1.Preparation and Determination of Indomethacin-hydroxypropy-cyclodextrin Inclusion Complex Ointments

OBJECTIVE: To prepare Indomethacin-hydroxypropy-cyclodextrin inclusion complex ointments and establish its assay method. METHODS: The preparation was prepared with O/W type emulsion ointment as base material, and the content of Indomethacin in which was determined by UV spectrophotometry. RESULTS: The calibration curve of Indomethacin was linear in the range of 16.76~41.91?g?mL-1(r=0.999 6),and its average recovery was 99.53%(RSD=1.50%). CONCLUSION: The ointments were convenient in preparation technique, and simple and accurate in determination.
2.Investigation on teachers' and students' reform expectation of classroom teaching methods for eight-year program medical education

Objective To discuss some expectation on reform of classroom teaching method for eight-year program medical education, in order to provide a basis for the evaluation and reform of classroom teaching method. Methods Investigation that mainly included expectation on reform of classroom teaching method, classroom teaching method of most expected application, and ex-pected value of classroom teaching method was investigated and analyzed with self-made question-naire, to total 78 medical students (22 of Grade 2008, 20 of Grade 2009, 20 of Grade 2010, 16 of Grade 2011) of eight-year program who had completed the stage before medical education in the uni-versity, and related 32 teachers. Software of SPSS 20.0 was used to make statistical analysis. Results In the survey, 55.8% students and 78.1% teachers hoped to reform the current classroom teaching methods (χ2=4.782,P=0.029). 61%students and 90.6% teachers expected comprehensive application of diverse method(χ2=15.464,P=0.017), including instruction combined with writing on blackboard, teaching by multimedia, PBL, inversion-type class, independent learning, a lot of questions for exami-nation, and so on. The value expectation (>60%) of classroom teaching method was multivariate, in-cluding to stimulate students' interest in learning, to cultivate ability of finding and solving problems, conducive to understanding of knowledge and memory, conducive to comprehension and memory of knowledge, conducive to cultivation of learning method, able to improve ability of creativity and criti-cal thinking, conducive to strengthening interaction and communication of teachers and students, to improve the subject consciousness of student in classroom, helpful to cultivate the spirit of teamwork, to improve the efficiency of teaching, and able to make full use of information technology. Conclusions For the diversified value demand and the highly expectation on reform of current classroom teaching method, more attention should be paid to evaluation and guidance on reform of classroom teaching method for eight-year program medical education.
3.Mechanism investigation of activation of ALDH2 against diabetes induced liver injury in rat model

Aim To observe whether the activation of aldehyde dehydrogenase 2 ( ALDH2 ) can protect a-gainst diabetes induced liver injury in rat model, and analyze the role of JNK pathway in the liver protection induced by activation of ALDH2 . Methods All male SD rats were randomly divided into three groups: nor-mal control group ( Con ) , diabetes group ( DM ) and ethanol + diabetes group ( EtOH + DM ) . After 8 weeks, the fasting blood glucose ( FBG) level, glyco-sylated hemoglobin ( HbA1c) level, serum AST and ALT levels were measured. The changes of hepatic pa-thology were observed by hematoxylin and eosin ( HE) staining method. The protein expressions of ALDH2, JNK and p-JNK in liver tissue were measured. Result Compared with control group, in DM group, the lev-els of FBG and HbA1c, serum AST and ALT levels were increased significantly. The structure of liver mor-phology was destroyed, disarranged and unclear, the hepatocyte was swollen, and a large number of inflam-matory cells were infiltrated. ALDH2 protein expres-sion was decreased, while the expressions of JNK, p-JNK and the ratio of p-JNK/JNK were increased. Com-pared with DM group, in EtOH+DM group, the levels of FBG and HbA1c, serum AST and ALT levels were decreased. The expression of ALDH2 protein was in-creased, accompanying with the decrease of JNK, p-JNK protein expressions and the ratio of p-JNK/JNK. Conclusion Activation of ALDH2 can protect the liv-er against diabetes induced liver damage in rat model, which may be relevant with inhibiting the JNK path-way.
4.Oxygen free radical injury of myocardial mitochondria in the experimental type 2 diabetic rats

AIM: To study the mechanism of diabetic cardiomyopathy and abnormality of oxygen free radicals. METHODS: The contents of myocardial cytosolic cytochrome C, mitochondria cytochrome C, mitochondrial calcium, NO, MDA and the activity of SOD and NOS were determined in diabetic rats induced by STZ. The pathological changes were observed under transmission electron microscope. RESULTS: Compared to the normal and ganoderma group, the levels of mitochondrial NO, iNOS, MDA, calcium and plasma Cyt-C in rat myocardium were higher (P<0.05), while mitochondrial Cyt-C and SOD were lowered in model group (P<0.05). The bouncary indistinct, disorganization, a focal loss of muscular fibril, myocardium mitochondria swelling, pulmonary vascular endothelial cellular swelling and obstructed lumen of the capillary were also observed under transmission electronic microscope. CONCLUSION: The findings indicate that oxyradical and lipid peroxidation might be associated with the damage of myocardial mitochondria in NIDDM rats. Cyt-C and mitochondrial calcium is also involved in the process.
5.Curriculum reform of Higher Medical Education based on education outcome: learning outcome framework and competency requirements

The establishment of outcome-based or competency-based education is an important feature of today's curriculum plans. This paper reviews the development and application of outcome-based education (OBE) in medical education. Based on the concept of OBE, the curriculum framework system of Higher Medical Education is reformed and optimized, and the frameworks of course learning outcome, curriculum teaching theme and curriculum competency are redesigned and determined. Outcome-based curriculum reform reflects the requirements of global healthcare education for medical talents in the new era. Driven by the learning outcomes of postgraduates, the curriculum reform focuses on cultivating the postgraduates' recognition, understanding, analysis and research of the phenomena and problem-solving abilities in medical education by providing courses with different teaching topics, clear learning objectives and requirements. The systematic improvement of teaching quality is helpful to further promote the scientific research of medical education and the cultivation of professional talents in medical education management.
6.Expression of ALC1 in tissues of esophageal squamous cell carcinoma and its effect on ESCC cell proliferation, invasion, and migration

Objective: To investigate the expression of ALC1 protein during esophageal squamous cell carcinoma (ESCC) development and progression, so as to explore its association with clinicopathological characteristics and overall survival of ESCC patients, and the effect of ALC1 overexpression on malignant biological behavior of esophageal squamous cells. Methods: Immunohistochemistry (IHC) was used to detect ALC1 protein expression in 245 primary ESCC tissues and their paired normal esophageal mucous membranes, and to determine its correlation to gender, age, tumor cell differentiation, invasion, TNM stage, lymph nodes metastasis, and overall surviv-al rate of ESCC patients. MTT assay, colony formation assay, transwell invasion, and wound healing assay were used to observe the ef-fect of ALC1 on ESCC cell proliferation, invasion, and migration. Results: The expression ratio of ALC1 in esophageal squamous cell car-cinoma was higher compared with that in their paired normal esophageal mucous membranes (41.6% vs . 21.2% , P<0.05). Upregula-tion of ALC1 was associated with ESCC invasion, TNM stage, and lymph node metastasis (P<0.05). The overall survival of ESCC patients with ALC1 overexpression was significantly lower than that in patients with downregulated ALC1 expression (P=0.002). Therefore, ALC1 may promote the proliferation, invasion, and migration of ESCC cells. Conclusions: ALC1 upregulation may play an important role in the progression and development of ESCC. Upregulation of ALC1 leads to poorer disease prognosis, and could promote the prolifera-tion, invasion, and migration of the KYSE30 ESCC cells. Therefore, ALC1 may have potential prognostic value for ESCC patients.
7.Construction of a self-regulated learning structure model for clinical medical undergraduates in the stage of professional courses

The purpose of this study is to investigate the basic situation of self-regulated learning among clinical medical undergraduates in the stage of professional courses and construct a structural model suitable for them to carry out self-regulated learning . Zimmerman's self-regulated learning concept framework was determined as the theoretical basis of this study. A questionnaire survey was performed to identify five main dimensions affecting self-regulated learning including expression of learning intention, self-efficacy, adjustment of learning behavior, management of curriculum strategies, and adaption of learning environment, and then, Amos 24.0 software was used to construct the structural model and draw the conclusion that self-regulated learning in this group centers on the regulation of learning behavior, with learning intention and self-efficacy as sub-centers.
8.LncRNA SATB2-AS1 affects proliferation and apoptosis of cervical cancer cells by modulating miR-373-5p/BTG3 axis

Objective:To explore the effects of long non-coding RNA SATB2-AS1 in the proliferation and apoptosis of cervical cancer cells as an endogenous competitive RNA to regulate miR-373-5p/BTG3 axis.Methods:qRT-PCR was used to detect the expression of LncRNA SATB2-AS1, miR-373-5p and BTG3 in paracancerous tissue and cancerous tissue of patients with CC. The interaction between LncRNA SATB2/miR-373-5p/BTG3 was then predicted and verified. The expression of SATB2-AS1 and miR-373-5p in cells was intervened, subsequently the CC cells were divided into different groups. The proliferation activity of cells in each group was detected by MTT, and the apoptosis of cells in each group was detected by flow cytometry.Results:qRT-PCR showed that the expression of SATB2-AS1 and BTG3 in cancer tissue and CC cells was significantly decreased, while the expression of miR-373-5p was significantly increased in cancer tissue and CC cells compared with paracancerous tissue and normal cervical cells.The targeting relationship between SATB2-AS1 and miR-373-5p was confirmed. Compared with NC group, overexpression of SATB2-AS1 inhibited cell proliferation butinduced apoptosis. Overexpression of miR-373-5p promoted cell proliferation, inhibited apoptosis, but this effect was partially saved by SATB2-AS1.Conclusion:Up-regulation of LncRNA SATB2-AS1 expression regulated the miR-373-5p/BTG3 axis and participated in the progression of cervical cancer, subsequently inhibited cancer cell proliferation but induced apoptosis.
9.Asurvey of career growth recognition amongphysicians at the stage of standardized training

Objective:To investigatethe heterogeneity of career growth recognition amongphysicians at the stage of standardized training and the growth characteristics of clinical physicians in the early stage, and to provide a reference for improving the quality of standardized training.Methods:The trainees who received standardized residency trainingwere randomly selected from three grade A tertiary hospitals in Chongqing, China. Mplus7.4 was used for latent profile analysis to explain the relationship between explicit continuous indicators, and the subtypes of career growth recognition were analyzed.Results:Career growth recognition at the stage of standardized training was classifiedinto three subtypes of all-round growth recognition, high resource and expectation recognition, and unclassified growth recognition, among which the subtype of unclassified growth recognition accounted for the highest percentage of 40.27% (207 trainees).Conclusions:Physicians at the stage of standardized training show obvious uncertainty aboutcareer growth recognition, and it is necessary to strengthen the education on career growth recognition during standardized training and design a development pathway based on the characteristics of career growth recognition.
10.Features of peripheral nerve injuries in workers exposed to vibration: an analysis of 197 cases

Objective To investigate the features of peripheral nerve injuries in workers exposed to vibration.Methods A total of 197 male workers [median age:34 years (21-50 years);median working years of vibration exposure:7.3 years (1-20 years)] engaged in grinding in an enterprise were enrolled.Their clinical data and electromyography results were analyzed to investigate the features of peripheral nerve impairment.Results Of all workers,96 (48.73%) had abnormal electromyography results.Of all workers,88 (44.7%) had simple mild median nerve injury in the wrist,who accounted for 91.7%(88/96) of all workers with abnormal electromyography results.Six workers had ulnar nerve injury,superficial radial nerve injury,or/and superficial peroneal nerve injury and accounted for 6.3% of all workers with abnormal electromyography results.Of all workers,88 had a reduced amplitude of median nerve sensory transduction,and 28 had slowed median nerve sensory transduction.A total of 46 workers were diagnosed with occupational hand-arm vibration disease and hospitalized for treatment.They were followed up for more than 4 months after leaving their jobs,and most of them showed improvements in neural electromyography results and returned to a normal state.Conclusion Workers exposed to vibration have a high incidence rate of nerve injury in the hand,mainly sensory function impairment at the distal end of the median nerve,and all injuries are mild peripheral nerve injuries.After leaving the vibration job and being treated,most workers can achieve improvements and return to a normal state.
