2.GPCR-G? fusion protein and its applications on ligand screening of orphan G protein-coupled receptors

GPCR-G alpha fusion protein has been used to investig ate receptors in recent years. The expression of GPCR-G alpha fusion protein en sured defined 1∶1 stoichiometry, the physical proximity of GPCR to G alpha subu nit, and was suitable for high-throughput ligand screening. The fusion protein technology provided a novel strategy to study orphan GPCRs. Obviously, it will p lay important roles in ligand screening of the orphan GPCRs and exert its signif icances in the R&D of novel drugs targeted at oGPCRs.
3.Experimental study of the effect of baicalin on treatment of lung injury following severe acute pancreatitis

Objective To explore the effects of baicalin on the expression of ICAM-1 in lung injury induced by severe acute pancreatitis(SAP). Methods SAP model was induced by injection of 62mmol/L sodium taurocholate solution into biliopancreatic duct. A total of 72 SD rats were randomly divided into 3 groups: sham operation group, SAP group and SAP treated with BA group. Myeloperoxidase(MPO) activity in lung tissue was examined at 3, 6 and 12h. The expression of ICAM-1 in lung tissue was determined by immunohistochemical technique. The pathological changes of pancreas and lung tissue were examined under optical microscope. Results The levels of MPO activity, which were similar between SAP group and Baicalin group in the early 3 hours, were dramatically lower in Baicalin group at 6h, 12h(P

8.Clinical analysis on 177 patients with measles

Objective To analyze the clinical characteristics,prevention,treatment and prognosis of patients with measles,and provide data for further prevention and complication control.Methods Data of 177 patients with measles were collected and analyzed retrospectively.Results Infants (79.1%) and adults (20.9%) accounted for the most part.The clinical symptoms of measles were typical and Koplik's was very common.The incidences of bronchitis and bronchial pneumonia were higher(33/140,22.6%) in infant patients than that in adults,which was more serious in adult patients,and usually accompanied by liver damage (34/37,89.2%).Conclusion Currently measles is mainly typical,and the incidence in children and adults is increasing.In order to control measles,measles antibody titer monitoring and vaccination of measles should be strengthened.
9.Detection of promoter methylation of p27 gene in gastric carcinoma by methylation-specific PCR technique

ObjectiveTo detect methylation status of the 5'CpG island locating in the promoter region of p27 gene in gastric carcinoma and to analyze the relationship between the aberrant methylaion of p27 gene and the bionomics of gastric carcinoma.MethodsMethylation-specific PCR were used to detect the methylation status of p27 in gastric carcinoma( n =49), adjacent tissue of carcinoma ( n =20) and normal tissue(n =10), respectively.ResultsThe mathylation rate is 49% (24/49) for gastria cancer, 25%(5/20) for tumor-sideward mucosa and 0 (0/10)for normal mucosa.There was a significant positive correlation between gastric carcinoma specimens and normal mucosa specimens ( P < 0. 05 ). There was no significant diference between normal gastric group and tumor-sideward mucosa group ( P > 0. 05 ) and no significant diference between tumor-sideward mucosa group and tumor group ( P > 0. 05 ). The mathylation rate of p27 gene with lymph node metastases (61%) are higher than those without metastasis (28%) (P < 0. 05). And there is no relation on patients' age, sex, tumor size and location.Conclusionp27 gene methylation of the p27 gene is correlated to oncogenesis and metastasis of gastric carcinoma and could lead to the loss of p27mRNA transcription.
10.CT imaging for diagnosis of thoracic sarcoidosis

To investigate the CT findings of thoracic sarcoidosis and improve the understanding and diagnostic level of the disease. Retrospective analysis of the pathology or clinical treatment were observed in 20 patients with thoracic sarcoidosis accorded with the diagnostic standard of CT performance. There were 15 cases confirmed by pathology and 5 cases verified after clinical treatment. There were hilar or mediastinal lymph nodes in 90% cas-es, pulmonary mainly nodules in 60% case, pulmonary fibrosis in 20% cases, bronchovascular bundle thickening in 15%cases, pleural effusion in 10% cases, interlobular septal thickening in 15% cases, ground-glass opacity in 10% cases and pulmonary consolidation in 5% cases. CT findings of thoracic sarcoidosis may help improve the rate of its diagnosis.
