2.Effect of Gegenqinlian decoction combined with berberine in the treatment of biguanides resistant type 2 diabetes in elderly patients in glycosylated hemoglobin and homocysteine

Objective To investigate the effects of Gegenqinlian decoction combined with berberine in the treatment of biguanides resistant type 2 diabetes in elderly patients in glycosylated hemoglobin and homocysteine.Methods 82 elderly patients from March 2015 to March 2016 in our hospital who were diagnosed with type 2 diabetes with biguanides resistant were selected,and randomly divided into two groups,41 cases in each groups.Control group receiced routine treatment,experimental group received more with Gegenqinlian Decoction combined with berberine treatment,and all the patients were treated for two weeks.Levels of FPG,2h PG,serum HbA1c,Hcy,hs-CRP,APN,TNF-α,ICAM-1 and clinical efficacy were compared,and the results were analyzed by statistical software SPSS 19 data analysis.ResultsCompared with before treatment,levels of serum FPG,2h PG,HbA1c,Hcy,hs-CRP,TNF-α and ICAM-1decreased in two groups after the treatment(P<0.05),levels of AFN increased(P<0.05),Compared with the control group,levels of serum FPG,2hPG,HbA1c,Hcy,hs-CRP,TNF-α and ICAM-1 in the experiment group were lower(P<0.05),levels of AFN were higher(P<0.05);The total effective rate of the experimental group was significantly higher than the control group(P<0.05).Conclusion Gegenqinlian decoction combined with berberine can effectively reduce the levels of FPG,2h PG,HbA1c,Hcy,hs-CRP,TNF-α in serum and ICAM-1 levels,increase serum APN level,and good curative effect,worthy of clinical application and promotion.
3.Dose comparison of different scan projections of Implagraphy cone beam computed tomography for dental maxillofacial use

Objective To evaluate the subject's absorbed dose,equivalent dose and effective dose.Methods The CBCT unit was Implagraphy and three scan projections were selected such as mandible,maxilla and temporamandibular joint (TMJ).Thermoluminescent dosimeter tubes were used to record the absorbed dose at special positions in the head and neck region of an adult skull and tissue-equivalent phantom.16 interested organs included pituitary,lens,parotid glands,submandibular glands,sublingual glands、diploe,spongy bone of the chin and cervical vertebra,skins of cheeks and nuchal region,thyroid and esophagus.The absorbed dose was measured in these organs,and then the effective dose(E1990,E2007)were calculated according to different ICRP tissue weighting factors.Results The absorbed dose of mandible,maxilla and TMJ scan varied from (0.99 ±0.09) to (12.85 ±0.09)mGy,(0.93 ±0.01) to (13.07 ±0.02) mGy and (0.68 ±0.01) to (10.18 ± 0.04)mGy.There was significant difference among the three scan projections (F =19.61-30992.27,p < 0.05).The equivalent doses of lens and skin were (1.11± 0.07)-(5.76 ± 0.06) mSv and (6.96 ± 0.06)-(10.64 ± 0.07) mSv.There was significant difference among the three scan projections(F =4473.02,9385.50,P <0.05).The effective dose(E1990,E2oo7) was [(191.35±1.53),(325.17 ±2.58) μSv] for mandible scan,[(106.62 ±2.17),(226.28 ±2.81)μSv] for maxilla scan,[(104.21 ± 1.02),(142.36 ± 1.90) μSv]for TMJ scan,respectively.Conclusions The valid measurement should be taken to reduce the subject' s dose such as a careful history and clinical examination before the performance of CBCT,the latest risk/benefit assessment,precise scan position,the shielding of thyroid as well as brain and the smaller volume size as well.
4.DETECTION AND ANALYSIS OF HUMAN PAPILLOMAVIRUS DNA IN CONDYLOMA ACUMINATUM

Condyloma acuminatum is closely associated with human papillomavirus (HPV). In this study the method of slotting hybridization was used to detect the Specimens of 28 cases of condyloma acuminatum. Fourteen out of the 28 samples were positive for HPV 11 probe, the positive rate was 50% (14/28). Nine out of the 14 positive samples were digested with Bamll Ⅰ and Pst Ⅰ respectively to study the present state of DNA after using Southern blotting hybridization. It was shown that HPV 11 DNA were mainly present in a simple monometric episomal state in the condyloma acuminatum cells.
5.SELECTION AND SUBCLONE OF THE TRANSFORMING GENES OF HUMAN PAPILLOMAVIRUS TYPE 16

The transformation of human papillomavirus type 16 (HPV16) may play somc important roles in the oncogenesis of cervical carcinoma Studies showed that the transforming gence mainly located in the E6. E7 open reading frames (ORFs). We digested HPV16 DNA with PstI+EcoRI. Using pUC-19 as the vector, E. coli JM103 strain as the host cell, after two times of directive subelone, plasmid pEP-8 was construeted, confirmed by RE analysis and Southern blot hybridization, the inserted band of pEP-8 was 1.43Kb long contained the intact E6 E7 ORFs, and also a cat-box region, two TATA-boxics and a cell-type specific cnhancer in the upstream swquence. The pEP-8 could be used as the specific probe of HPV-16 E region for identifing virol mRNAs in the HPV infected cells. Meanwhile it was a basework for studying the transformation and transforming proteins of HPV-16.
6.Effect of Earle's solution and MEM on activities of neurons in culture

ObjectiveTo observe the activities of cultured neurons incubated with Earle's solution, new minimal essential medium (MEM) or former MEM.MethodsActivity changes of cultured neurons were measured by determining the leakage of lactate dehydrogenase (LDH) and metabolic rate of 3-(4,5-dimethylthiazol)2,5 diphenyltetrazolium bromide (MTT).ResultsActivities of neurons incubated with different culture medium for 12 h or 24 h were significantly different (P<0.01).ConclusionDifferent culture medium can influence neuronal activities.
7.The protective effect of LY367385 on impairment of cultured mouse cerebral cortical neurons induced by sodium glutamate or oxygen-glucose deprivation

ObjectiveTo investigate the protective effect of LY367385 on impairment of cultured mouse cerebral cortical neurons induced by sodium glutamate (Glu) or oxygen-glucose deprivation (OGD).MethodsNeuron damage induced by Glu or OGD, as well as the action of (S)-(+)-a-amino-4-carboxy-2-methylbenzeneacetic acid (LY367385) were measured by determining the leakage of lactate dehydrogenase (LDH) from neurons. Immunocytochemistry and immunofluorescent methods were used to detect the expression of anti-mGluR1α. Morphological observation of primary cortical neurons was performed by phase contrast microscope.ResultsFollowing the exposure to 0.1 mmol/L Glu for 1 h or OGD for 1 h, LDH leakage from neurons obviously increased (P< 0.01 ). 50 mmol/L LY367385, when co-incubated with Glu or OGD, markedly reduced the LDH leakage (P<0.01). The 24-h leakage of LDH was increased from cells exposed to 0.1 mmol/L Glu for 15 min. Pre-and post-treatment with LY367385 (50 mmol/L ) decreased the leakage of LDH. The cultured neurons expressed mGluR1α.ConclusionLY367385 has protective effect on neurons damaged by Glu or OGD. It may be related to antagonizing mGluR1α.
9.A Scratch-wound Model in Cultured Rat Astrocytes

Objective To reproduce a scratch-wound model in cultured rat astrocytes (AST).Methods The secondary cultured AST prepared from newborn Wistar rat cerebral cortex were scratched with plastic pipette tips. The morphologic change of AST was observed through microscope at 10 min before and 1, 3, 6, 12, and 24 h after injury, meanwhile the lactate dehydrogenase (LDH) leakages in the cultured medium were determined.Results Immediately after injury the edge of the scratch was lined with irregularly shaped cell. 6 h after injury the AST processes began extending to cell-free area, and elongated further at 12 and 24 h after injury, with presented of new generated cells in the denuded area. At different times after injury, the LDH leakages of the experiment group were higher than that before injury ( P<0.05), and were higher than that of the control group ( P<0.05).Conclusion According to observed AST morphologic changes and determined LDH leakages in culture medium, the scratch-wound model in cultured rat AST is successfully reproduced.
10.Thoughts and suggestions on personnel training assessment index in performance appraisal of tertiary public hospitals

Personnel training marks a key assurance for the healthy and sustainable development of healthcare sector and healthcare institutions in China, rendering it critical importance to include personnel training into performance appraisal. The authors analyzed the connotation and orientation of personnel training index assessment in the performance appraisal at tertiary public hospitals in the country, sorted out the assessment details to be optimized. On such basis, the authors recommended on optimal assessment methodology, combining the gradual improvement of medical personnel training assessment indexes and interval range assessment. These efforts aim at providing references for further improving the scientificity, standardization, homogeneity, as well as vertical and horizontal comparability and operability of personnel training index assessment.
