Education of medical morality is an important part of medical education and must be through the whole career life of medical workers.Now our society is in the period of transition,but education of medial morality fails to get enough attention.Based on the present situation,the paper first makes a systematic exposition of the importance,intension,element and process of the reception of medical morality education of medical workers,then points out the rule of this reception and the approach to realize the validity of medical morality education.

Objective To study the transcription level of exogenetic glial cell line-derived neurotrophic factor(GDNF)in rat bone marrow mesenchymal stem cells(BMSCs).Methods The rat BMSCs were isolated and cultured in vitro,and randomly divided into three groups:GDNF-transfected group,blank plasmid-transfected group and non-transfected group.GDNF-transfected group and blank plasmid-transfected group were infected by the recombinant adenovirus of GDNF gene and blank plasmid,respectively.Each of the groups was divided again into 4 subgroups according to the cultured time(3d,6d,9d and 12d)after infection.The transcription level of GDNF gene in BMSCs was tested by quantitative real-time PCR.Results Blank plasmid-transfected group and non-transfected group had no expression of GDNF mRNA,and the relative level of GDNF mRNA transcription was 0.00751?0.00067 in GDNF-transfected group during the following 3~12 days after transfection.Conclusion Gene transfection technoqie mediated by adenoviral vector can transfect GDNF gene into BMSCs with the expression of exogenetic GDNF mRNA.BMSCs transfected with GDNF gene will be used for further research on therapy for the disease of central nervous system.

BACKGROUND:Delayed onset muscle soreness is closely related to skeletal muscle micro-injury, but the exact mechanism of skeletal muscle micro-injury is not yet clear. OBJECTIVE:To observe the histomorphological and ultrastructure changes of skeletal muscle micro-injury models induced by eccentric exercise. METHODS: Forty male Sprague-Dawley rats were randomly divided into quiet control group, immediately after exercise group, post-exercise 24 hours group, post-exercise 48 hours group and post-exercise 72 hours group. In the latter four groups, the rats were subjected to intermittent running on the-16° slope at a speed of 16 m/min: 5 minutes movement, 2 minutes rest and totaly 120 minutes. Rats in the latter four groups were observed immediately, at 24, 48, 72 hours after exercise. RESULTS AND CONCLUSION:After eccentric exercise, the morphology and ultrastructure of rat’s skeletal muscle were both changed to different extents, and Desmin and Vimentin were dyed off for anti-desmin antibody staining at varying degrees. It indicates that one-time eccentric exercise can cause delayed skeletal muscle micro-injury.

Objective To observe the changes of astrocytes and expressions of bcl 2 and bax in neuron apoptosis in facial nerve nucleus after facial nerve transection in rats Methods A total of 72 rats were equally divided into normal control, and facial neurotmesis groups. Six rats were killed 3,7,15,21,30 and 60 d after transection respectively and same amount of rats were killed in same time interval in the control group. Objective To clone CYP3A4 gene and to construct CYP3A4 recombinant mammalian expression vectors which are transferred into K562 cells for expression,and detect the anti-tumor effect of CYP3A4 combined with Cyclophosphamide (CPA) in vitro.Methods Mammalian expression vector containing CYP3A4 gene cloned from human hepatocytes by RT-PCR were constructed and transferred into K562 cells via liposome. The expression of CYP3A4 was detected by RT-PCR and Western blot respectively. MTT detected the anti-tumor effect of CYP3A4 recombinant mammalian expression vectors combined with CPA.Results We cloned CYP3A4 gene and constructed the recombinant mammalian expression vectors pcDNA3, 1/myc-His A (+)-CYP3A4 successfully. Both RT-PCR and Western blot showed significantly higher mRNA and protein expression of CYP3A4 in gene-transfected group than in empty vector-transfected controls and in empty cells controls. The IC_(50) values were 1. 090 16 mmol/L in gene-transfected group, which were significantly lower than in other two groups and could be reduced and increased by the revulsant and inhibitor respectively.Conclusions CYP3A4 recombinant mammalian expression vectors had anti-tumor effect cooperating with CPA, and the effect could be increased and reduced by the revulsant and inhibitor of CYP3A4 respectively.

Objective To investigate the endoluminal ultrasonographie characteristics of adenoearcinoma and mucous adenocareinoma of the rectum. Methods The ultrasonographie characteristics of thirty eases of advanced adenoeareinoma of the rectum and twenty-eight eases of rectal mucous adenoearcinoma were analyzed retrospectively in comparison with their eorresponding pathologic images. Results In advanced adenocareinoma of the rectum, 28 eases manifested rough endoluminal surface, of which 19 marked by thickened changes without arrangement of layers, 10 had indistinct arrangement of layers in some areas and one had clear arrangement of layers. In the deepest margin of infiltration, 17 eases manifested sentus, homed or knot-like protuberance, while 5 marked by wavy margins, and 8 cases had clear and fiat margins. In 25 eases of rectal mucous adenocareinoma, 20 manifested smooth endoluminal surface. With regard to infiltration of tumors to rectal wall, 6 cases manifested clear arrangement of layers, while 13 had clear arrangement of layers in part and 6 had no clear arrangement. In the deepest part of infiltration, 8 eases manifested slippery and fiat margins and 10 manifested wavy margins and 7 had a few sentus, homed or knot-like protuberance locally. According to ultrasonic results of the depth of infiltration to rectal wall in comparison with pathologic findings, 25 eases were diagnosed accurately, while in two cases the diagnosis was too shallow and in three eases it was too deep. In rectal mucous adenocarcinoma, 10 cases got correct diagnosis and 14 were diagnosed superficially and one got too deep. The accuracy of ultrasonic diagnosis on rectal mueoas adenoearcinoma was apparently lower than that of adenoeareinoma and most of the misdiagnosis were too superfieiai. Conclusion There are different ultrasonographie characteristics of rectal adenoeareinoma and rectal mucous adenoeareinoma, which can be helpful for the typing of rectal carcinoma and avoiding excessively superficial misdiagnosis.

Objective To establish a real-time fluorescence quantitative methylation assay to investigate the methylation status of GSH-sulphur-transferase P1(GSTP1) gene promoter region in hepatocellular carcinoma(HCC) and to investigate whether which can be used as the early diagnostic indicator of HCC .Methods Ninety-five serum samples were collected from 40 patients with HCC ,30 patients with liver cirrhosis and 25 individuals with healthy physical examination as controls .The methylation level of GSTP1 gene in these serum samples were quantitatively determined by using the real-time fluorescence quantitative methylated spe-cific PCR technique .The receiver-operation characteristic(ROC) curves were adopted to evaluate its diagnostic value for HCC .Re-sults The methylation quantitative level of GSTP1 gene in HCC serum was significantly higher than that in the healthy controls (P<0 .05) .The ROC curve analysis demonstrated that the methylation quantitative analysis of GSTP1 gene could efficiently distin-guish HCC and cirrhosis from healthy controls (AUC=0 .8641) .With the methylation rate of 2% as the critical value for diagno-sing HCC ,its diagnostic specificity was 87 .5% ,the sensitivity was 69 .6% ;the combination detection of serum GSTP1 gene methy-lation and serum AFP could increase the detection rate of HCC to 75% .Conclusion The real-time fluorescence quantitative methyl-ation assay can accurately quantify the methylation level of serum GSTP1 gene ,which has certain application value for the early di-agnosis of HCC .

ObjectiveTo investigate the relationship between platelet-derived growth factor (PDGF)-B and pre-eclampsia or eclampsia by comparing the expression of PDGF-B mRNA and PDGF-BB protein in the placentas from pregnant women with normal pregnancy,pre-eclampsia or eclampsia.Methods The expression of PDGF-B mRNA and PDGF-BB protein were respectively determined with RT-PCR and immunohistochemistry combined with the analysis of computer microphotography in the placentas from 20 mild pre-eclampsia (mild pre-eclampsia group),20 severe pre-eclampsia (severe pre-eclampsia group),3 eclampsia (eclampsia group) and 20 normal pregnancy (control group).ResultsThe expression of PDGF-B mRNA and PDGF-BB protein in the placentas of mild pre-eclampsia group,severe pre-eclampsia group and eclampsia group ( 1.56 ± 0.30 and 1.32 ± 0.34,1.91 ± 0.28 and 2.00 ± 0.41,2.57 ± 0.06 and 2.73 ± 0.15)were higher than those of control group ( 1.38 ± 0.25 and 1.01 ± 0.26) with significant differences (P＜ 0.01 ).The expression of PDGF-B mRNA and PDGF-BB protein in the placentas of severe pre-eclampsia group and eclampsia group were higher than those of mild pre-eclampsia group (P ＜ 0.01 ).The expression of PDGF-B mRNA and PDGF-BB protein were positively correlated with the degree of the disease (r =0.691,0.797,P ＜0.01 ).ConclusionsPregnant women with pre-eclampsia and eclampsia have higher expression of pDGF-B in the placentas.PDGF-B may play an important role in the development and progression of pre-eclampsia and eclampsia by promoting atherogenesis in the placental vessels.
