High mobility group box 1 protein,a kind of damage-associated molecular patterns,is a group of nonhistone DNA-binding protein in the nuclear.It not only involves in DNA replication,recombination,repair,regulation of gene transcription,but also induces activation of innate immunity and T cell generation and activates the adaptive immune response through Toll-like receptor.It is also more closely related to autoimmune diseases,inflammatory diseases,tumors and so on.Currently there are many researches on the role of TLR-mediated HMGB1 signaling pathway in diseases,which is involved in a variety of pathological processes and has a broad clinical outlook.

Hepatitis C virus (HCV)was easily developed into hepatitis,cirrhosis and liver cancer after infecting human.Cur-rently,the traditional method of treatment of HCV was pegylated interferon and ribavirin program,which was ineffective and had poor side effects,while the new direct antiviral drugs were expensive.Therefore,looking for cheap and non-toxic side effects of treatment was the focus of current research.More and more evidence indicate that micro-Ribose Nucleic Acid (miRNAs)play an important role in the development of liver disease,regeneration and functional regulation.This review studies the relationship between miR-122 and Hepatitis C virus and hepatocellular carcinoma.
3.Psychological Investigation of Patients with Oral Carcinoma and Therapeutic Efficacy Analysis of Peri-operative Psychological Intervention

Objective: To explore the psychological status and therapeutic efficacy of peri-operative psychological intervention among patients with oral carcinoma.Method: psychological statuses of 60 patients with oral carcinoma were investigated,and specific peri-operative psychological interventions were performed according to features and initiating factors of their psychological crisis,in order to provide references for the therapy of oral carcinoma.Result: Common psychological crises among patients with oral carcinoma such as anxiety and fear have been dramatically decreased by proper psychological investigation and peri-operative psychological intervention.Conclusion: Psychological investigation contributes to a great improvement of psychological status of patients with oral carcinoma.Therefore,in addition to routine peri-operative medical treatment,an effective psychological investigation is also crucial for patients with oral carcinoma to cure the disease and regain health.
4.Expression of RIG-Ⅰgene in peripheral blood of patients with chronic hepatitis C and its ;correlations with hepatitis C virus load and typeⅠinterferon gene expression

Objective To detect the expression of retinoic acid-inducible gene Ⅰ ( RIG-Ⅰ) in peripheral blood mononuclear cells (PBMCs) of patients with chronic hepatitis C (CHC), and to explore its correlations with type Ⅰinterferon gene expression and serum level of hepatitis C virus .Methods A total of 101 na?ve patients with CHC were enrolled from Renmin Hospital of Wuhan University during July 2014 and July 2015, and 69 healthy subjects served as controls .The expressions of RIG-Ⅰ, IFNαand IFNβmRNA in PBMCs and serum level of HCV RNA were detected by real-time quantitative polymerase chain reaction.Pearson correlation analysis was performed to investigate the correlations of RIG-Ⅰ mRNA expression with IFNα/βmRNA expression and serum level of HCV RNA .Results The relative expressions of RIG-Ⅰ, IFNα, IFNβmRNA to healthy controls in CHC group were 0.082, 0.022 and 0.156, respectively (t=7.83, 3.65 and 2.13, all P<0.05).RIG-Ⅰ mRNA expression in patients with HCV loads <(1 ×106 ) IU/mL was 1.79 times higher than that in patients with HCV loads≥(1 ×106 ) IU/mL (t=2.60, P <0.05).Pearson correlation analysis showed that the expression of RIG-Ⅰ mRNA was positively correlated with that of IFNαmRNA and IFNβmRNA (r=0.247 and 0.489, P<0.05), but not correlated with HCV load (r=0.187, P>0.05).Conclusion Expression of RIG-ⅠmRNA decreases in patients with CHC , and is positively correlated with that of IFNαand IFNβmRNA.
5.Application of Risk Management in Outpatient Blood Collection Work in Grade 3 and First Class Hospital

Objective:To explore the function of risk management in outpatient blood collection work under the conditions of informationization. Method:This paper retrospectively reviewed the nursing risk management in out-patient blood collection work from January 2014 to January 2015 in our hospital, analyzed the causes of risk in out-patient blood collection work, evaluated the possible adverse outcomes, and put forward the measures to prevent and control risks. Results:Through the nursing risk management, the nurses′ risk prevention consciousness was enhanced, as well, both nurse and patient satisfaction was improved. Conclusion:Application of risk management in outpatient blood collection work could improve the quality of nursing, conform to the ethical requirements of guaranteeing patient safety, and effectively reduce the incidence of medical risks and accident.
6.Differentiation of mouse embryonic stem cells into cardiomyogenic cells incubated with cardiomyocytes

Objective To explore the effect of embryonic stem cells(ESCs) differentiating into cardiomyocytes(CM) by cardiomyocytes.Methods The blastocytes of 3.5 d from Kunming mouse,cultured on mouse embryonic fibroblaste cell feeder layers for 4~5 d,the mouse ESCs were isolated and subsequently cultured.Day 2~3 embryoid bodies(EBs) were derived from ESCs of passage 3 to 5 and then incubated with neonatal cardiomyocyte(CM) to induce into cardiomyocytes and the expression of cardiac specific cardiac troponin-T(TnT) and ?-actin were detected by immunofluoresence.Results The rhythmic beating embryoid bodies were observed on day 3 and reached 93% of rhythmic beating embryoid bodies on 12 day in group 2.All the beating cardiomyocytes derived from ESCs expressed cardiac-specific proteins for TnT and ?-actin,and the group induced by direct cell contact acquired the highest differentiating ratio as 56.5%,and was the highest differentiating ratio compared with other groups(P

ABSTRACT:Objective To analyze the stress changes of tooth tissues in the medial-occlusal (Class Ⅱ)cavity restored with different kinds of inlay materials by the three-dimensional (3-D)finite element.Methods The inlay restored 3-D finite element model of medial-occlusal (Class Ⅱ)cavity was established by CBCT scanning method and reverse engineering software Mimics,Geomagic Studio,Pro/E5.0 software,and finite element analysis software. The Von-mises stress and change tendency of tooth tissues and four different kinds of inlay materials were compared after taking vertical loading and tongue 45°to loading.Results There were different stress levels of tooth tissues among different inlays of medial-occlusal (MO)Class Ⅱ cavity.The stress level of the cobalt chromium alloy inlay was the largest,the stress level of the gold alloy inlay was lower,and the stress level of the composite inlay was the lowest.Dental tissues stress distribution was similar for the four different restorative materials,and focused on hole bottom of dentin near the pulp in dentin.Conclusion Compared with ceramics,gold alloy and cobalt chromium inlay restoration,restoration with composite resin inlay can reduce the organization stress of the remaining tooth in the medial-occlusal (Class Ⅱ)cavity type.
8.Study on Expression Interferon Stimulating Gene and Related Signal Pathway Gene in Peripheral Blood Mononuclear Cells of Patients with Chronic Hepatitis C

Objective To detect mRNA expressions of STING and type Ⅰ interferons(IFN-α and IFN-β) in peripheral blood mononuclear ceils (PBMCs) of patients with chronic hepatitis C(CHC).Methods 113 patients with CHC and 94 healthy controls were collected from Renmin Hospital of Wuhan University during January 2015 and January 2016.Expressions of mRNA of STING,RIG-1,IFN-α and IFN-β were detected by real-time quantitative PCR,and their relative expression values were obtained by 2-△△Ct method and the results were statistically analyzed.Results The expression of mRNA of STING,RIG-1,IFN-α and IFN-β in CHC were 0.018,0.361,0.578 and 0.573 times than its in healthy controls respectively and the differences were statistically significant (t-8.28,4.26,2.18 and 2.07,all P < 0.05).Pearson correlation analysis showed that mRNA expressions of STING in healthy controls were positively correlated with that of IFN-α mRNA and IFN-β mRNA (r=0.487 and 0.207,all P<0.05),which had no correlation in CHC (r=0.174 and 0.091,all P>0.05).The expressions of STING mRNA was positively correlated with that of RIG-1 mRNA in healthycontrols (r=0.222,P<0.05),but there was no correlation of expression in CHC between STING mRNA and RIG-1 mRNA (r=-0.029,P>0.05).Conclusion Compared with healthy controls,the expression of STING,RIG-1,IFN-α and IFN-β mRNA were all reduced.There was positive correlation between STING and RIG-1,IFN-α and IFN-β in healthy controls,but no correlation in CHC.
9.Isolation and culture of murine embryonic stem cells of Kunming species

BACKGROUND: Hundreds of mouse embryonic stem cell lines are established presently. Most of these cell lines are collected from 129, C57BL/6J and BALB/C mice. Success rate of creating embryonic stem cell lines with Kunming mice is very low. OBJECTIVE: To explore the method of isolation and culture of murine embryonic stem cells of kunming species to increase the efficiency of establishment of embryonic stem cell lines from Kunming species mouse. DESIGN, TIME AND SETTING: The cell experiment was performed at the Chongqing Key Laboratory of Neurology from April 2006 to June 2007. MATERIALS: Blastocysts of 3.5 days and 4 days from Kunming species mouse were collected. METHODS: The blastocysts of 3.5 days and 4 days from Kunming species mouse were cultured on the fibroblast cell feeder layers for 4-5 days, and the cells from inner cell mass were isolated and subsequently cultured in vitro in 2.5 g/L trypase-0.4 g/L ethylenediamine tetraacetic acid (EDTA) and 1 g/L trypase-0.2 g/L EDTA at room temperature for 2-4 minutes. Embryonic stem cell colony was mechanically straggled. MAIN OUTCOME MEASURES: Colony growth was observed and determined by alkaline phosphatase staining, OCT-4 staining and karyotype analysis. 10.Drug Resistance of Clinical Common Pathogens in Our Hospital in 2008

OBJECTIVE:To analyze the resistance of the main pathogens in our hospital to antimicrobials for clinical reference of rational use of antimicrobial agents. METHODS:The antibiotic resistance of the clinical pathogens in our hospital in 2008 was retrospectively analyzed. RESULTS:Of the total 938 clinical pathogenic bacteria isolated in our hospital,the most common pathogens(leading the first 10 places)were E. coli,staphylococcus aureus,enterobacter spp,staphylococcus epidermidis,pseudomonas aeruginosa,Klebsiella,acinetobacter,enterococcus,hemolytic streptococcus and bacillus proteus. CONCLUSION:Clinicians should use antimicrobial agents rationally to reduce the antibiotic resistance.
