1.Application of glucose descriptive chart in monitoring of stress hyperglycemia

ObjectiveTo study the application of glucose descriptive chart in monitoring of stress hyperglycemia,and provide continuous,dynamic change of blood glucose levels for clinic. Methods80 patients with stress hyperglycemia who adopted intensive insulin therapy were randomly divided into the experimental group and the control group with 40 cases in each group.The experimental group used glucose descriptive charts to record blool glucose,the control group used the traditional registration form to record blood glucose.The incidence of hypoglycemia and evaluation of medical staff for the glucose descriptive chart were compared between two groups. ResultsThe incidence of hypoglycemia in the experimental group was significantly lower than the control group.The medical staff considered that glucose descriptive chart helped understand the dynamic change of blood glucose,and provided a more comprehensive basis for regulating the use of insulin. ConclusionsApplication of glucose descriptive chart in monitoring of stress hyperglycemia can provide continuous and dynamic change of blood glucose,which facilitates the medical staff to monitor the blood glucose and decrease the incidence of hypoglycemia.
2.Application of self-made retractor in laparoscopic thyroidectomy

Objective To compare the difference between self-made retractor and thyroid suspension in laparoscopic thyroidectomy,as well as exploring the feasibility and efficacy of self-made retractor.Methods Fifty nine thyroid patients were included in this study from Jan.2012 to Jun.2013 in our hospital.30 cases of laparoscopic thyroidectomy were completed by the help of self-made retractor while 29 cases were completed with thyroid suspension.The owration time,blood loss,drainage flow,hospital stays and short term effect were compared.Result The average operation time of retractor group (71.57 ± 9.56) min was shorter than the suspension group (79.90 ±10.61) min (P =0.002) The blood loss of retractor group (11.83 ±6.76) mL was less than the suspension group (17.24 ± 9.41) mL (P =0.014).The hospitalization time of retractor group (4.07 ± 0.45) days was similar with the suspension group (4.14 ± 0.52) min (P =0.573),The retractor group had shorter operation time,blood loss.There was statistical significance between them.There was no significance difference of hospital stays and discomfort.Neither group appeared of hoarseness,bucking,angulus oris numbness and other symptom.Conclusion Self-made retractor can reduce the operation time,blood loss.It is worth of clinical extension.
3. Lanthanum chloride induces apoptosis in HeLa cells via Caspase-3 pathway

Objective: To explore apoptosis-inducing effect of lanthanum chloride in cervical cancer cells in vitro and the related mechanism, so as to provide a theoretical basis for medical use of rare earth complex. Methods: The anti-tumor activity of lanthanum chloride against HeLa cells was examined by morphological observation and MTT (3-[4, 5-dimethyrthiazol-2-yl] - 2, 5-diphenyltetrazolium bromide) assay. Distribution of cell cycle and cell apoptosis were analyzed using flow cytometry (FCM). The expression and activation of Caspass-3 were examined by Western blotting assay and spectrophotometry. Results: Morphological observation and MTT assay showed that lanthanum chloride significantly induced apoptosis in HeLa cells and inhibited cell growth in a doss-dependent manner (P<0.05). FCM analysis showed that the cell ratio of G0/G1 phase increased and that of S phase decreased significantly with the increase of lanthanum chloride doses. Lanthanum treatment also induced cell apoptosis doss-dependently. The minimum dose of lanthanum chloride to trigger apoptosis was 5 μmol/L, and the apoptosis rate increased in a doss-dependent manner compared with the control group (P<0.01). Western blotting analysis showed that lanthanum chloride up-regulated Caspass-3 expression in a doss-dependent manner. Conclusion: Lanthanum chloride can inhibit growth of human cervical cancer HeLa cells and induce cell apoptosis via Caspase pathway.
4.Clinical value of Antibody D5F3 and Survivin in patients with non- small cell lung cancer

Objective To investigate the clinical value of D5F3 and Survivin in patients with non-small cell lung cancer. Methods 100 patients with non - small cell lung cancer admitted in our hospital from January 2016 to June 2017 were examined by immunohistochemical method, to analyze the clinical value of D5F3 and Survivin antibody in non - small cell lung cancer (NSCLC). Results The coincidence rate of D5F3 was (20%), (++) was 33.33%, and the coincidence rate was (87.50%), and the difference was statistically significant (P<0.05). The positive rate of Survivin protein expression was correlated with the differentiation degree of non-small cell lung cancer, TNM stage and lymph node metastasis (P<0.05). Conclusion D5F3 has high accuracy, sensitivity and specificity, and has important clinical diagnostic value; The detection of Survivin expression is helpful for clinical differentiation, clinical staging and lymph node metastasis in patients with non-small cell lung cancer.
5.Effects of erythropoietin on proliferation and differentiation of neural stem cells originated from adult rats brain

Objective To approach the influence of erythropoietin on proliferation and differentiation of neural stem cells originated from the hippocampi of adult rats in vitro. Methods Neural stem cells were isolated from the hippocampi of adult rats and cultured in vitro with serum-free incubation and single-cell cloning technique; immunochemistry technique was employed to identify the neural stem cells and neural stem cells-derived progeny. Erythropoietin (EPO) were added to cultures in different concentrations during proliferation and differentiation of neural stem cells, respectively. Results Compared with the control group, EPO supplementation group displayed an average twofold to fourfold increase in Nestin positive cells over a wide range of plating densities during proliferation, whereas showed a striking decrease in Nestin positive cells during differentiation,following an early appearance of Tuj1 or GFAP positive cells. 6.Laser peripheral iridoplasty combined with iridectomy for the unmanageable acute angle-closure glaucoma by medication

AIM: To investigate the effect and safety of laser peripheral iridoplasty combined with iridectomy in the unmanageable acute angle - closure glaucoma by medication.
METHODS:Totally 19 cases (21 eyes) with acute angle-closure glaucoma, including 15 cases ( 17 eyes ) with primary glaucoma and 4 cases (4 eyes) with intumescent cataract - induced glaucoma, were recruited into the study. The intraocular pressure ( IOP ) of all cases were still >21mmHg after 24h drug treatment, and then were treated by laser peripheral iridoplasty combined with iridectomy. The visual accurity, IOP, cornea, peripheral anterior chamber depth, anterior chamber angle and complications were observed at 24h after the surgery.
RESULTS:The mean IOP of all cases was reduced from 53. 09±11. 01mmHg before the surgery to 14. 98±4. 21mmHg at 24h after the treatment, with significant statistical difference ( P< 0. 01 ). The visual acurity of all cases increased in different degrees from handle move to 0. 3 to 0-1-1. 0 at 24h after the treatment. In all cases, cornea edema reduced or cleared up, peripheral anterior chamber depth increased, and anterior chamber angle reopened in different degrees. Complications included iris hemorrhage in 11 eyes (52. 4%), mild iritis in 21 eyes (100%). No cornea burn was encountered.
CONCLUSION: Laser peripheral iridoplasty combined with iridectomy is an effective and safe method for the treatment of the unmanageable acute angle - closure glaucoma by medication.
7. A goat model for research of anterior cervical-adjustable fusion fixator

Objective: To establish an animal model for anterior cervical-adjustable fusion fixator (AC-AFF), and to compare the operation time, blood loss, and recovery time of food intake and movement between AC-AFF group and other groups with different cervical reconstruction fixators. Methods: Eighteen goats were evenly randomized into 3 groups: AC-AFF, steel plate+ titanium mesh, and steel plate+autogenous iliac bone group. The 3 fixtors were implanted after corpectomy. The operation time, blood loss during operation, and recovery time of food intake and movement were recorded in each animal and were compared. Results: All experiment animals survived after operation; one animal of steel plate+titanium mesh group suffered from incision infection and recovered 5 days after intramuscular penicillin therapy. Two animals in steel plate+autogenous iliac bone group could not stand on forehoof, but food intake was not influenced; the animals could stand 1 week later without treatment and walked with difficulty, and they recovered normal walk 2 weeks later. Four animals received C3 corpectomy (2 received AC-AFF and 2 received steel plate+titanium mesh) and the rest received C4 corpectomy. It was found that AC-AFF group had the shortest operation time (P<0.01), and there was no difference between the other 2 groups. The intra-operation blood loss in the steel plate+autogenous iliac bone group was significantly higher than that in the other 2 groups (P<0.01), with no significant difference found between the latter 2 groups. Conclusion: AC-AFF is easier to manage than the other 2 fixators; besides, AC-AFF has less blood loss, short operation time, less post-operation complication, and early recovery of food intake and movement.
8. Bone graft fusion in goat model implanted with anterior cervical-adjustable fusion fixator after anterior cervical corpectomy: An observation of outcome

Objective: To observe the bone graft fusion of goat cervical model implanted with anterior cervical-adjustable fusion fixator(AC-AFF), laying a foundation for future studies. Methods: Eighteen experiment goats were implanted with AC-AFF, titanium mesh and autogenous iliac bone combined cervical plate after corpectomy. Six months later, the fused cervical spine vertebrae were collected, prepared, and observed grossly, under microscope, by X-ray photographs and CT scan; the fusion of the graft was evaluated. Meanwhile, the fusion of AC-AFF with the adjacent vertebral surface was observed. Results: All the experiment animals survived after operation; all the implants were stable, without displace or loosening. The interface where the titanium or AC-AFF contacts the vertebral body surface was bulged and ossified. All iliac bone graft fused well and a great deal of bony callus was found covering the vertebral body surface. Lucency area was not seen surrounding the bone graft, titanium and AC-AFF on the X-ray films, and there was new bone formation in the interface of internal fixation and bone. The central region of titanium and AC-AFF became vague and new bone formation was found in the intracavitary area of implants by CT scan. And part of the new bone transited the parietal reticulation and connected with normal ossea at titanium and AC-AFF. A large number of phoroblasts and chondrocytes were found microscopically in the bone fusion sites in all cases; however, the region without bony callus still existed in the titanium mesh lateral wall. Conclusion: There are no differences in bone fusion between AC-AFF with titanium mesh and autogenous iliac bone combined cervical plate. New phoroblasts and chondrocytes can be generated in the bone graft interface. AC-AFF has been proven as an important method for cervical stability reconstruction after decompression operation.
9.Experimental Study of Needle Knife Treatment for Knee Osteoarthritis

Objective To explore the mechanism of therapeutic action of needle knife “regulating sinews and treating bones” on knee osteoarthritis (KOA) by observing the effect of needle knife intervention on KOA rabbit behaviors, mechanical characteristics of patellar ligament (PL), and expressions of interleukin-4 (IL-4), matrix metalloproteinase-3 (MMP-3) and aggrecan in knee cartilages.Methods Forty New Zealand rabbits were randomly allocated to blank, model, needle knife and electroacupuncture groups, 10 rabbits each. A rabbit model of KOA was made by the modified Videman method of immobilization in extension position. After model making, the needle knife and electroacupuncture groups received needle knife and electroacupuncture treatments, respectively. A behavioral assessment was made using the modified Lequesne MG knee grade evaluation method in every group after model making and treatment. The samples were taken after treatment. PL tension, stress relaxation and creep state were tested using a Bose Electro Force 3300 protracted test machine. Cartilage cell IL-4 expression was examined by ELISA. MMP-3 mRNA and aggrecan mRNA expressions were detected by real-time PCR.Results After model making, there was a statistically significant difference in the Lequesne MG score between the model and blank groups (P<0.01); there was no statistically significant difference in the Lequesne MG score between the needle knife or electroacupuncture group and the model group (P>0.05). There was a statistically significant post-treatment difference in the Lequesne MG score between the needle knife or electroacupuncture group and the model group (P<0.01,P<0.05) and between the needle knife and electroacupuncture groups (P<0.05). There were statistically significant post-treatment differences in PL maximum stress, maximum displacement, elastic modulus, stress relaxation rate and creep rate between the model and blank groups (P<0.01,P<0.05). There were statistically significant post-treatment post-treatment differences in PL maximum stress, maximum displacement, elastic modulus, stress relaxation rate and creep rate between the needle knife and model groups (P<0.01,P<0.05). There was a statistically significant post-treatment difference in elastic modulus between the electroacupuncture and model groups (P<0.01). The IL-4 content and aggrecan mRNA expression decreased significantly and MMP-3 mRNA expression increased significantly in the model group after model making and there were statistically significant differences compared with the blank group (P<0.01,P<0.05). After treatment, the IL-4 content increased significantly in the needle knife and electroacupuncture groups compared with the model group (P<0.01,P<0.05) and aggrecan mRNA expression tended to increase in the two groups. The regulation of aggrecan mRNA and MMP-3 mRNA expressions was better in the needle knife group than in the electroacupuncture groups, but there was no statistically significant difference compared with the model group (P>0.05).Conclusion The mechanism of action of needle knife treatment on KOA may be that it improves ligament mechanical characteristics, regulates intra-articular stress environment, and modulates aggrecan mRNA and MMP-3 mRNA expressions and inhibits cartilage degeneration through IL-4 mechanical signal pathway, to produce the therapeutic effect of “regulating sinews and treating bones”.
