2.Prognostic value of iASPP for nasopharyngeal carcinoma

Objective To evaluate the prognostic value of iASPP for nasopharyngeal carcinoma (NPC).Methods One hundred and thirty patients with nasopharyngeal carcinoma were initially diagnosed and treated between January and December 2012 in Department of Radiation Oncology of the First Affiliated Hospital of Guangxi Medical University.The clinical staging was classified according to the cancer staging criteria 2009 AJCC/UICC.All patients were treated by IMRT.Cisplatin-based concurrent chemotherapy was given to patients with stages Ⅲ-ⅣB disease.Immunohistochemistry was used to detect the expression of iASPP in the carcinoma tissues,and the clinicopathological features were compared among the patients with different expressions of iASPP.Furthermore,the relationship between the expression of iASPP and the efficacy in patients was explored.Results Of 130 patients,positive expression of iASPP was observed in 86 patients (66.2%),and negative expression in 44 patients (33.8%).There was significant difference in the positive expression rate of iASPP among the patients with different N-stage and clinical stages(x2 =7.565,4.947,P < 0.01).At three months after treatment,no significant difference was found in the response rate of tumor with different expression of iASPP.In univariate analysis,the expression of iASPP was significant predictor of 3 year-DMFS (x2 =4.335,P =0.037) and PFS (x2 =6.640,P =0.01).Furthermore,N-stage was significant predictor of 3y-DMFS (x2 =8.058,P =0.005),PFS (x2 =9.554,P =0.002) and OS (x2 =6.987,P =0.008),respectively.By using multivariate Cox analysis,the expression of iASPP and N-stage was independent prognostic factors for PFS (x2 =4.336,5.228,P < 0.05),respectively.Conclusions Positive expression of iASPP may be a poor prognostic factor for NPC patients.
3.Effects of co-grafts of bone marrow stromal cells and nerve growth factor suspension on repair of spinal cord injury

Objective To investigate effects of co-grafts of bone marrow stromal cells (BMSCs) combined with nerve growth factor (NGF) suspension on repair of spinal cord injury (SCI) in adult rats so as to determine whether the co-grafts are more effective than a single protocol.Methods Spinal cords of adult rats (n=32) were injured by the modified Allen's method.One week after injury,the injured cords were injected with Dubecco-modified Eagles Medium,BMSCs,NGF,and BMSCs plus NGF respectively.One month and two months after injury,rats were respectively sacrificed-and their injured cord tissues were sectioned to identify the transplanted cells under fluorescent microscopy.We observed axonal regeneration and differentiation of BMSCs through immunocytochemical staining conducted with neurofilament (NF) and Neuron-Specific Nuclear Protein (NeuN) and glial fibrillary acidic protein (GFAP).We measured the cavity volume by staining with hematoxylin-eosin (HE).At the same time,rats were subjected to behavioral tests using the open-field BBB (blood brain battier) scoring system.Results One month and two months after transplantation,immunocytochemical staining showed that transplanted cells partially expressed NeuN and GFAP.At the same time,significant reduction in lesion area (P<0.05) and improvement in BBB lo- comotor rating scale (P<0.05) were observed in the cases that received implantation,as compared with those that received vehicle injection.More importantly,further reduction in lesion area and improvement in function were ob- served in the combined treatment group.Conclusions BMSCs can differentiate into neurons and astroeytes. BMSCs and NGF can reduce lesion size,promote axonal regeneration and improve functional recovery,and may have a synergistic effect.
4.Quercetin suppresses HeLa cells by blocking PI3K/Akt pathway.

To explore the effect of quercetin on the proliferation and apoptosis of HeLa cells, HeLa cells were incubated with quercetin at different concentrations. Cell viability was evaluated by MTT assay, cell apoptosis was detected by Annexin-V/PI double labeled cytometry and DNA ladder assay. Cell cycle was flow cytometrically determined and the morphological changes of the cells were observed under a fluorescence microscope after Hoechst 33258 staining and the apoptosis-related proteins in the HeLa cells were assessed by Western blotting. The results showed that quercetin significantly inhibited the growth of HeLa cells and induced obvious apoptosis in vitro in a time- and dose-dependent manner. Moreover, quercetin induced apoptosis of HeLa cells in cell cycle-dependent manner because quercetin could induce arrest of HeLa cells at G0/G1 phase. Quercetin treatment down-regulated the expression of the PI3K and p-Akt. In addition, quercetin could down-regulate expression of bcl-2, up-regulate Bax, but exerted no effect on the overall expression of Akt. We are led to conclude that quercetin induces apoptosis via PI3k/Akt pathways, and quercetin has potential to be used as an anti-tumor agent against human cervix cancer.
5.Comparison of the properties of three denture base materials treated and untreated by hydraulic pressure cooker

Objective:To compare the properties of three kinds of denture base materials treated and untreated by hydraulic pressure cooker. Methods: The impact strength, flexural strength, flexural elastic modulus and color difference roughness of three denture base materials, Kunshan Rijin(KR),GC UNIFAST Trad(GC), Shanghai Zhangjiang(SZ), were measured in vitro. All results were statistically analyzed with SPSS software to have ANOVA and Tamhane's T2 tests, α=0.05. Results: Comparison of the same materials between the treated materials and the untreated materials by hydraulic pressure cooker: there were significant statistical differences of impact strength for all the materials. There were significant statistical differences of color difference for KR and SZ. Comparison of the three materials: impact strength, GC>SZ>KR;flexural strength, KR>SZ>GC;flexural elastic modulus, KR>SZ>GC;roughness, GC>KR>SZ. Conclusion: The properties of denture base materials treated by hydraulic pressure cooker are good.
6.Clinical observation of early vitrectomy for open globe injuries with intraocular foreign bodies

Objective To evaluate the clinical effect of vitrectomy for open globe injuries with intraocular foreign bodies in 48 hours after injury.Methods Retrospective analysis was used to evaluate the patients with open globe injuries (OGIs) accompanied by intraocular foreign bodies (IOFB) who underwent vitrectomy of pars plana vitrectomy (PPV) within 48 hours during the period from January 1,2009 to March 1,2015 in our hospital.Results Among the 56 eyes,16 eyes with intraocular foreign bodies occurred endophthalmitis,while the other 40 eyes did not;the intraocular foreign bodies removal rate was 100%.Among all the patient eyes,52 eyes were saved after surgery,while 4 eyes failed.The pars plana vitrectomy operation ratio of the intraocular foreign bodies patients with endophthalmitis and operated within 24 to 48 hours was 2.09 times than that within 24 hours.There was no significant difference in terms of eye preservation,one time of retinal reposition and abnormal intraocular pressure no matter the PPV surgery was conducted within 24 hours or 24 to 48 hours after injury.Conclusion Early vitrectomy is a safe and effective method for the treatment of open globe injuries with intraocular foreign bodies.

BACKGROUND: At present, the proof of the cerebral protective effects of desflurane on morphology is insufficient and the mechanism is unclear.OBJECTIVE: To investigate the cerebral protective effects of desflurane on complete cerebral ischemia-reperfusion injury and its impacts on gene expression related with apoptosis and stress response.DESIGN: A randomized controlled trial based on the experimental animals.SETTING: Departments of anesthesiology and neurosurgery in a university hospital.MATERIALS: The study was conducted in Beijing Neurosurgery Institute Affiliated with Capital University of Medical Sciences between February 2003and February 2004. Seventeen adult male Wister rats were randomly divided into ischemia group ( n = 7), desflurane group ( n = 7) and sham-operation group ( n = 3 ).INTERVENTIONS: Rat complete cerebral ischemia-reperfusion model was established. Desflurane was immediately inhaled for 1 hour after the beginning of reperfusion in rats of desflurane group. Brains of were harvested from 3 rats of either ischemia group or desflurane group after 1 hour of reperfusion (after 1 hour of operation in sham-operation group) and the cortical ultrastructural changes were observed under electron microscope. The rest 4 rats of either ischemia group or desflurane group were used for the analysis of the variable expression of genes related with apoptosis and stress response by gene chip combined image analysis technique.variable expression of genes related with apoptosis and stress response analyzed by gene chip.RESULTS: Compared with ischemia group, desflurane group had less neuronal pyknosis with almost normal cell organs and ultrastructure in neurons. Compared with ischemia group, the apoptotic protease activating factor 1 (APAF1) of desflurane group down-regulated.CONCLUSION: Desflurane might have protective effects on neurons and cell skeleton and its mechanism might be related with APAF1 down-regulation.
9.Correlation of procalcitonin to the severity of injury, complications and outcome of simple brain trauma patients

Objective To study the changes of serum procalcitonin (PCT) in the patients with simple brain trauma and its relationships with injury of severity, complications and outcome of injury.Methods A total of 120 patients with simple brain trauma were evaluated in a prospective observational study.The serum levels of procalcitonin and C-reactive protein were measured 1,2,3,5,7 and 10 days after the admission.During the following ten days after admission,the clinical data of patients with simple brain trauma were recorded,and also the patients with 28-day survival were followed up.Another 30 healthy subjects were enrolled in normal group for comparison. Results In different periods after trauma,the PCT levels in brain injury patients with different degree of injury severity were significantly different ( P < 0.05 ).During the first two days after admission,there were no significant differences in serum PCT levels between non-infection group and infection group,but significant differences were found between two groups on the 3rd,5th,7th and 10th days ( P < 0.05 ).Kaplan-Meier survival analysis showed the 28 days survival between the group with high PCT level and the group with lower PCT level was statistically significant ( P <0.05 ). Conclusions Poor outcomes and complications such as infection were often occurred in simple brain trauma patients with initially high PCT.The frequent determination of serum PCT levels is a useful approach to the judgment of injury severity and proguosis in patients with simple brain trauma.
10.A retrospective-analysis of the correlation between diabetes mellitus and cancer

Objective To evaluate the influence of diabetes on cancer stage,treatment,and overall survival rate among newly diagnosed cancer patients,and the correlation between diabetes mellitus and cancer.Methods Total 16 890 newly diagnosed cancer patients were analyzed retrospectively.Morbidity rate of diabetes mellitus,stage of cancer,cancer treatment,survival rate,and comorbidities were collected and compared.Results 9.57% of those 16 890 cancer patients were suffering from diabetes by the time of cancer diagnosis.The prevalence of diabetes was high among patients with pancreas cancer (18.76%),renal cancer (16.76%),colorectal cancer (12.34%),and uterine cancer (10.97%).Colorectal cancer was often diagnosed at an advanced tumor stage.Compared with those without diabetes,diabetic patients with colorectal cancer,gastric cancer,and endometrial carcinoma were more likely to receive surgical treatment.Unadjusted analyses showed that the median survival time and percent alive at 3 years in cancer patients with diabetes were significantly reduced in all types of cancers,except for prostate cancer,as compared with those cases without diabetes.After adjustment for occurrence of cardiovascular disease,diabetic patients with colorectal,breast,endometrial,ovary,prostate,kidney,and lung cancers still had a 8%-55% increased risk of mortality compared to cancer patients without diabetes mellitus (P<0.05).Conclusion The prevalane of cancer in diabetes is higher than non-diabetics.Diabetic cancer patients are frequently treated less aggressively and have a worse prognosis compared to those without diabetes.
