1.Quality Analysis on 4 Kinds of Common Oral Antidiabetics in Beijing Drug Market in 2004

OBJECTIVE:To evaluate the quality of antidiabetics in Beijing so as to provide the reference for pharmaceutical manufacturers,patients and governmental departments of drug control.METHODS:236package samples of4kinds of common oral antidiabetics(glibenclamide,gliquidone,metformin,and phenformin)in Beijing medicine market were randomly selected and analyzed.RESULTS:4samples were found to be unqualified,with the disqualification rate of1.7%.CONCLUSION:The quality of4kinds of common oral antidiabetics in Beijing medicine market in2004is satisfactory on the whole.
2.Studies on conditions of tissue culture and accumulation of alkaloids at different period in Fritillaria mellea

Object To compare the contents of alkaloids of callus at different period with tissue culture of fresh bulbs of Fritillaria mellea S. Y. Tang et S. C. Yueh.Methods To study the best condition of induction frequency of callus about some phytohormones, temperature and light, and then to determine the contents of alkaloids of callus in F. mellea. Results The best condition of induction frequency of callus above was NAA 1 mg/L+ 6BA 3 mg/L; temperature at (22?1) ℃ and scattered light; the contents of alkaloids were kept highest in about 20th day ofter the callus appeared.Conclusion The contents of alkaloids of callus are kept higher level than fresh bulbs of F. mellea.
3.Effect of microbubble contrast agent on expression of plasmid EGFP in hepatocellular carcinoma of mice with exposure to low-frequency ultrasound

Objective To explore the optimal dose-effect relationship of gene transfer according to the expression of pEGFP in tumor cells under different parameters mediated by microbubble contrast agent and ultrasound in vivo.Methods C57BL/6J mice were inoculated subcutaneously in the middle of the right flank with hepal-6 tumor cells.The mixture of SonoVue and enhanced green fluorescent protein(EGFP)plasmid was injected into the tail vein of each mouse, groups were randomly established according to different output intensity( 1 W/cm2, 2 W/cm2, 3 W/cm2 ), exposure time ( 1 min, 5 min, 10 min) and volume of SonoVue(30 μl,60 μl,90 μl), the expression of EGFP in tumor cells under different parameter was examined by flow cytometry and fluorescence microscopy.Results The expression of pEGFP in tumor cells was significantly higher with the increase of exposure time,output intensity and volume of SonoVue (1 min vs 5 min, P<0.05; 1 W/cm2 vs 2 W/cm2, P<0.05 ; 30/μl vs 60 μl, P<0.05).However, the transfection efficiency of EGFP can not be enhanced continuously with the increasing exposure time,output intensity and volume of SonoVue (5 min vs 10 min, P>0.05; 2 W/cm2 vs 3 W/cm2, P>0.05; 60 μl vs 90 μl, P>0.05).The highest transfection effieieney was achieved under intensities was 2 W/cm2 [(21.02±1.45)%]with 5 min[(23.22±1.91)%] exposed to ultrasound, and 60 /μl SonoVue [ (21.02±1.45) % ].Conclusions Gene transfeetion efficiency changded with the difference parameters.2 W/cm2,5 min and 60 μl SonoVue led to the optimum dose-effect relationship of gene transfer in vivo .
4.Carotid atherosclerotic plaque predicts poor short-term outcomes of cerebral embolism due to non-valvular atrial fibrillation

Objective To investigate the impact of carotid atherosclerotic plaque on short-term outcomes of cardioembolic stroke due to non-valvular atrial fibrillation (NVAF).Methods A total of 288 patients with acute cerebral embolism due to NVAF were recruited in this study.All patients underwent carotid ultrasonography screening to estimate carotid intima-medium thickness (IMT) and atherosclerotic plaque.The short-term outcomes were assessed.The correlation between carotid atherosclerotic plaque and short-term outcomes of cardioembolic stroke due to NVAF were determined by partial correlation analysis.Results Among the 288 patients,carotid atherosclerosis occurred in 202 cases (70.1%) of the patients,poor outcomes in 113 cases (39.1%),worsening neurological function in 43 cases(14.9%),and stroke recurrence in 24 cases(8.3%).Carotid atherosclerosis plaque was positively associated with neurological worsening (r =0.247,P =0.000) and poor outcomes (r=0.139,P=0.018).The use of aspirin was negatively correlated with both neurological worsening (r=-0.235,P=0.000) and recurrence of stroke (r=-0.177,P=0.003).The use of statin was negatively correlated with recurrence of stroke (r =-0.223,P =0.000),neurological worsening (r=-0.147,P=0.013) and poor outcomes (r=-0.286,P=0.000).Conclusions Carotid atherosclerotic plaque is an independent predictive factor for poor short-term outcomes of cardioembolic stroke due to NVAF,and the aggressive management for carotid plaque can improve the poor short-term outcomes.
5.Analysis of irregular antibody screening results of voluntary blood donors in Shenzhen

Objective To retrospectively analyse the irregular antibodies of the local blood donors ,in order to find the rules ,To ensure the safety and effectiveness of clinical blood transfusion .Methods Collected 169 860 blood donors specimens with the spe‐cific antigen of red blood cells in Shenzhen area from February 2011 to December 2012 ,through Brine method for detection of irreg‐ular antibodies ,positive samples would be identified the specificity of antibodies in blood typing department .Results 169 860 healthy blood donors of irregular antibodies were positive in 36 cases(0 .021% ) ,with 12 cases anti‐M ,4 cases anti‐D ,1 case anti‐P1 ,2 cases anti‐A1 ,1 case of cold antibody ,anti‐M with other irregular antibody in 1 cases ;Male irregular antibody positive rate was 0 .013% (15/118 723) ,female irregular antibody positive rate was 0 .041% (21/51 137) .Conclusion The positive rate of irregular antibody in female blood donors is higher than male blood donors ,and the positive rate of irregular antibody of blood donors is low in this area ,which need to be combined with other irregular antibody detection method to improve the detection rate and ensure the safety and effective of blood transfusion .
6.Effects of Ginger Phenol on IL-17 and IL-23 in Serum of Rheumatoid Arthritis Model Rats

Objective To observe the effects of Chinese medicine ingredient ginger phenol on expressions of IL-17 and IL-23 in collagen-induced arthritis (CIA) rats;To investigate the role and mechanism of ginger phenol in CIA rats.Methods Forty female Wistar rats were randomly divided into 4 groups:blank group, model control group, ginger phenol treatment group and tripterygium glucosides control group. After CIA was established in rats successfully, ginger phenol treatment group and tripterygium glycosides control group were treated with ginger phenol and tripterygium glycosides by gavage, while the control group and model control group were fed with the same amount of saline. Four weeks after the treatment, the rats were collected serum and sacrificed. The pathological of CIA joint synovial was observed by electron microscope. The expressions of IL-17 and IL-23 in serum were detected by ELISA.Results The hyperplasia of synovial tissue and angiogenesis was reduced in gingerol phenol treatment group. Infiltration of inflammatory cells and edema was improved. IL-17 and IL-23 expressions of in serum of CIA rats were significantly higher than those of the blank group (P<0.01). Ginger phenol and tripterygium glycosides down-regulated the expressions of IL-23 and IL-17 (P<0.05,P<0.01). Ginger phenol group was better than that of tripterygium glycosides control group (P<0.05).Conclusion Chinese medicine ingredient ginger phenol can reduce expressions IL-17 and IL-23 in serum of CIA rats.
7.Observation of Dynamic Changes in Ultra-Micro-Structure of Pulmonary Arteries and Endogenous Hydrogen Sulfide in Rats with Left-Right Shunt

Objective To explore the relationship between dynamic changes in ultra-micro-structural of pulmonary arteries and endogenous hydrogen sulfide in rats with left-right shunt.Methods Rats in shunt group were subjected to an abdominal aorta-inferior vena cava shunt to create an animal model of pulmonary artery structural remodeling. After 1 day, 3 days, 1 week, 4 weeks and 8 weeks of experiment, the ultra-micro-morphologic changes of pulmonary arteries of rats were observed under electronic microscope and H_2S concentration in serum was evaluated by modified sulfide electrode method.Results The changes of ultra-micro-structure of pulmonary arteries were progressively exacerbated, endothelial cells became swollen and large in size on 3 days, smooth muscular cells increased in size as well as the change of endothelial cells in 1 week, and they changed from contractile phenotype to synthetic phenotype in 4 weeks.Conclusions Shunt exhibited changes of ultra-micro-structure of pulmonary arteries are accompanied by the changes of endogenous H_2S. It is suggested that endogenous H_2S might play a protective role in changes of ultra-micro-structure of pulmonary artery.

10.Expression of SP and CGRP in esophagus mucosa of nonerusive gastroesophageal reflux disease

Objective To evaluate the expression of substance P (SP) and caltenin gene related peptide (CGRP) in esophagueal mucosa from patients with non-erosive gnstroesophageal reflux disease (NERD) and reflux esophagitis (RE) and to explore their role in the development of NERD. Methods Fif-ty-one patients with typical symptoms of gnstroesophageal reflux disease (GERD) were evaluated with reflux disease questionnaire (RDQ), PPI test, endoscopy and 24hr esophageal pH monitoring. The patients were then divided into RE group (n = 21), NERD group with acid refluux (NERD+, n = 12) and NERD group without acid reflux (NERD-, n = 18) according to the evaluation results. The expression of SP and CGRP in esophagus mucosa from these patients and 10 healthy control subjects were assayed by immunohistochemis-try, and the stain positive index (PI) was calculated by Color patho-image analysis software and compared. Results The PIs of SP and CGRP in NERD- group were 96.77±31.74 and 24.76±29.15, respectively, which were significantly higher than those of NERD+ group (73.64±31.38, 9.78±10.30, respectively, P < 0.05), RE group (67.56±34.62, 9.61±6.20, respectively, P < 0.05) and control group (59.82± 46.15, 8.64±12.12, respectively, P < 0.05). Conclusion Expressions of SP and CGRP in esophagus mucosa from NERD patients without detectable acid reflux are significantly increased, they may play an im-portant role in esophageal visceral sensitivity.
