1.The effects of subthreshold electrical stimulation on apoptosis and caspase-3 expression of ischemic cardiomyocytes

Objective To explore the effects of electrical stimulation below the contraction threshold on apoptosis and the expression of caspase-3 in cardiomyocytes after myocardial infarction in rats.Methods Thirty-two rats were randomly divided into 4 groups:a sham-operated group,a myocardial infarction group,an ischemic electrical stimulation group and a non-ischemic electrical stimulation group.TdT-mediated X-dUTP nick end labeling (TUNEL)was used to investigate apoptosis of cardiomyocytes.Reverse transcription-polymerase chain reaction(RTPCR)and immunohistochemical techniques were used to detect the expression of caspase-3.Results ①Compared with the sham-operated group,the amount of apoptotic cardiomyocytes increased significantly in the two electrical stimulation groups and the ischemic myocardium group.Compared with the ischemic myocardium group,apoptotic cardiomyocytes decreased significantly in the two electrical stimulation groups.There was no significant difference between the two electrical stimulation groups.②Compared with the sham-operated group,in the ischemic myocardium group and the two electrical stimulation groups,expression of caspase-3 increased significantly.Compared with the ischemic myocardium group,expression of caspase-3 decreased significantly in the two electrical stimulation groups.There was again no significant difference between the two electrical stimulation groups.Conclusions Electrical stimulation below the contraction threshold can reduce ischemic cardiomyocyte apoptosis in rats.The mechanisms may be associated with down-regulation of the expression of caspase-3.Electrical stimulation below the contraction threshold of the myocardium can also reduce cardiomyocyte apoptosis in rats.

6.Application of MRI cranial surface locating device in localization of small intracranial space-occupying lesions

Objective To explore the feasibility of applying MRI cranial surface locating device in localization of small intracranial space-occupying lesions before the surgeries. Methods Thirty patients with small intracranial space-occupying lesion were localized by MRI cranial surface locating device before the surgeries. The clinical accuracy was verified during the surgeries and the results were compared with cases without using the device. Results According to the verifications during the surgeries , 30 cases which used MRI cranial surface locating device showed much higher accuracy than that using the traditional method. Conclusions MRI cranial surface locating device can precisely delineate the surgical target area. The device is easy to operate and also can help to design the rational surgical approach with minimum injuries , significantly improve accuracy of locating small intracranial space-occupying lesions and it is safe , effective and worthy of widely used clinically.
7.Value of liver biopsy in diagnosis of chronic hepatitis B

Objective To explore the value of liver biopsy in diagnosis of the severity of chronic hepatitis B (CHB).Methods The liver biopsy, laboratory examination and imaging were performed in 176 CHB patients. The clinical and histological diagnoses were compared. Biochemical indexes of liver function were also compared between patients with histological inflammation activity <G2 and≥G2 or the patients with fibrosis score <S2 and ≥S2. Rank sum test and χ2 test were used in univariate analysis. Binary Logistic regression was used in multivariate analysis. Results Among 176patients, the clinical diagnosis of CHB and cirrhosis were established in 171 patients and five patients,respectively. Among 171 CHB patients, the clinical diagnosis was consistent with histological diagnosis in 102 (59.6%) patients. The clinical diagnosis was more severe than histological diagnosis in 60 (35.1%) patients and the clinical diagnosis was less severe than histological diagnosis in nine (5.3%) patients. Among 44 patients with histological grade G2, 41 (93.2 %) patients had alanine aminotransferase (ALT)≥2 × upper limits of normal (ULN). According multiple Logistic regression analysis, age, overt symptom, ALT level, cholinesterase (CHE), albumin (Alb), total bile acid (TBA) and ultrasonic appearance of non-smooth liver surface were independent risk factors for the diagnosis of fibrosis score ≥S2. Conclusions The CHB diagnosis based on clinical evidences doesn't show acceptable consistency with the diagnosis based on histological evidences. G2 and S2 may be more reliable criteria for diagnosing moderate and severe CHB.
8.Preventive effects of sodium butyrate on gastric epithelial cells damaged by N-methyl-N’-nitro-N-nitrosoguanidine

Objective To investigate the effects of sodium butyrate (SB) on fetal gastric epithelial cells (GECs) in vitro damaged by N-methyl-N’-nitro-N-nitrosoguanidine (MNNG). Methods The fetal gastric epithelial cells were isolated, cultured and identified. Then the cells of the second generation were cocultured with 10 -5 mol/L, 10 -6 mol/L, 10 -7 mol/L SB for 4 h, then added with 10 -5 mol/L MNNG culturing for another 24 h. After the treated cells were finally cultured in normal culture medium for 40 d, the unscheduled DNA synthesis (UDS), lipid peroxidation (LPO) and ras p21 were detected. The cells were only cultured with 10 -5 mol/L MNNG served as negative control and without MNNG or SB as normal control. Results The level of UDS, the contents of lipid peroxidation products and the concentrations of ras p21 in GECs treated with 10 -5 mol/L and 10 -6 mol/L SB decreased obviously, and had significant differences with that of GECs treated without SB. Conclusion SB can effectively prevent the damage of GECs induced from MNNG.

In order to investigate the relationship between the expression of heme oxygenase-2 (HO-2) mRNA and the pathogenesis of Hirschsprung's disease (HD), total ribonucleic acid (RNA) was extracted in the aganglionic and ganglionic segments of colon respectively from 15 cases of HD. The single-stranded cDNA of HO-2 was synthesized and further amplified by reverse transcription-polymerase chain reaction (RT-PCR). The expression of HO-2 mRNA was normal in ganglionic segments, but absent in aganglionic segments. It is concluded that the absence of HO-2 mRNA expression may be an important mechanism responsible for HD.
