1. Effect of Janus kinase inhibition on extracellular signal-regulated protein kinase during invasion and metastasis of human breast cancer cell MDA-MB-231

Objective: To explore the influence of Janus kinase inhibitor AG490 on phosphorylation of extracellular signal-regulated protein kinase (ERK) in the human breast cancer cell line MDA-MB-231, so as to discuss the effect of JAK on ERK signal transduction pathway and on invasion/ metastasis of breast cancer cells. Methods: MDA-MB-231 cells were treated with Janus kinase inhibitor AG490; MTT assay was used to examine the proliferation of MDA-MB-231 cells and the adhesion of MDA-MB-231 cells to artificial basement membrane matrigel after AG490 treatment. Invasion and metastasis of MDA-MB-231 cells were evaluated with transwell chamber after treated with AG490. The expression of P-ERK and MMP-9 protein was determined by Western blotting assay. Results: AG490 inhibited the proliferation of MDA-MB-231 cells in a time- and dose-dependent manner; AG490 also depressed the adhesion, invasion and metastasis of MDA-MB-231 cells. Meanwhile, the expression of P-ERK and MMP-9 protein was decreased after treatment with AG490. Conclusion: Our study indicates that JAK kinase can affect the activity of ERK signal transduction pathway through the phosphorylation of ERK. The inhibitory effects of JAK kinase on MMP-9 expression and invasion of breast cancer cells are associated with the blocking of the ERK signaling pathway.

Factors in uterus may be important in the aetiology of type 2 diabetes.Malnutrition during early life can affect insulin secretion and result in insulin resistance,and such effects are markedly exaggerated when facing over-nutrition in later life.This paper gives a review about the population evidence and epigenetic mechanism in the generational susceptibility to type 2 diabetes.
3.Effect Evaluation of Clinical Pharmacists in Continued Service for Patients with Ischemic Stroke

Objective:To evaluate the effect of clinical pharmacists in the continued therapy of stroke patients .Methods:Totally 146 cases were selected and randomly divided into the observation group and the control group according to the standard matching meth -od.The observation group was treated with the continued therapy .The control group was given one-month telephone follow-up after the discharge.The differences in the drug compliance (Morisky scale), systolic blood pressure (SBP), diastolic blood pressure (DBP), glycosylated hemoglobin (HbA1C), serum total cholesterol ( TC), low density lipoprotein ( LDL-C), health behaviors ( HPLPⅡscale) and relapse rate were compared between the two groups in the 3rd, 6th and 12th month after the discharge.Results:The differ-ences in the above indices were statistically significant between the two groups ( P <0.05) at the various time points , and those were more significant with time extending (P<0.01).The difference of the relapse rate in the 3rd, 6th and 12th month after the discharge between the two groups were significant .Conclusion: Clinical pharmacists can improve the drug compliance and health behaviors of stroke patients and reduce the relapse rate .It is worthy to be recommended in clinics .
4.PROTECTIVE EFFECTS OF JATRORRHIZINE ON MYOCARDIAL ISCHAEMIA AND REPERFUSION DAMAGE IN EXPERIMENTAL ANIMALS

Jat(lOmg/kg iv) administered 5 min prior to ligation of left anterior descending coronary artery significantly decreased the incidence of arrhy-thmias(VT and VF)and mortality of rats during ischaemia and reper-fusion.It also delayed the onset and shortened the duration of arrhythmias. The size of myocardial infarction caused by coronary artery ligation was decreased by Jat ( 0.75mg/kg iv. 1 h before and 6 h after operation) in rabbits.
5.Efficacy of Venlafaxine and Reboxitine in treatment of selective serotonin reuptake inhibitors non-responder with major depressive disorder

Objective The purpose of this study was to access the efficacy of Venlafaxine and Reboxitine on treatment of depressive patients who had not responded to selective serotonin reuptake inhibitors for eight weeks. Methods 117 cases depressive patients who had not responded to selective serotonin reuptake inhibitors for eight weeks were divided into three Venlafaxine group(n =41),Reboxitine group(n =39),Original medicine group(n =37) by randomly and were treated by corresponding drugs for 8 weeks. The therapeutic response and safety were evaluated by scale of HAMD and TESS at before and after treatment the end of 1,2,4,8 week. Results The score of HAMD in Venlafaxine and Reboxitine groups were significantly lower than Original medicine group at the end of 4,8 weeks after treatment and the score of HAMD in Venlafaxine group were significantly lower than Reboxitine group at the end of 8 weeks after treatment. The score of HAMD in Venlafaxine and Original medicine groups were marked descendanter than before treatment from after treatment second weekend(P <0.01),while reboxitine group did not marked descent until the 4th weekend after treatment. The reduce score of factors in cognition,retardition and desperation of Venlafaxine and Reboxitine groups were significantly higher than Original medicine group at the end of treatment. The curative effect of Venlafaxine group were supper than Original medicine group. The side reaction of three groups were not difference. Conclusions There are obviously curative effect using Venlafaxine to treat the depressive patients who had not responded to selective serotonin reuptake inhibitors. The time of initial effect in Venlafaxine were rapider than Reboxitine group.
6.Methylenetetrahydrofolate Reductase Gene Polymorphisms and Ischemic Stroke

Recent studies have demonstrated that hyperhomocysteinemia is an independent risk factor for stroke,which may cause vascular endothelial injury by multiple mechanisms,destroy blood coagulation and fibrinolytic systems,and influence on lipid metabolism.Methylenetetra hydrofolate reductase(MTHFR)is a key enzyme of homocysteine metabolic pathway in the human body.If it is insufficient or its activity decreases,it will directly result in the accumula tion of homocysteine in the human body,and callse hyperhomocysteinemia.Although a number of studies have found that MTHFR gene mutation is the main cause of MTHFR insufficient and its activity decrease,the correlation between MTHFR gene mutation and ischemic cerebral vascular disease is quite controversial now.This article reviews the relationship between MTHFR gene polymorphisms and ischemic stroke.
7.Detection of large deletions in mitochondrial DNA during skin aging

Objective To quantitatively observe the actual levels of 4977 bp and 3895 bp mitochon-drial DNA (mtDNA) deletion in healthy human skin of different age, and to explore their relationship with intrinsic aging and photoaging. Methods Seventy-one samples of skin tissue were obtained from healthy volunteers, including 40 samples from UV-protected areas (buttock, thigh, waist or abdomen) and 31 from UV-exposed areas (neck, back of hands, forehead or face). Nuclear and mitochundrial DNA was extracted from these samples. Conventional PCR was performed to detect the incidence of 4977 bp and 3895 bp mtDNA deletion. Then, SYBR Green real-time PCR was applied to quantitatively analyze the target mutations in posi-tive samples. Results Conventional PCR showed that the incidence of both 4977 bp deletion and 3895 bp deletion increased with age. For example, the incidence of 4977 bp deletion and 3895 bp deletion accounted for 47.5% (19/40) and 30% (12/40), respectively in samples from volunteers older than 40 years, signifi-cantly higher than that in those from volunteers younger than 40 years (X2 = 4.673, 6.118, respectively, both P < 0.05). The total incidence of 4977 bp deletion and 3895 bp deletion in UV-exposed areas was 48.4% (15/31) and 32.3% (9/31), respectively, which did not differ from those in UV-protected areas. The results from real-time PCR revealed a positive correlation of the copy number of 4977 10p deletion and 3895 bp deletion with age (rg = 0.907, 0.845, respectively, both P < 0.05). When the UV-exposed area was compared with the UV-protected area, no significant difference was found in the copy number of 4977 bp deletion ( P = 0.264), whereas a higher level of 3895 bp deletion was noticed in UV-exposed area (P = 0.014). Conclu-sions The 4977 bp mtDNA deletion is primarily associated with chronological aging, and might serve as a biomarker for the process of chronological aging of skin. Deletion of 3895 bp mtDNA seems to be more sus-ceptible to ultraviolet radiation.
