1.Association of serum leptin with the tale of “two hits” in patients with nonalcoholic fatty liver disease

Objective To study the effect of serum leptin in “two hits” of nonalcoholic fatty liver disease (NAFLD).Methods Fasting serum leptin(LEP) ,malondialdehyde(MDA) ,hyaluronic- acid(HA) ,type Ⅲ procollagen ( PCⅢ), laminin ( LN), and type Ⅳ collagen ( Ⅳ-C ) were detected in 43 simple nonalcoholic fatty liver( NAFL ) patients ,41 NASH patients and 40 healthy subjects. Insulin resistance was estimated by HOMA value and its relatio nship with leptin level in NAFLD was analyzed. The severity of lipid peroxidation was estimated by MDA and its relationship with leptin level in NAFLD was analyzed. The relationship of serum fibrosis markers with leptin level in NAFLD also was analyzed. Results Serum leptin, HOMA value in NAFLD were higher than those of healthy controis. MDA,PC Ⅲ 、Ⅳ-C 、LN 、HA in NASH were higher than those of healthy controls or simple NAFL patients. ,HOMA value, in NAFLD positively correlated with leptin. MDA, PC Ⅲ、 Ⅳ-C 、LN、HA in NASH positively correlated with leptin. Conclusion Serum leptin closely correlated with insulin resistance which results in “First hit” of NAFLD. There was certain relationship of leptin with “Second hit” of NAFLD. Leptin should be one of the factors which result in liver fibrosis.

3.Clinical efficacy and safety of Yanhuning in the treatment of children’ s hand-foot-and-mouth disease

Objective To evaluate the clinical efficacy and safety of Yanhuning in the treatment of children ’ s hand -foot -and -mouth disease ( HFMD).Methods One hundred cases with HFMD were ran-domly divided into treatment group ( n=50) and control group (n=50). Patients in the treatment group were given 5-10 mg · kg-1 · d-1 Yanhu-ning +glucose injection 50-100 mL, once a day.Patients in control group were received 10 -15 mg · kg-1 · d -1 ribavirin +glucose injection 100 mL, twice a day.The course of treatment was 7 days for two group.After treatment, the clinical efficacy , clinical symptoms subside time ( herpes disappearance time , temperature returned to normal time, cure time) , alanine aminotransferase ( ALT) , blood urea nitrogen (BUN), creatinine (Cr)at different times (before treatment, after 24, 48 , 72 h ) and adverse drug reactions in two groups were compared.Results After treatment, the total efficiency of treatment group and control group were 96.00% and 84.00%, respectively , showing the better therapeutic effect of the treatment group than the control group.Herpes disappearance time , temperature returned to normal time , healing time of treatment group were shorter than those of control group , there was a significant difference (P<0.05).After treatment 24, 48, 72 h, the ALT, BUN, Cr of treatment group were lower than those of control group , more closer to normal ( P<0.05 ).The incidence of adverse reactions in treatment group and the control group was 12.00%and 16.00%, there was no significant difference ( P>0.05 ).Conclusion The Yanhuning for the treatment of HFMD of children had a good effect , and it can shorten the time of clinical symp-toms and hospitalization time.It can effectively reduce the content of ALT , BUN, Cr and have good effect.
4.Alveolar bone thickness around maxillary central incisors of different inclination assessed with cone-beam computed tomography.

OBJECTIVE: To assess the labial and lingual alveolar bone thickness in adults with maxillary central incisors of different inclination by cone-beam computed tomography (CBCT). METHODS: Ninety maxillary central incisors from 45 patients were divided into three groups based on the maxillary central incisors to palatal plane angle; lingual-inclined, normal, and labial-inclined. Reformatted CBCT images were used to measure the labial and lingual alveolar bone thickness (ABT) at intervals corresponding to every 1/10 of the root length. The sum of labial ABT and lingual ABT at the level of the root apex was used to calculate the total ABT (TABT). The number of teeth exhibiting alveolar fenestration and dehiscence in each group was also tallied. One-way analysis of variance and Tukey's honestly significant difference test were applied for statistical analysis. RESULTS: The labial ABT and TABT values at the root apex in the lingual-inclined group were significantly lower than in the other groups (p < 0.05). Lingual and labial ABT values were very low at the cervical level in the lingual-inclined and normal groups. There was a higher prevalence of alveolar fenestration in the lingual-inclined group. CONCLUSIONS: Lingual-inclined maxillary central incisors have less bone support at the level of the root apex and a greater frequency of alveolar bone defects than normal maxillary central incisors. The bone plate at the marginal level is also very thin.

5.The effect of separating red blood cells from bone marrow graft in vitro by methylcellulose.

It has need to separate red blood cells (RBC) from marrow graft in ABO group unmatched BMT and auto-BMT with purging tumor cells, the separating effect of methylcellulose was observed. The mixture of 0.5% methylcellulose and bone marrow was laid up in an open transfusion system, and then sedimentation of RBC was performed in the transfusion tube. The separating results of 18 marrow grafts showed that the recovery rates of mononuclear cells and CD34(+) cells were (83.8 +/- 55.2)% and (90.3 +/- 7.2)%, respectively. RBC residual rate was (4.3 +/- 1.5)%. The yield of CFU-GM was (60.8 +/- 22.4)/2 x 10(5) MNC, and there was no difference to [(69.8 +/- 23.4)/2 x 10(5) MNC] yielded from same marrow samples, separated by Ficoll-Hypaque separation. It is concluded that this method could be used for bone marrow transplantation.

6.Studies on chemical constituents in rhizome of Elaeagnus bockii I.

OBJECTIVETo study the chemical constituents in the rhizome of Elaeagnus bockii.
METHODCompounds were isolated by silica gel and Sephadex LH-20 chromatography. Their structures were elucidated by means of spectral analysis.
RESULTSeven compounds were isolated from the rhizomes of E. bockii, and their structures were identified as angelicin (1), psoralen (2), vanillic acid (3), bakuchiol (4), oleanolic acid (5), ursolic acid (6) and beta-sitosterol (7 ).
CONCLUSIONAll these compounds were obtained from E. bockii for the first time.

7.Transforming growth factor-β1 and Snail1 mediate tubular epithelial-mesenchymal transition in diabetic rats.

The present study was aimed to explore the expressions of transforming growth factor-β1 (TGF-β1) and Snail1 in renal tissues of diabetic rats, and their role in tubular epithelial-mesenchymal transition (TEMT). Induced diabetic rats were randomly divided into 2-, 4-, 8-, 12-, 16-, 20-, 24-week and 16wA, 20wA, 24wA groups. The rats in 16wA, 20wA and 24wA groups were treated with insulin to control blood glucose to the normal level from the 13th week. The age-matched rats were set as controls. Blood glucose, 24-hour urine protein, serum creatinine (Scr), kidney index of rats were measured. PAS staining was used to observe the renal pathological changes. Immunohistochemical staining and (or) Western blot were employed to determine the expressions of TGF-β1, Snail1, E-cadherin, α-smooth muscle actin (α-SMA) and fibronectin (FN) proteins. The expressions of Snail1 and E-cadherin mRNAs in renal cortex were examined by RT-PCR. Blood glucose, 24-hour urine protein, Scr and kidney index increased remarkably in diabetic rats as compared with those in the control groups (P<0.05, P<0.01) and insulin-treated rats (P<0.01). TGF-β1 and Snail1 protein expressions could not be detected by immunohistochemical staining in the normal renal tissues, however, the strongly positive staining was observed in diabetic rat renal tubules. A time-dependent loss of TGF-β1 and Snail1 expressions was detected in the kidney of insulin-treated rats. In diabetic rats tubular α-SMA positive staining was seen at the 16th week. E-cadherin expression was lost in diabetic rats. The expressions of TGF-β1, Snail1 proteins and Snail1 mRNA were significantly up-regulated in diabetic rats, while down-regulated in insulin-treated rats (P<0.01). The expressions of E-cadherin protein and mRNA in the cortex were contrary to the expressions of TGF-β1 and Snail1. Therefore, TGF-β1 and Snail1 are possibly involved in the pathogenesis of TEMT in diabetic nephropathy rats.

8.The study on methylation of gene IGSF4 promoter in acute leukemia cells.

To study whether gene IGSF4 was inactived by methylation in leukocytic cells, expression of IGSF4 was examined before and after treatment with demethylating agent in U937, Molt4 and HL-60 leukemia cell lines by means of RT-PCR. The methylation of promoter in U937, Molt4 and HL-60 cells as well as 21 acute leukemia patients was analyzed by MS-PCR. The results showed that methylation of IGSF4 promoter was inactived and could be reversed by treatment with a demethylating agent in U937, Molt4 and HL-60 cell. IGSF4 promoter methylation was detected in 57.1% of acute leukemia patients. There is no difference in incidence of IGSF4 promoter methylation between acute myelocytic leukemia and acute lymphocytic leukemia. In conclusion, IGSF4 is frequently inactived in acute leukemia and is a good candidate for the leukemia suppressor gene. As a normal suppressor gene, it may play an important role in inhibiting the development of leukemia, and the methylation of gene IGSF4 may be a good index in monitoring relapse of leukemia.

9.Effect of inhibitors for demethylation and histone deacetylase on proliferation of cell line K562 and expression of tumor related genes.

In order to observe the effect of inhibitors for demethylation and histone deacetylase on the growth of leukemia cell line K562 and the expressin of tumor related genes, the K562 cells were treated with 5-aza-2' deoxycytidine (DAC) and trichostatin A (TSA) in co-culture; the growth curves were observed; the cell cycle was detected by flow cytometry (FCM); the gene expression pattern before and after drug treatment was measured with Atlas7742-1 microarray. The results showed that the combination treatment of DAC and TSA inhibited the proliferation of K562 cells, the growth of most cells were stopped in G(1)/S phases after drug treatment, the gene expression after treatment was more than before, and a few gene expression were down-regulated. In conclusion, combination treatment of DAC and TSA had an inhibitive effect on the leukemia cell line K562, combination of DAC and TSA with microarray could be used for screening candidate genes inhibiting leukemia cells.

10.Studies on chemical constituents of Helwingia chinensis.

OBJECTIVETo investigate the chemical constituents of Helwingia Chinensis.
METHODCompounds were isolated with silica gel, Sephadex LH - 20 and polyamide chromatography, and their structures were elucidated by means of spectral analysis.
RESULTSix compounds were isolated and identified as cinnamic acid (1), gult-5-en-3beta-ol (2), friedelin (3), alpha-amyrin (4), luteolin-7-O-beta-D-glucoside (5) and 4, 5-dimethoxy-1, 2-benzoquinone (6).
CONCLUSIONAll these Compounds were obtained from H. chinensis for the first time.
