Clinical Medicine of China 2008;24(7):648-650
Objective To explore the relationship between P-selection,nitric oxide(NO),high-sensitive-C-reactive protein(hs-CRP)and type 2 diabetes mellitus(T2DM)with macroangiopathy. Methods In sernm of 125 T2DM subjects(60 subjects with macroangiopathy and 65 subjects without macroangiopathy)and 55 normal controls, the activity of P-selection,NO and hs-CRP was detected by ELISA,Nitrate Reductase Assay and scattering immunoturbidimetric method, respectivley. Results The activity of serum P-selection and hs-CRP was significantly higher in diabetic macroangiopathie group than in non macroangiopathic group and control normal.Serum NO was significantly lower in diabetic rnacroanglopathic group than in non macroangiopathic group and control normal.Serum P-selection was positively correlated with hs-CRP-systolic blood pressure(SBP),LDL-C,and was negatively correla- ted with NO:P-selection,hs-CRP,LDL-C and SBP were independently risk factors for T2DM with macroangiopathy. Conclusion Determination of P-selection and NO is very important for prevention and diagnosis of diabetic mac- roangiopathy in early stage.

Objective To investigate the inducible effect of immature dendritic cells (DC) on immune tolerance in coordination islet xenotransplantation. Methods Mature and immature DC were cultured from BALB/c mice with 20 ?g/L GM-CSF and 500 ?g/L GM-CSF+ 200 ?g/L IL-4, respectively, then loaded with major histocompatibility complex (MHC) antigen of Wistar rat. Mature and immature DC were injected into the mice with diabetes. After a week, the islets of Wistar rat or SD rat were transplanted under the envelop of left renal of the diabetes mice. The survival time of grafts was monitored. The immune reactivity of T cell was evaluated by mixed lymphocyte culture. Results Grafts in BALB/c mice pretreated with mature DC rejected quickly, while the survival time of grafts was significantly prolonged in the mice pretreated with immature DC. However, whether pretreated with mature DC or immature DC, grafts from SD rat were rejected soon. The splenocytes of tolerant mice showed mild proliferation. Conclusion The recipient pretreated with immature DC will lead to the anergy of T lymphocyte, and prolong the survival time of xeno-islet.

Objective To investigate whether TGF-?_1 gene modified dendritic cells could induce immune tolerance in islet xenotransplantation. Methods Mature and immature DC were cultured from bone marrow of BALB/C mice, and then transfected the immature DC with TGF?_1 gene. Two kinds of DC were loaded with antigen of islet from Wistar rats, and injected into BALB/C mice with diabetes. After one week, the islets of Wistar or SD rats were transplanted under the capsular of left kidney. Survival time of grafts, proliferation and cytotoxicity of host T cell were evaluated. Results Grafts survival time in normal group was (10.2?1.1)d, (7.0?1.3)d in mature DC pretreated group (P0.05. The proliferation and cytotoxicity of T cell were enhanced in mature DC pretreated group (SI=6.92, KRmax=73%, P

Objective To report the surgical approach for the treatment of giant or extensive skin lesions due to the infection of human papilloma virus (HPV). Method Surgical treatment was performed for 10 patients with verruca vulgris, 20 with condyloma acuminatum, and 8 with epidermodyplasia verruciformis, whose giant or extensive lesions were poorly responsive to routine non-surgical therapy. Results Recurrence of lesions was found in 2 of 10 patients with verruca vulgris and 5 of 20 condyloma acuminatum in 6 months after operation, with a total recurrent rate of 25%. The extensive skin lesions were removed and the functions of affected limbs were recovered in patients with epidermodyplasia verruciformis after operation, however, the long-term topical retinoids was needed to control the recurrence of the lesions. Conclusion Surgical approach is effective for the treatment of the giant or extensive skin lesions due to HPV infection.

Objective To observe the treatment outcome of different surgical strategies in nevus-like benign melanocytic tumors with various size and shape in the head and face. Methods A total of 180 cases with nevus-like benign melanocytic tumors received surgical treatment from February 2005 to February 2008.The following surgical strategies were selected, including direct excision, partial excision, skin grafting, skin soft tissue expanding and application of local flaps (inter-flap, diamond-shaped flap, A-T flap, kite flap, etc),to treat skin lesions in the head, nose, cheek and eyelid. All patients were followed up for 3 to 6 months. Results The incisions for direct and partial excision were well healed without infection or recurrence. Of those cases treated with local flaps or expander, 3 (1 on the face and 2 on the head) experienced recurrence, 2 on the head suffered from post-operational infection. Conclusion To select different surgical strategies according to lesion size, sites and shape may lead to a favorable cosmetic result in the treatment of nevus-like benign melanocytic tumors in the head and face.
