1."Seed cells" for the therapy of liver failure with hepatocyte transplantation
International Journal of Biomedical Engineering 2006;0(06):-
At present, the survival of patients with liver failure mainly depends on orthotopic liver transplantation(OLT), howerer, the clinical use of OLT therapy has been limited because of many practical difficulties. Although the emergence of hepatocyte transplantation offered assist great or substitute for OLT owing to its unique advantages. Finding a best sources of hepatocyte become an pressing problem we are facing. The cells used in present studies include bone marrow stem cells(BSC) , embryonic stem cells (ESC) and umbrilical stem cells (USC), et al. This review summarized progresses on the differentiation of these stem cells into hepatocyte and we hope these basic concepts may benefit a better understanding for ″seed cells″ choice in hepatocyte transplantation.

Tumor microenvironment is the key factor that influences the invasion and metastasis of tumor cells.Exosome is the particle by which cells can make communication in microenvironment.MicroRNA (miRNA)is widely involved in physiological processes, and plays an important role in the stability of tumor environment.MiRNAs in exosome play roles in tumor cell proliferation, extracellular matrix remodeling, and then promote the metastasis, invasion and angiogenesis of tumor. Thus, the expression of oncogenes can be blocked by regulating the exosome production and secretion. It may shed light on the development of a thera-peutic strategy for tumor treatment.

Objective: To investigate the relationship between the Smad4-independent pathway of TGF-?1 and drug-resistance of pancreatic cancer. Methods: The sensitivities of Smad4 homozygous-deleted pancreatic cancer cell line BxPC3 to different kinds of anti-cancer drugs (5-Fu, Gemzar, Oxaliplatin, Cisplatin, CPT-11 and Epirubicin) were observed by MTT assay before and after they were transfected with full-length cDNA of TGF-?1 or treated with TGF-?1 (5 and 10 ng/ml) solution. Western blot was used to detect p170 protein expression after stimulation with different concentrations of TGF-?1. Results: Cisplatin had the most powerful killing effect on BxPC3 cells, followed by Oxaliplatin, 5-Fu and CPT-11 with moderate effect and Gemzar and Epirubicin with the least effect. Cells transfected with full-length cDNA of TGF-?1 or treated with TGF-?1 solution became less sensitive to Cisplatin. Western blot revealed upregulation of p170 expression by TGF-?1. Conclusion: The Smad4-independent pathway of TGF-?1 can increase the drug resistance of pancreatic cancer cells through upregulating expression of p170.

Objective To investigate the correlation between G753A and C1040T polymorphisms in the gene encoding region of thrombin-activatable fibrinolysis inhibitor (TAFI )and cerebral infarction in patients with cerebral infarction in Chinese Han population. Methods C1040T and G753A poly-morphisms in the TAFI gene encoding region in 130 patients with cerebral infarction and 118 healthy subjects (control group)were analyzed retrospectively and they were detected by using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP ). Results The GG genotyping of the TAFI gene G753A polymorphism in the cerebral group was 41. 5%(n=54)and the A allele carriers were 58. 5%(n=76),while those in the control group were 44. 9%(n=53)and 55. 1%(n=65)respectively. There were no significant differences in the GG genotyping of TAFI gene G753A polymorphism and the A allele carriers between the cerebral infarction group and the control group (χ2 =0. 288,P=0. 592). In the cerebral infarction group, the CC genotyping of C1040T polymorphism was 50. 0%(n=65)and T allele carriers were 50. 0%(n=65),while those in the control group were 51. 7%(n=61)and 48. 3%(n=57)respectively. There were no significant differences in the GG genotyping of C1040T polymorphism and the T allele carriers between the two groups (χ2 =0.071,P =0.790 ). Multivariate logistic regression analysis showed that G753A and C1040T single nucleotide polymorphisms (GA or AA genotype)in the TAFI gene encoding region were not the independent risk factors for cerebral infarction. Conclusion There are no significant differences in the correlation between the G753A and C1040T polymorphisms in the TAFI gene encoding region and cerebral infarction. They are not the independent risk factors for the onset of cerebral infarction.

Objective To evaluate the association between the single-nucleotide polymorphism (SNP) of the 5-59A/G (rs2241027) of Htra2 gene and Parkinson's disease in Han population of the western GuangDong province. MethodsThe restriction fragment length polymorphism (PCR-RFLP) was used to determine the 5-59A/G polymorphism in the intron region of Htra2 gene in the case group (n=56) and healthy control group (n=109). ResultsA allele frequency of 5-59 A/G in cases (46.4%) was trended to more than that in controls (36.7%) (P=0.073), as well as the AA genotyping frequency (21.4% vs 11.0%, P=0.072). For the male, the frequency of AA genotype was significantly more in cases (25.7%) than that in controls (10.3%) (P=0.041), and the frequency of A allele was trended to more in cases (48.6%) than in controls (34.6%) (P=0.051). ConclusionA allele and AA genotype of the 5-59A/G (rs2241027) of Htra2 gene may increase the risk of suffering from Parkinson's disease, especially for males.

Chinese Journal of Clinical Nutrition 2022;30(6):340-345
Patients with Crohn's disease (CD) are often complicated with malnutrition and enteral nutrition is the preferred option for nutritional support. Enteral nutrition is the first-line therapy to induce remission of CD in children, but its application in adults has not been well established. In recent years, studies have found that enteral nutrition can not only improve the nutritional status in adult CD patients, but also induce and maintain CD remission through various mechanisms. Given its favorable nutritional efficacy and safety, enteral nutrition has been considered as the basic treatment for adult CD patients and often used in combination with other treatment approaches. However, any combination therapy should be assessed in terms of benefit and risk. This review aims to describe the efficacy and safety of enteral nutrition combined with other treatment approaches in adult CD patients and enumerate common applicable clinical settings, so as to better inform clinical treatment.
