1.Effect of reconstruction on medial collateral ligament healing of knee in rabbits

Objective To provide histological evidence for clinic tendon reconstruction on severe medial collateral ligament injury in rabbits. Methods Medial collateral ligaments of two knee joints were completely incised to create own control model of MCL rupture. The operation group intimated the procedure of human being MCL reconstruction with semitendinous tendon. In the 4th,12th,16th,24th weeks, healing of MCL was observed by anatomy and the picrosirius-polarization method. Results In the 4th week, the MCL healing tissue of ex-periment group is more than control group. The average ratios of type Ⅲ collagen to Ⅰ, Ⅲ collagen in the experiment group [(0.263±0.075), (0.235±0.041), (0.210±0.045), (0.197±0.029)] were significantly lower than those in control group [(0.310±0.072), (0.286±0.045), (0.261±0.046), (0.236±0.043) ] in the 4th, 12th, 16th, 24th weeks, respectively (P < 0.05). Conclusions The semitendinons tendon reconstruction can improve MCL healing in early and middle period after injury.
2.Selective pregnancy reduction in second trimester

Objective To investigate selective pregnancy reduction in second-trimester and its notices. Methods Selective pregnancy reduction in 19 cases of multiple pregnancy during 12~25 gestational age was performed,including 6 cases of twins,1 cases of triplets,1 case of quadruplet and 1 case of sextuplet. Ultrasonographically guided transabdominal intracardiac injection of high concentrate potassium chloride (KCl) solution was used. Results Performances of selective feticide in all twins were successful,except one monochorionic twins. In 11 cases of triplets,one case was aborted,one case was monozygotic triplet. Selective feticide was performed successfully in 7 of the rest 9 cases and the quadruplet Two cases were aborted after the procedure. The sextuplet was aborted after feticide. Conclusion Multifetal pregnancy reduction can be successfully performed in the second trimester without severe maternal complications. But abortion might occur afterwards. It is possible to reduce the abortion rate if monozygotic twin or triplet can be excluded before feticide,choosing the gestational sac which near the fundus,reducing the amniotic fluid volume of the target fetus or postponing feticide.
3.Effect of different pressure oxygen pre-breathe in diving decompression sickness of rats.

OBJECTIVETo investigate the effect of different pressure oxygen pre-breathing in preventing decompression sickness of rats.
METHODSForty male SD rats were randomly divided into 4 groups: decompression sickness (DCS) group and three oxygen pre-breathing groups with 1 ATA, 2 ATA and 3 ATA pressure respectively. The rats of DCS group were placed in the hyperbaric chamber and the chamber was compressed evenly within 3 minutes to depths of 7 absolute atmosphere(ATA) and held at the designated depth for 60 min, then decompressed (3 min) at constant speed to the surface pressure. After that, the rats were taken out for further detection. While the rats of oxygen pretreatment groups pre-breathed different pressure oxygen for 20 min before entering into chamber. The mortality and behavioral of rats were observed with 30 min post decompression. The dry/wet ratio of the lung, protein levels in the bronchoalveolar lavage fluid (BALF), and the inflammatory cytokine tumor necrosis factor (TNF-alpha) expression were also tested.
RESULTSCompared with that of the DCS group, the mortality and morbidity of oxygen pre-breathe groups didn't change obviously. But the total BALF protein level and the inflammatory cytokine TNF-alpha expression of 1 ATA oxygen pre-breathe group were obviously decreased, while the dry/wet ratio of lung as obviously increased instead (P < 0.05).
CONCLUSIONAlthough preoxygenation can' t obviously change the mortality and mobidity of rats, normal pressure oxygen pre-breathing can mitigate the protein infiltration in BALF and the expression of inflammatory cytokine in lung tissue.

4. Studies on chemical constituents of Chloranthus fortunei

Objective: To study the antitumor constituents from Chloranthus fortunei. Methods: Various chromatographic techniques and spectroscopic methods were applied to investigate the chemical constituents from C. fortunei, and some of the compounds were screened for their antitumor activities by MTT method. Results: Sixteen compounds were obtained from the whole plants of C. fortunei and identified as rosmarinic acid (1), 2’-hydroxy-4,3’,4’,6’-tetramethoxychalcone (2), flavokawain A (3), cycloshizukaol A (4), atractylenolide III (5), 4β-hydroxy-8,12-epoxyeudesma-7,11-diene-1,6-dione (6), (8α)-6,8-dihydroxycadina-7 (11),10 (15)-dien-12-oic acid γ-lactone (7), curcolonol (8), 11-hydroxyldrim-8,12-en-14-oic acid (9), friedelin (10), isovanillic acid (11), 6β-hydroxystigmast-4-en-3-one (12), 3,4-dihydroxybenzoic acid (13), shikimic acid (14), scopolin (15) and N-acetyltyramine 1-O-β-D-glucoside (16). Compounds 4 and 5 showed weak cytotoxicity with IC50 ranged from 46 to 85 μmol/L. Conclusion: Compounds 2, 10, 11, and 13-15 are obtained from the genus Chloranthus for the first time and compounds 1-3 and 6-16 are isolated from C. fortunei for the first time. Some sesquiterpenoids from C. fortunei exhibited weak antitumor activities.
5. Chemical constituents from roots of Chloranthus serratus

Objective: To study chemical constituents contained in the ethanol extracts from the roots of Chloranthus serratus. Methods: Fifteen compounds were separated from the roots of M. serratus by using various chromatographic techniques. Results: Their structures were identified on the basis of physicochemical properties and spectral data as 1α, 9α-dihydroxy-8,12-expoxy-eudesma-4,7,11-trien-6-one (1), 1β, 5α-guaiane-4β, 10α-diol-6-one (2), zedoalactone E (3), multistalactone C (4), 1β, 8β-dihydroxy-eudesman-3,7(11)-dien-8α, 12-olide (5), 1β, 8β-dihydroxy-eudesman-4(15), 7(11)-dien-8α, 12 olide (6), sobrerol (7), umbelliferone (8), isofraxidin (9), 5-methoxy-6,7-methylene-dioxy coumarin (10), trans-N-p-coumaroyl tyramine (11), N-trans-feruloyl tyramin (12), N-cis-feruloyl tyramin (13), catechin (14), and 7-hydroxy-5,8-dimethoxyflavanone (15). Conclusion: Compounds 2,5-7,10,14 an 15 are obtained from the plants of Chloranthus Sw. for the first time, and compounds 1,3, and 4 are isolated from C. serratus for the first time.

This research explored the synergistic effects and the potential mechanisms of RCE-4 and various nonsteroidal anti-inflammatory drugs (NSAIDs) on the proliferation of cervical cancer Ca Ski cells. The MTT assay and CalcuSyn V2.0 software were used to detect cell proliferation and calculate the combination index (CI); the expression levels of various proteins were analyzed using Western blot assay; mitochondrial membrane potential (MMP) was assessed using JC-1 staining; acridine orange/ethidium bromide (AO/EB) double-fluorescence staining was used to detect the apoptosis of Ca Ski cells; a co-immunoprecipitation (Co-IP) assay was used to analyze the relative content of Bcl-2-Beclin 1 complex in Ca Ski cells. The results demonstrate that the combination of RCE-4 and NSAIDs increases the inhibition of Ca Ski cells compared to the single-RCE-4 group, and celecoxib provided the best synergistic effect among the four NSAIDs tested, with a CI of 0.32. 7.Pedicle screw fixation into the injured vertebra for thoracolumbar fractures:a long-term effect and biomechanical analysis

BACKGROUND: Pedicle screw fixation is a common repair method for thoracolumbar fractures. Of them, pedicle screw placement into the injured vertebra is an important method. However, the long-term effects and the related biomechanical properties deserve further analysis. OBJECTIVE: To analyze the long-term effect and biomechanical characteristics of the internal fixation of thoracolumbar fractures by pedicle screw implantation. METHODS: A retrospective analysis was performed in 126 cases of thoracolumbar fractures. The patients were treated with pedicle screw implantation and the treatment was fol owed up regularly. Ten specimens of fresh calf chest and lumbar spines were obtained. Five specimens served as an experimental group to prepare models of anterior middle column fractures, and these specimens received pedicle screw fixation into the injured vertebra. The other five specimens served as the control group, and did not receive any treatment. In the two groups, along the sagittal axis, axial compression displacement, anterior flexion displacement, posterior extension displacement, torsion angle and pul -out strength were measured and recorded. RESULTS AND CONCLUSION: (1) Clinical trial results: 126 patients were fol owed up for 10-36 months. Compared with pre-treatment, compression ratio of the injured vertebra, the height of intervertebral space, vertebral slip distance, visual analogue score and Cobb angle were significantly improved at 1 month after treatment and during final fol ow-up (al P < 0.05). No significant difference in above indexed was detected between 1 month and final fol ow-up (al P > 0.05). At the end of fol ow-up, Frankel spinal cord injury grade was significantly improved (Z=-4.127, P < 0.05). After operation, three patients suffered from pulmonary infection, four patients affected urinary tract infection, and one patient experienced delayed wound healing. (2) Biomechanical results: axial compression displacement, anterior flexion displacement, posterior extension displacement, torsion angle and pul -out strength were smal er in the experimental group than in the control group (al P > 0.05). (3) Above findings showed that pedicle screw fixation into the injured vertebra for thoracolumbar fractures obtained good long-term effects, and had good biomechanical properties.
8.The importance of serum IL-1β,TNF-α in diabetic retinopathy patients with hyperuricemia

Objective To detected the levels of inflammatory markers in diabetic retinopathy (DR) patients with hyperuricemia(HUA) ,and to investigated the relationship between diabetic retinopathy and hyperurice-mia .Methods 190 DR patients were enrolled in to our study at January 2016 to June 2017 ,patients were di-vided into group of diabetic retinopathy with hyperuricemia (HUDR group) and group of diabetic retinopathy (DR group) according to level of serum UA ,and selected diabetes mellitus patient (DM group) and health per-son(HC group) as control group .Detected serum interleukin 1β(IL-1β) and tumor necrosis factor-α(TNF-α) levels of participants and statistical analysis .Results The IL-1β and TNF-α levels have significant difference between HUDR group ,DR group ,DM group and HC group ,the difference was statistically significant (P<0 .05) ;Among them ,the levels of IL-1β 、TNF-α in HUDR group was significantly higher than that of DR group ,DM group and HC group ,and the difference was statistically significant (P<0 .05) .The proportion of proliferative retinopathy in group HUDR was 42 .1%,was significantly higher DR group which was 27 .4%, the difference was statistically significant (P=0 .033) .Serum UA level was positively corrected to IL-1β(r=0 .356 ,P=0 .000) and TNF-α(r=0 .660 ,P=0 .000) levels of HUDR patients .Logistic regression analysis re-veal that UA(OR :1 .857 ,95% CI :1 .123-3 .229) ,IL-1β(OR :2 .124 ,95% CI :1 .336-3 .715) and TNF-α(OR :1 .673 ,95% CI :1 .108 -2 .275) were independent risk factors of diabetic retinopathy .Conclusion There are higher levels of IL-1β and TNF-α in diabetic retinopathy patients with hyperuricemia ,hyperuricemia may pro-mote the progression of diabetic retinopathy and hyperuricemia is an independent risk factor of the disease .
9.Identification and Function of Acid-sensing Ion Channels in RAW 264.7 Macrophage Cells

Activation of acid-sensing ion channels (ASICs) plays an important role in neuroinflammation.Macrophage recruitment to the sites of inflammation is an essential step in host defense.ASIC 1 and ASIC3 have been reported to mediate the endocytosis and maturation of bone marrow derived macrophages.However,the expression and inflammation-related functions of ASICs in RAW 264.7 cells,another common macrophage,are still elusive.In the present study,we first demonstrated the presence of ASIC 1,ASIC2a and ASIC3 in RAW 264.7 macrophage cell line by using reverse transcriptase polymerase chain reaction (RT-PCR),Western blotting and immunofluorescence experiments.The non-specific ASICs inhibitor amiloride and specific homomeric ASIC 1 a blocker PcTx 1 reduced the production of iNOS and COX-2 by LPS-induced activating RAW 264.7 cells.Furthermore,not only amiloride but also PcTx 1 inhibited the migration and LPS-induced apoptosis of RAW 264.7 cells.Taken together,our findings suggest that ASICs promote the inflammatory response and apoptosis of RAW 264.7 cells,and ASICs may serve as a potential novel target for immunological disease therapy.
