1.Effect of carthamus tinctorius L.injection on caspase-3,bcl-2 expression related to renal ischemia/reperfusion injury in rats

Objective: To investigate the effect of carthamus tinctorius injection on expression of caspase-3 and bcl-2 after renal ischemia/reperfusion injury.Methods: Seventy-five SD rats were randomly divided into three groups(n=25,in each):normal control group,ischemia/reperfusion group and treatment group.After the renal ischemia/reperfusion injury model was established,the serum creatinine、the urine osmotic pressure and the blood osmotic pressure were measured to evaluate the change of renal function;the apoptosis rate in ischemia/reperfusion renal tissure were measured by TUNEL method;the caspase-3 and bcl-2 concentration were measured by immunohistochemistry.Results:The renal function got worse,and the apoptosis rate,bcl-2 and caspase-3 concentration were significantly higher in ischemia/reperfusion group than those in normal control group.As compared with ischemia/reperfusion group,the renal function was improved,the apoptosis rate decreased,bcl-2 expression increased and caspase-3 expression decreased significantly in treatment group.Conclusion:Carthamus tinctorius injection has significant effect on the up-regulation of bcl-2 and down-regulation of caspase-3 expression in renal ischemia/reperfusion injury.
2.Efficacy and Safety of Ambrette Capsule Combined with Tripterygium Glycosides Tablet versus Tripterygium Glycosides Tablet Alone in the Treatment of Diabetic Nephropathy:A Meta-analysis

OBJECTIVE:To systematically review the efficacy and safety of Ambrette capsule combined with Tripterygium gly-cosides tablet versus Tripterygium glycosides tablet alone in the treatment of diabetic nephropathy to provide evidence-based refer-ence for clinical treatment. METHODS:Retrieved from CJFD,Wanfang database,VIP database,Cochrane Library,EMBase and Medline randomized controlled trials(RCT)about Ambrette capsule combined with Tripterygium glycosides tablet(test group)ver-sus Tripterygium glycosides tablet alone(control group)in the treatment of diabetic nephropathy were collected. Meta-analysis was performed by using Rev Man 5.1 software after data extraction and quality evaluation. RESULTS:A total of 5 RCTs were includ-ed,involving 400 patients. Results of Meta-analysis showed the effective rate [OR=4.10,95%CI(2.27,7.38),P<0.001],24 h uri-nary protein quantificative value[MD=-0.54,95%CI(-0.72,-0.36),P<0.001] in test group were significantly higher than con-trol group,there were statistical significances. alanine aminotransferase value [MD=0.14,95%CI(-1.08,1.36),P=0.82] and changes value of peripheral white blood cells(WBC)[MD=-0.31,95%CI(-0.64,0.03),P=0.07] were not statistical difference between 2 groups. CONCLUSIONS:Efficacy of Ambrette capsule combined with Tripterygium glycosides tablet in the treatment of diabetic nephropathy were superior to Tripterygium glycosides tablet alone with similar safety. Due to limit of methodological quali-ty,more strict,long-term follow-up large-scale RCTs were required to validate the conclusion.
3.Clinical observation of long-term combined drug therapy for benign prostatic hyperplasia

ObjectiveTo observe the curative effect of finasteride combined with terazosin on benign prostatic hyperplasia (BPH).Methods53 elderly patients with BPH continuously received finasteride (5 mg daily) and terazosin (2 mg daily) in our hospital from June 2002 to December 2010.The rating of international prostate symtom score (IPSS),nocturia,prostate volume and bladder residual urine were recorded before and 2 and 5 years after treatment.ResultsThe IPSS,nocturia,prostate volume,and bladder residual urine were better after treatment than before treatment (P< 0.05).The rating of IPSS,nocturia and prostate volume after 5 years of treatment also improved than after 2 years treatment (P<0.05),but there was no difference in bladder residual urine between 5 years and 2 years of treatment.ConclusionsLong-term use finasteride combined with terazosin for treatment of BPH is effective with little side effects.
4. Detection and analysis of methylation status of HHIP gene in human pancreatic cancer cell lines

Objective: To explore the relationship between HHIP expression and its hypermethylation in human pancreatic cancer cell lines, so as to provide new insights into the tumorigenesis of pancreatic cancer. Methods: Human pancreatic cancer cell lines BxPC-3, CFPAC-1, PANC-1, AsPC-1 and PaTu8988s were used in the present study. Reverse transcriptase polymerase chain reaction(RT-PCR) and immunocytochemical S-P method were used to detect the expression of HHIP at mRNA and protein levels before and after treatment with 5-aza-2′-deoxycytidine (5-Aza-CdR), a DNA methyltransferase inhibitor. Methylation-specific PCR (MSP) combined with DNA sequencing were adopted to identify the CpG island methylation of HHIP gene. Results: Expression of HHIP mRNA/protein was absent in the five pancreatic cancer cell lines before 5-Aza-CdR treatment, and re-expressed after treatment with 5-Aza-CdR. CpG island hypermethylation of HHIP gene was observed in the five pancreatic cell lines by MSP and DNA sequencing. Conclusion: Hypermethylation in CpG island of HHIP gene is correlated with the inhibition of HHIP expression in human pancreatic cancer cell lines, and it might play an important role in the development and progression of pancreatic cancer.
5.Feasibility study of differentiation of invitro induced rat bone marrow-derived mesenchymal stem cells into retinal pigment epithelial cells

Objective To investigate the feasibility of differentiation of invitro induced rat bone marrow-derived mesenchymal stem cells(rMSCs) into retinal pigment epithelial (RPE) cells.Methods The rMSCs from Brwon-Norway (BN) rats were isolated and cultured by adherent screening method.RPE cells lysate made by repeated freeze-thawing was put into the rMSCs culture system to identify whether the induced cells could express characteristic label cytokeratin(CK)and S-100 simultaneously or not.Results The growth rate of rMSCs induced by RPE cells lysate was slower and protuberant burr surrounded the fusiform cells.The results of immunoblotting and double immunofluorescence showed that partial induced cells expressed CK and S-100 simultaneously.The result of flow cytometry indicated that 14.1% induced cells expressed CK and S-100 simultaneously.Conclusion Induced by RPE cells lysate,rMSCs can differentiate into RPE cells.

7.Comparison of clinical efficacy of retroperitoneal laparoscopic ureterolithotomy, ureteroscopy or microchannel percutaneous nephrolithotomy in the treatment of impacted upper ureteral calculi

Objective To evaluate the effect of retroperitoneal laparoscopic ureterolithotomy (RLUL),ureteroscopy(URS),microchannel percutaneous nephrolithotomy (mPCNL) in the treatment of impacted upper ureteral calculi.Methods Clinical data of 105 cases with impacted upper ureteral calculi were retrospectively analyzed.The average residence time of stone in the same position was 8.6 (2-49) months and the average stone surface area were 171 (71-361)mm2 in 41 cases received RLUL treatment.The average residence time of stone in the same position was 9.5 (2-47) months and the average stone surface area were 159 (71-342)mm2 in 29 cases received URS treatment.The average residence time of stone in the same position was 8.9 (2-51) months and the average stone surface area were 165(71-353)mm2 in 35 cases received mPCNL treatment.Results In the RLUL group,surgery was successful in all patients and the stone-free rate was 100% ;In the URS group,the surgery success rate was 93.1% and the stone-free rate 1 month later was 92.6% ;In the mPCNL group,the surgery success rate was 97.1% and the stone-free rate 1 month later was 97.0%.The operation time in the URS group (mean 65 min,ranged 45-95 min)was longer than that in the RLUL(mean 46 min,ranged 25-70 min) and mPCNL(mean 51 min,ranged 39-80 min)groups (P<0.05).The hospital stay in the URS group (mean 5.1 days,ranged 4-12 days) was shorter than that in the RLUL(mean 7.9 days,ranged 6-11 days) and mPCNL(mean 8.1 days,ranged 5-18 days)groups (P < 0.05).The operation time and the hospital stay had no significant differences between the RLUL group and mPCNL group (P > 0.05).The surgery success rate,stone-free rate and the serious complications of the three groups had no significant differences (P > 0.05).Conclusion The operation time of URS is shorter,but postoperative recovery is faster than that of RLUL and mPCNL groups.Although the incidence of serious complications has no statistically significant differences among the three treatments,but URS and mPCNL surgery have the potential risk of serious complications.RLUL does not increase the difficulty of operation,has less serious complications,and is a remedy for failure in URS and mPCNL operation.
8.Increased expression of Hes1 and its effect on tumour-formation ability in colon cancer

tumours was found to be slightly faster than those of the controls.Conclusion:Hes1 is overexpressed in colon cancer than that in normal tissue, and Hes1 is upregulated in poorly differentiated cancer samples compared with well-differentiated tumour samples. Hes1 has a positive inlfuence on the ability of tumour-formation in SW620 cellsin vivo.
9.Islet β-cell mass and clinical progression of diabetes mellitus

Normally, pancreatic islet?-cell mass is regulated by?-cell replication, neogenesis, apoptosis and cell size.?-cell mass in diabetic patients is conspicuously less than that in normal persons. Decline of?-cell mass is mainly caused by increased?-cell apoptosis. The change of?-cell mass is clinically becoming more and more important in regard to the progression of diabetes mellitus.
10. Screening of the effective cellulose-degradable strain and its application in the production of cellulose bioethanol

Strains from the cellulose-containing environment were collected. Primary screening (by filter-paper Hutchison solid culture medium and sodium carboxymethylcellulose solid culture medium) and reelection(by filter-paper inorganic salt culture medium and sodium carboxymethylcellulose Congo red culture medium) indicated that five strains obtained were best suited for high performance cellulose degradation. Determination of sodium carboxymethylcellulose activity(CMCA) and filter paper activity(FPA) was accomplished for each of the five. The strongest of the five in CMCA and FPA was applied to the production of cellulose bioethanol by separate hydrolysis and fermentation(SHF) and simultaneous saccharification and fermentation(SSF) respectively.
