1.ISOLATION,CULTURE AND VERIFICATION OF STROMAL AND GLANDULAR CELLS OF HUMAN ENDOMETRIUM

Objective Stromal and glandular cells of human endometrium were successfully isolated and cultured in this experiment. Methods These two types of cells were observed by light microscopy and verified by immunocytochemical staining. Results The result showed that some of stromal cells became adherent after.0 5h of culture.Two shapes of stromal cells were found.One was spindle and the another was polygonal.Immunocytochemical staining method showed that both shapes of stromal cells were positive for vimentin,with 95% of positive rate,and negative for cytokeratin 1,indicating that these cells were endometrial stromal cells.Most of glandular cells became adherent after 24h of culture and formed tightly packed whorls after 4d.Individual cells were polygonal and had a large and round nucleus.Immunocytochemical staining method showed that glandular epithelial cells were positive for cytokeratin 1,with 90% of positive rate,suggesting their nature of epithelial cells. Conclusion Stromal and glandular cells of human endometrium were successfully isolated by using two series of filter and cultured in this experiment. [
2.Monitoring the Process of Bovine Insulin Unfolding by Capillary Zone Electrophoresis

Capillary zone electrophoresis (CZE) was used to monitor the whole unfolding process of bovine insulin resulted from the dithiothreitol (DTT) reduction. An off-line matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS) was employed to determine the molecular weight of the reaction products at the same time to confirm the observation results obtained by CZE. The structural change during the process of bovine insulin unfolding could be observed directly from the electropherogram and information of protein unfolding could be obtained simultaneously. The results indicated that as an effective tool of monitoring the conformational change of protein,the method of CZE was simple,quick,sensitive and lower sample consumption.
3.Differential gene expression profiles in paclitaxel-induced cell cycle arrest and apoptosis in human breast cancer MCF-7 cells

Aim To elucidate the molecular mechanism of cell cycle arrest and apoptosis of MCF-7 cells induced by paclitaxel. Methods Flow cytometry was used to determine the cell cycle changes of MCF-7 cells upon paclitaxel treatment. Gene expression profiles of MCF-7 cells induced by paclitaxel were obtained by using cDNA microarrays containing 9 984 genes and expressed sequence tags (ESTs).Results Cell cycle analysis showed that 77.8% of cells arrested at G2/M phase and 1.3% of cells caused G2/M cell cycle arrest and apoptotic cell death in a concentration-dependent manner, which is associated with the regulation of selected genes related to microtubule assembly and cytoskeleton, cell cycle regulation, and DNA repair and apoptosis.
4.Preliminary study on the development of germ cells from human fetal testicular tissues xenografted into the mouse.

OBJECTIVETo investigate the development of xenografted primitive human germ cells by using fetal testicular tissues as donor tissues and an immunodeficient mouse as the recipient.
METHODSTesticular tissue fragments of a 26-week fetus were grafted under the back skin of a castrated immunodeficient mouse. Grafts were taken out after 135 days and processed for morphological and histological analyses.
RESULTSThe mass of grafts grew from about 1 mm in diameter and 5 mg in wet weight to about 3 mm and more than 20 mg 135 days after grafting. Histological observations showed a significant expansion of seminiferous tubules after grafting (80 +/- 25 microm in diameter) in comparison with seminiferous cords at the time of grafting (60 +/- 15 microm in diameter). The seminiferous cords developed into seminiferous tubules with the epithelial border and lumen. After 135 days of grafting, most of the dispersedly distributed primitive Sertoli cells and germ cells migrated to the basal part of seminiferous epithelium, located on the basement membrane and few of germ cells differentiated into spermatogonia.
CONCLUSIONHuman fetal testicular tissues could survive and continuously develop after being xenograft into castrated immunodeficient mice.

5.Karyotype analysis and fluorescence in situ hybridization detection of 122 patients with myelodysplastic syndrome.

OBJECTIVETo assess the value of karyotype analysis and fluorescence in situ hybridization(FISH) assay for the diagnosis of myelodysplastic syndrome (MDS).
METHODSThe karyotypes of 122 initially treated MDS patients were analyzed with conventional R-banding and FISH using probes including GLP CSF1R/D5S23, D5S721, GLP EGR1/D5S23, D5S721, GLP D7S486/CSP7, GLP D7S522/CSP7, GLP D20S108, CSP8 and CSP X/Y.
RESULTSThe detection rate of chromosomal abnormalities was 54.9% for the 122 patients. Among these, those involving 3 or more chromosomes are most common (16.4%), followed by +8(14.8%), -7/7q-(7.4%), -5/5q-(5.7%), 20q-(2.5%), and -Y in male patients (5.0%). Two MDS-RAEB II patients detected with t(8;21) should be diagnosed with acute myelocytic leukemia. FISH analysis showed that 54 patients were positive (44.3%). Among these, 30.3% had CSP8 amplification, followed by GLP D7S486/CSP7 and GLP D7S522/CSP7 deletion (12.3%), GLP CSF1R/D5S23, D5S721 and GLP EGR1/D5S23, D5S721 deletion (9.8%), GLP D20S108 deletion (7.4%), and CSPX/Y deletion (5%).
CONCLUSIONWith a detection rate of 54.9%, R-banding still constitutes the basic examination for MDS. As detection of interstitial chromosomal abnormalities in MDS can be greatly enhanced by FISH, combined karyotype analysis and FISH can improve the diagnosis of MDS and facilitate assessment of its prognosis.

6.Bibliometric analysis of researches on liver organoids

BACKGROUND:In recent years,the development of liver organoids has made it a hot spot in the field of international liver disease research,but there is still no article on the bibliometric analysis of liver organoids. OBJECTIVE:To explore the hot trends in liver organoids in the last 20 years based on bibliometrics and visualization analysis. METHODS:We searched the articles about liver organoids in the Web of Science Core Collection from January 1,2002 to November 12,2022.Origin,Office,and CiteSpace software were used for bibliometrics and visualization analysis.We statistically analyzed the number of annually published articles,countries,institutions,authors,journals,and keywords of the articles by generating charts. RESULTS AND CONCLUSION:The number of articles,citation frequency,institutions and personnel involved in the research about liver organoids showed an overall upward trend in the last 20 years,indicating that the field was growing rapidly and attention was increasing.The USA had published the most papers and had the strongest influence in this field.Although it had invested a lot of time and energy,the number of papers published by a single research institution in the USA was not the highest among many research institutions.China was second only to the USA in the number of publications,with the Chinese Academy of Sciences and Fudan University leading the list.Utrecht University in the Netherlands was the institution with the most publications.Clevers H was the author with the highest number of articles.The article with the highest co-citation frequency was"Long-term culture of genome-stable bipotent stem cells from adult human liver".The main fields of study for liver organoids were Molecular Science,Biology,and Immunology.The most frequently occurring keywords were stem cell,in vitro,and culture.The research hotspots in the liver organoids field were mainly focused on in vitro stem cell three-dimensional culture,differentiation and gene expression.
7.Effects of Forsythia suspensa ethanol extract on the proliferation,migration and invasion of lung cancer cells NCI-H226

OBJECTIVE To investigate the effects of Forsythia suspensa ethanol extract on the proliferation, migration and invasion of lung cancer cells NCI-H226. METHODS As research objects, lung cancer cells NCI-H226 were divided into control group, F. suspensa ethanol extract low-, medium- and high-concentration groups (5, 10, 20 mg/mL), activator group [10 mg/mL F. suspensa ethanol extract+0.5 μmol/L nuclear factor kappa B (NF-κB) signaling pathway activator PMA], inhibitor group (10 mg/mL F. suspensa ethanol extract+10 μmol/L NF-κB signaling pathway inhibitor BAY 11-7082) and positive control group (20 μg/mL cisplatin). Except for the control group of cells without intervention, all other groups of cells were cultured with corresponding drugs for 24 hours; the proliferation, migration and invasion of cells were all detected, and the proliferation rate, migration rate, and the number of invading cells were also calculated; protein expressions of NF-κB p65, NF-κB inhibitory protein α (IκBα), phosphorylated NF-κB p65 (p-NF-κB p65) and phosphorylated IκBα (p-IκBα) were determined. RESULTS Compared with control group, the proliferation rate, migration rate, and the number of invading cells as well as the protein expressions of p- IκBα and p-NF-κB p65 were decreased significantly in F. suspensa ethanol extract groups and positive control group (P<0.05). Compared with F. suspensa ethanol extract medium-concentration group, the proliferation rate, migration rate, and the number of invading cells as well as above protein expressions were all decreased significantly in inhibitor group (P<0.05), while those of activator group were increased significantly (P<0.05). CONCLUSIONS F. suspensa ethanol extract can inhibit the proliferation, migration and invasion of lung cancer cells NCI-H226, and the mechanism of which may be related to the inhibition of NF-κB signaling pathway.
8.Study on the value of pharmaceutical service related to risdiplam based on follow-up data of DTP pharmacy

OBJECTIVE To explore the value of providing pharmaceutical service related to risdiplam in direct-to-patient (DTP) pharmacies. METHODS The follow-up data of spinal muscular atrophy (SMA) patients who purchased and used risdiplam from Shangyao Yunjiankang Yiyao Pharmacy (Shanghai) Co., Ltd. from May 2021 to January 2023 were collected. The medication information, therapeutic efficacy and the occurrence of adverse events were retrospectively analyzed. RESULTS A total of 42 prescriptions were checked by pharmacists in the DTP pharmacies, and 7 prescriptions were found to be unreasonable (16.7%, 7/42), which were corrected after the timely intervention. During the follow-up management, pharmacists replied to 4 patients (9.5%, 4/42) regarding medication consultation about medication requirements and adverse events. Two patients with type Ⅰ SMA experienced adverse events: one of them presented with fever and the other presented with skin dryness with darkening. Both of them were grade Ⅰ toxic reactions and generally did not require clinical treatment. Considering that the patient sustained low-grade fever for a long time, the pharmacist suggested symptomatic treatment under the guidance of the doctor. CONCLUSIONS Pharmacists in DTP pharmacies conducting follow-up management of risdiplam use for rare disease SMA patients can help promote rational, standardized medication for patients.
9.Analgesic Effect and Mechanism of Osteoking on Dorsal Root Ganglion Compression Model Rats

ObjectiveTo investigate the analgesic effect and mechanism of Osteoking (OK) on nerve compression in lumbar disc herniation. MethodThe rat model of chronic compression of dorsal root ganglion (CCD) was established to simulate clinical lumbar disc herniation. The CCD rats were randomly divided into model group, low, medium, and high dose OK groups (1.31, 2.63, 5.25 mL·kg-1·d-1), and pregabalin group (5 mg·kg-1), with eight rats in each group. Another eight SD rats were taken as the blank group, and the same volume of normal saline was given by gavage. Behavioral tests, side effect evaluation, network analysis, Western blot, immunofluorescence, and antagonist application were used to explore the effect. ResultCompared with the blank group, the mechanical hyperalgesia threshold, thermal hyperalgesia threshold, and the expression of inflammatory factors in the spinal dorsal horn of the model group are significantly increased (P<0.01), and the related indicators of the affected foot footprints are significantly down-regulated (P<0.01). The expression of signal transducer and activator of transcription 3 (STAT3), vascular endothelial growth factor A (VEGFA), and phosphorylated extracellular regulated protein kinase (p-ERK) in microglia in the spinal dorsal horn is significantly increased in the model group (P<0.01). Compared with the model group, low, medium, and high dose OK groups can increase the mechanical hyperalgesia and thermal hyperalgesia thresholds of CCD rats (P<0.05, P<0.01) in a dose-dependent manner, improve the gait of CCD rats (P<0.05, P<0.01), and reduce the expression of inflammatory factors in the spinal dorsal horn (P<0.05, P<0.01). The expression of STAT3, VEGFA, and p-ERK in the spinal dorsal horn microglia of CCD rats is significantly decreased (P<0.05, P<0.01), and the acetic acid-induced nociceptive response in rats is effectively reduced (P<0.05, P<0.01). In addition, there is no tolerance. The results of the body mass test, organ index, forced swimming, and rotation show that OK has no obvious toxic or side effects. Further antagonist experiments show that MRS1523 and RS127445 can reverse the transient analgesic effect of OK compared with the high dose OK group (P<0.01). ConclusionOK has a good analgesic effect on the CCD model without obvious toxic side effects, and its mechanism may be related to the activation of ADORA3 and HTR2B and the inhibition of STAT3, VEGFA, p-ERK, and other elements in microglia.
