactamase(MBLs). RESTUTS:A total of 2089 strains of Gram-negative bacillus were detected in our hospital during 2013-2015,among which there were 1456 strains of enterobacteria (69.70%) and 633 strains of non-fermentative bacteria,mainly involving Escherichia coli,Pseudomonas aeruginosa,Klebsiella pneumoniae,Acinetobacter bau-mannii and Enterobacter cloacae. A total of 406 strains of carbapenems-resistant bacteria were detected (19.44%),including 367 strains of non-fermentative bacteria and 39 strains of enterobacteria. The resistant rates of carbapenems-resistant strains to 16 antibi-otics were all higher than 50%,but those of non-carbapenems-resistant strains were in relative low level. Except for aztreonam,re-sistant rates of carbapenems-resistant strains to other 15 antbiotics were all higher than those of non-carbapenems-resistant strains, with statistical significance(P<0.05). A total of 36 strains of producing MBLs were detected(8.87%),including 13 strains of pro-ducing MBLs drug-resistant P. aeruginosa and 23 strains of producing MBLs drug-resistant A. baumannii;producing MBLs drug re-sistant enterobacteria had not been found. CONCLUSIONS:Gram-negative bacillus are mainly enterobacteria in our hospital;car-bapenems-resistant strains are mainly non-fermentative bacteria,resistant rate of them are commonly higher than that of non-drug-re-sistant strain. The situation of producing MBLs is serious,and enzyme producing strains are mainly non-fermentative bacteria. It is necessary to strengthen drug resistance of pathogen and enzyme producing strain monitoring,avoid the generation and spreading of drug-resistant strains due to irrational use of antibiotics.

To analyze the reasons of injuries during surgical practices and explore the hazards and the corresponding preventive measures. The surgical interns don't receive enough training of injury prevention. They don't pay enough attention to the occupational prevention and are not skilled in operation, which causes high rate of sharp edged instrument injuries. Therefore, a perfect plan of education, training and treatment must be made to reduce the occupational injuries and blood sourced diseases.
10.Significance of Expression of CD_(14)~+ CD_(16)~+ on Peripheral Monocytes in Children with Kawasaki Disease

Objective To observe the significance of expressions of CD14+CD16+ on peripheral monocytes in children with Kawasaki di-sease (KD).Methods The expression of CD14+ and CD14+CD16+ monocytes in 16 children with KD (1-11 years old) were analyzed by flow cytomety both pre-treatment and post-treatment.And the percentages of CD14+CD16+ monocytes among CD14+ monocytes were calculated.Sixteen healthy children (10 months -10 years old) were served as normal control group.Statistical analysis was performed using t test.Results The levels of CD14+ monocytes,percentage of CD14+CD16+ monocytes among CD14+ monocytes and CD14+CD16+ monocytes in children with KD during acute phase (n=16) were (1.03?0.58)?109 L-1,(12.53?5.31)% and(1.20?0.79)?108 L-1.They were significantly higher than those in the normal controls[(0.57?0.21)?109 L-1,(3.86?1.84)% and (0.21?0.10)?108 L-1](Pa0.05).And the expressive levels remained high when the patient recurred.Conclusions The expressive levels of CD14+CD16+ monocytes increase in children with KD.And they change when the patient's clinical condition change.
