4.Application of biodegradable materials in the clinical treatment and care of gynecology and obstetrics

BACKGROUND:Recently, the biodegradable materials with good biocompatibility and with no adverse reaction have been widely applied in the clinical treatment and care of gynecology and obstetrics. The development of the biodegradable material is very rapid. OBJECTIVE:To summarize the application of biodegradable materials in gynecology and obstetrics. METHODS: The first author searched PubMed and CNKI databases for articles related to biodegradable materials in gynecology and obstetrics using the keywords of “degraded materials, biodegradable materials, gynecological care, surgery” in Chinese and English respectively. This article has an outlook of the potential application in gynecology and obstetrics based on clinical experience. RESULTS AND CONCLUSION:Biodegradable materials with good biocompatibility and biological security play an important role in biomedical materials. Their polymers and degradation products show smal adverse reactions to the body, and have good biomechanical properties, physical and chemical properties and good workability, which have been widely used in gynecology and obstetrics, such as absorbable stylolite and tampon tape. Although we have made a great progress on the biomaterial research, it stil has the limitation and safety flaws in the clinic. As the biomaterial research is further developed, the biomaterial application prospect wil be more promising.
5.Effects of triptolide on endothelin-1 and its gene expression in lung of asthmatic guinea-pigs

AIM To explore the effects of trip-tolide on endothelin-1 and its gene expression in lung of asthmatic guinea-pigs. METHODS The contents of ET-1 in lung of asthmatic guinea-pigs were measured with radioimmunoassay, and the expressions of ET-1, ETA, ETB and ECEmRNA with reverse transcription DNA polymerase chain reaction (RT-PCR). RESULTS 0 All of the indexes were increased after guinea-pigs were induced asthma. At the 4th hour after first induction of asthma, ET-1 reached the top, but at the 2rd hour, ET-1mRNA, ETAmRNA and ECEmRNA were markedly higher than control group (P
6.Prokaryotic Expression of the Partial gB Gene of the Marek’s Disease Virus

The partial segment of Marek′s disease virus (MDV) glycoprotein B (gB) gene was amplified by PCR. The segment was cloned into pET-28a vector to obtain the recombinant pET-gB plasmid. The recombinant plasmid was transformed into E.coli BL21,and expressed in very high level as inclusion body after induced with 1.0mmol/L IPTG. The inclusion body was solubilized in urea (8mol/L) . The purified protein was obtained by use of His?Bind affinity chromatography. Mice were immunized i.p. by the purified protein to make the polyclonal antibody. The titer of the antibody by indirect ELISA was 1?10~ -5 . Moreover, the analysis by western blot proved that antibody was specific to the recombinant protein. These works lay a favorable foundation for the study of the immune response by MDV gB.
7.A study on the facial morphometry of the attractive Uygur females in Xinjiang

Objective:To obtain the normal values of the facial soft tissue measurements of the attractive Uygur females in Xinjiang by photogrammetric analysis,and to study the relationship between the facial feature of attractive Uygur females and the neoclassical canons. Methods:31 attractive Uygur women were selected, standardized and referenced frontal photographs of faces were taken,and 14 standard anthropometric marks were determined,then measurements were performed by photoshopCS software.Results:Intercanthal width was rather smaller than nose width and larger than eye fissure width;nose width occupied more than 1/4 of face width or 2/3 of mouth width.The chin occupied 2/3 of the entire mandibular height.There was positive correlation among many horizontal and vertical facial soft tissue measurements.Conclusion:The facial features of the attractive Uygur females in Xinjiang fit the classical facial canons to some extent. There is pertinence in attractive facial form and features.
8.Evaluation of left ventricular systolic synchrony in patients with Ⅲ grade atrio-ventricular block after pacemaker implantation by real-time three-dimensional echocardiography

Objective To evaluate left ventricular systolic synchrony in patients with Ⅲ grade atrio-ventricular block(Ⅲ grade AVB) after pacemaker implantation by real-time three-dimensional echocardiography. Methods Methods Twelve patients with Ⅲ grade AVB implanted by VVI pacemaker, seven patients with Ⅲ grade AVB implanted by DDD pacemaker, twenty subjects with normal LV function were examined by Philips iE33 with a X3-1 probe. Results The minimal excursion in patients with Ⅲ grade AVB implanted by VVI pacemaker and DDD pacemaker was less than that in subjects with normal LV function (P <0.05) ,and two "bull eyes" displayed that some regional systolic timing delayed or ahead, but also some regional systolic excursion hypokinesis or dykinesis. Parameter index [including Tmsv 16-SD, Tmsv 12-SD, Tmsv 6-SD, Tmsv 16-Dif, Tmsv 12-Dif, Tmsv 6-Dif, Tmsv 16-SD (%), Tmsv 12-SD (%), Tmsv 6-SD (%), Tmsv 16-Dif (%), Tmsv 12-Dif (%), Tmsv 6-Dif(%)] of 17 segment time to minimal systolic volume in patients with Ⅲ grade AVB implanted by VVI pacemaker and DDD pacemaker was more than that in subjects with normal LV function (all P < 0.05). Conclusions Real-time three-dimensional echocardiography can rapidly evaluate left ventricular systolic synchrony in patients with Ⅲ grade AVB implanted by DDD pacemaker and VVI pacemaker.

10.Parathyroid hormone on the adipogenic potential of rat bone marrow mesenchymal stem cells

Objective To observe the effect of different concentrations of parathyroid hormone 1-34 on the adipogenic potential of rat bone marrow mesenchymal stem cells (BMSCs).Methods ① Rat bone marrow mesenchymal cells were separated and expanded by adherent culture.The morphology of cells was observed and cell surface markers were examined by flow cytometry.② The multi-lineage differentiation capability of cells was examined by culturing cells under conditions favorable for adipogenic and osteogenic differentiation.③ Taken P3 of BMSCs for test,different concentrations of PTH1-34 (0,10-10,10-9,10-8 mol/L) were used to stimulate BMSCs respectively,14 days later,lipoprotein lipase (LPL) activity were measured by enzyme linked immuno-sorbent assay (ELISA),mRNA expression of alkaline phosphatase (ALP) and PPARγ-2 were measured by realiime polymerase chain reaction (PCR).④ Statistical analysis:data were presented as x±s.All statistical analysis was performed with windows Statistical Praduct and Serice Solutions (SPSS) 13.0.One-way analysis of variance (ANOVA) was applied to determine the difference between groups.Least signisicant difference (LSD) was used to determine the difference between the two randomized groups.Differences were considered significant at a value of P<0.05.Results The cells expressed CD44,CD29 but without expression of CD45.By culturing in adipogenic medium for 3 weeks and in osteogenic medium for 4 weeks respectively,and then identified by oil red O and Alizarin red,the cells were successfully induced to adipocytes and osteogenesis.Expressions of LPL were 11.20±0.16,7.62±0.48,5.84±0.57,5.32±0.52,mRNA expressions of PPARγ-2 were 2.80±0.05,1.36±0.23,0.94-±0.11,0.78±0.04,ALP activity were 0.191 ±0.016,0.333±0.024,0.549±0.025,0.684±0.021 respectively.Compared with the control group,different concentrations of PTH1-34 groups could decrease mRNA expression of LPL and PPARγ-2.ALP activity were increased(P<0.05).Conclusion PTH1-34 inhibits BMSCs of adipogenic differentiation and promotes osteogenic differentiation in a dose-dependent manner.
