1.IMMUNE RESPONSES OF MICE VACCINATED WITH RECOMBINANTS CODED DIFFERENT STAGE GENES FROM FCC 1/HN STRAIN OF PLASMODIUM FALCIPARUM

Aim To search the effective malaria mixed gene vaccine and to explore the immune response of vaccinated host. The recombinant pcDNA3-EBA175/HRP Ⅱ was injected alone or mixed with pcDNA3-Pfs25 of Plasmodium falciparum into mice by intramuscular route. The muscle of injected parts were treated previously, e.g. injecting 50μl of 0. 5%bupivacaine into the muscle of left latter leg, with 2mm depth. To observed the changes of IgG antibody value, the splenic lymphocyte proliferation, the ratio of CD4+/CD8+ subgroups and NK cell killing activity. After injecting recombinant pcDNA3-EBA175/HRP Ⅱ alone or mixed with pcDNA3-Pfs25 into mice by intramuscular route, it showed that sera IgG value increased, the splenic T lymphocyte proliferation stimulated by specific antigen of Plasmodium falciparum increased, the ratio of CD4+/CD8+ decreased and NK cell activity increased. Enhanced immune injection could improve host's immune reaction.It is suggested intramuscular injection is an effective immune route, and mice inoculated with coding two gene recombinant alone or that mixed with sexual stage gene recombinant could all induce increased humoral and cellular immune response, and NK cell activity.
2.Spiral CT Diagnosis of Small Amounts of Traumatic Suharachnoid Hemorrhage

Objective To improve spiral CT diagnosis and differential diagnosis of small amounts of traumatic subarachnoid hemorrhage(TSAH).Methods SCT features of 105 cases with small amounts of TSAH , which were demonstrated by SCT reexaminations , were retrospectively analyzed. Results The distributions of small amounts of TSAH included cerebral longitudinal fissure cistern in 47 cases, cerebral lateral fissure cistern in 30 cases , cerebral surface sulcus in 22 cases, tentorium of cerebellum in 6 cases. There were 12 cases of small amounts of TSAH with contusion and laceration of brain , Subdural hematoma or bone fracture of skull.Conclusions Spiral CT scan has important value in qualitation and level diagnosisa of small amounts of TSAH.
3.Study on HPLC-DAD-ELSD Fingerprint in Different Species of Dendrobii Caulis

This study was aimed to set up the fingerprint chromatograms of chemical principles of Dendrobii Caulis by HPLC-DAD-ELSD. This analysis was performed at 30°C on a Dikma Cl8 column (250 mm × 4.6 mm, 5 μm) with a mobile phase consisting of acetonitrile-0.5% glacial acetic acid, gradient elution, at the flow rate of 1.0 mL?min-1 with DAD and ELSD detection. The HPLC fingerprint of Dendrobii Caulis was established. And 9 fingerprint peaks were marked. The similarity among different species was low. This method was stable, reliable and repeatable. It is useful for the study on the relationship between chemical components and species of Dendrobii Cauli.
4. Association between hyponatremia and hemodynamic and prognosis in patients with intermediate-risk acute pulmonary embolism

Objective:
To investigate the association between hyponatremia and hemodynamic and prognosis in patients with intermediate-risk acute pulmonary embolism.
Methods:
We retrospectively recruited 110 intermediate-risk acute pulmonary embolism patients (right ventricular dysfunction was confirmed by echocardiography and CT scan with or without the elevated levels of cardiac injury biomarkers) in the first and the second affiliated hospital of Harbin medical university from January 1,2011 to December 31, 2014. The patients were aged (58.4±14.9) years old.There were 49 males and 61 females.Patients were divided into 2 groups as non-hyponatremia group (plasma sodium>135 mmol/L, 93 cases) and hyponatremia group (plasma sodium≤135 mmol/L, 17 cases). Baseline clinical and hemodynamic parameters were obtained from these patients. All enrolled patients were followed up after discharge.

BACKGROUNDOver-expression of RAB5A gene has been proved to be associated with neoplasia metastasis. This study is to explore the effect of RAB5A gene on invasion and metastasis of human lung adenocarcinoma cell lines.
METHODSConstituted basement membrane invasion technique, adhesion capability of tumor cell assay, the chemotactic migration of tumor cells assay, and gelatinases SDS-PAGE analysis method were used to detect the changes of invasive and metastatic capability of Anip973 (with high metastatic capability) and its parent AGZY83-a cell lines (with low metastatic capability).
RESULTSAfter AGZY83-a cells were transfected by PcDNA3.1-RAB5A plasmid, its invasion was significantly increased (t=24.36, P < 0.000 5); adhesion capability of cell was promoted (P < 0.05); the chemotactic migration of cells was higher than that of the parent lines (t=14.18, P < 0.000 5); and the activity of gelatinases secreted from transfected AGZY83-a was enhanced. The invasion of the transfected Anip973 cells with PcDNA3-AntiRAB5A was lower (t= 16.510 4, P < 0.002 5); adhesion capability of cell was decreased (P < 0.05); the chemotactic migration of cells was lower than that of the parent lines (t=6.062, P < 0.005); and the activity of gelatinases was obviously decreased.
CONCLUSIONSThis study in vitro indicates that over-expression of RAB5A genes plays an important role in tumor invasion and metastatic phenotype formation of human lung adenocarcinoma cells; and antisense RNA can interrupt the translation of RAB5A gene.
