Objective To investigate the expression and clinical significance of RhoC protein in breast cancer.Methods Expression of RhoC protein was detected by immunohistochemlstry in 112 cases of breast cancer,20 cases of adjscent non-cancerous normal tissues and 15 cases of normal mammary gland tissues.The relation between expression of RhoC protein and clinicopathological characteristics was analyzed.Expression of CD34 in breast cancer tissues was detected.Microvessel density(MVD)was calculated and its correlation with RhoC protein Was analyzed.Results RhoC had a positive expression rate of 40.2%(45/112)in breast cancer,while it was not expressed in adjacent non-cancerous tissues or normal mammary gland tissues.Rhoc expression in breast tissues was related to lymph node metastasis and pathological stages(P<0.05).MVD value in patients with positive RhoC expression was significantly higher than that in patients with negative RhoC expression(P<0.05).Conclusions The expression of RhoC protein is related to lymphatic metastasis,pathological stages and MVD value.The upregulation of RhoC protein expression may promote the metastasis and invasion of breast cancer.

Objective To investigate modulatory effects of prostaglancdin E1 on the expressions of NF-?B and cytokines in serum, and to explore the protection mechanisms of PGE1 in ALI.Methods Forty-five male SD rats were randomly divided into three groups: group A(normal group,n=15), group B(LPS group,n=15) and group C(LPS +lipo-PGE1 group,n=15). The pathological changes and activity of NF-?B in the lung tissue, TNF-?, IL-10 and IL-12 concentration in serum at 1 hours,2 hours and 4 hours after intraperitoneal LPS were observed.Results Hyperemia and edema of lung tissue in the LPS +lipo-PGE1 group was obviously less than that in the LPS group. Activity of NF-?B and serum concentrations of TNF-? and IL-12 in the LPS +lipo-PGE1 treated group were more decreased than that in the LPS group, and serum IL-10 concentration was obviously increased in the LPS +lipo-PGE1 group. Conclusion PGE1 reduced LPS-induced ALI through doun-regulating NF-?B activity and inhibiting expression of harmful inflammatory cytokines.

Objective To obserVe the effect of gyPenoside on liPid Peroxidation and hePatic lesion in rats with tyPe 2 diabetes mellitus and nonalcohol fatty liVer disease. Methods Totally,65 SPF male SD rats were randomly diVided into blank control grouP (grouP N),NAFLD model grouP (grouP NM),and NAFLD with T2DM model grouP. The NAFLD with T2DM model grouP was further diVided into three subgrouPs:JH grouP,Perfused with 1 g·kg-1 ·d-1 GPS;JL grouP,Perfused with 0. 5 g·kg-1 ·d-1 GPS;model control grouP,Perfused with the same Volume of water. Blood sugar,triglycerides ( TG) ,total cholesterol ( TC) ,alanine aminotransferase ( ALT) ,asPart aminotransferase ( AST) ,adePonectin ( ADP) in the Plasma were measured. TG, malondialdehyde (MDA),and suPeroxide dismutase (SOD) in the liVer tissue were also tested. Results ADP leVel was (7. 46±1. 12),(3. 58±0. 98),(4. 89±1. 02),(4. 79±1. 01) and (4. 13±0. 89) ng·mL-1 in N,M,NM,JH and JL grouPs, resPectiVely. The ADP leVel was significantly higher in grouP JH and JL than in grouP M (P<0. 01),and significantly higher in grouP JH than in grouP JL (P<0. 05). MDA leVel was (2. 98±0. 09),(4. 22±0. 11),(3. 66±0. 10),(3. 72±0. 11),(3. 99±0. 13) nmol·mL_1 in N,M,NM,JH and JL grouPs,resPectiVely. The MDA leVel was significantly lower in grouP JH and JL than in grouP M (P<0. 01),and significantly lower in grouP JH than in grouP JL (P<0. 05). SOD leVel was (240. 8±17. 4), (149. 9±20. 6),(181. 6±19. 4),(209. 8±19. 2),(189. 4±18. 9) U·mL_1 in N,M,NM,JH,and JL grouPs,resPectiVely. ALT and AST were significantly lower in grouP JH and JL than in grouP M (P<0. 01),and significantly lower in grouP JH than in grouP JL (P<0. 05). Conclusion The liPid Peroxidation in the liVer of rats with T2DM comPlicated with NAFLD can be reduced by gyPenoside,and hePatic lesion may be alleViated through inhibition of liPid Peroxidation.

Objective To compare the specificity and sensitivity of antibodies against SSA and SSB,anti-M3 receptor polypeptide antibodies,anti-α-fodrin(IgG)antibodies in primary SjSgren's syndrome (pSS).Methods One hundred and ten pSS patients(mean age was 49.2±14.8.mean disease duration was 5.6±4.6),80 systemic lupus erythmatosis(SLE)patients(mean age was 25.5 4-4.6,mean disease duration was 2.5±1.2)and 80 rheumatoid arthritis(RA)patients(mean age was 44.6±3.5.mean disease duration was 4.2±1.1)were studied.Enzyme-linked immunosorbent assay was used to measure these antibodies.Results The seropositive rates of anti-SSA,anti-SSB antibodies,anti-M3 receptor polypeptide antibodies and anti-α-fodrin(IgG)antibodies were 45.5%,30.9%,78.2%and 77.3%,respectivelv in pSS.They were much higher than those in RA and SLE patients(P<0.05).Specificities of SSA、SSB antibodies,anti-M3 receptor antibodies and anti-α-fodrin IgG were 83.8%,97.7%.92.0%and 90.O% respectively.With the combination of these antibodies in the diagnosis of pSS.the Sellsitivity can be increased at least to 88.2%and the specificity was not decreased significantly.Conclusion Combination of these antibodies can significantly improve the sensitivity of these antibodies in the diagnosis of pSS.Anti-SSA and SSB antibodies,anti-M3 receptor antibodies and anti-α-fodrin(IgG)antibodies are specific antibodies for the diagnosis of pSS.

[Objective]To simulate the biomechanics mechanism and environment of humeral fracture caused by indirect impact force for the purpose of biomechanics understanding and treatment of such fracture.[Method]Based on the data source, which was high-resolution anatomic slice images from approximal clavicle to distal humerus, 1 mm thickness and totally 380 layers, the geometric model of total shoulder joint was established according to the order:point, line,area, and further meshed to set up the three dimension finite element model of shoulder, fracture sites and instantaneous stress and strain of humerus were simulated and analyzed under the condition which longitudinal impact force was loaded on the humerus based on the 12 functional positions of shoulder(abduction 30?、 45?、 60?、 90?, and simultaneous neutrality, internal rotation 45?,external rotation 45?).[Result]According to the humeral shaft load-strain curve in different functional positions of shoulder, linear relation was found when load changed from 0 N to 250 N, after which non-linear come out, and even load was removed , bone was deformed eternally. With the rise in load amount, the increase in stress was detected. When abduction degree changed from 90? to 30?, the strain of humerus, both the lateral and the medial increased gradually,and increase in internal rotation 45?and external rotation 45? was more significant than that in neutrality. Meanwhile, stress difference could be seen between the lateral and the medial , and medial was larger than the lateral. Increase in stress in rotation positions was quicker and more than that in other functional positions.[Conclusion]Based on 4 abduction degrees (30?, 45?, 60?, 90?) and 3 rotation degrees(neutrality, internal rotation 45?,external rotation 45?) ,the three dimensional finite element shoulder could simulate precisely stress, strain, general trend of fracture line, three dimension images of bone failure. Three dimension finite element simulation and analysis of shoulder is a valuable mechanical method for research on biomechanics theory related to humerus fracture.

Objective: To develop parental punishment style the questionnaire. Methods: Based upon previous literature about parental punishment style and expert interview, data were collected for the parental punishment style questionnaire. A sample of 1028 junior high school students were asked to complete the questionnaire and 158 of them was given retest after a month. Results: Taken from the questionnaire by exploratory factor analysis four factors were compatible. The results of confirmatory factor analysis showed a good fit to the data. The internal consistency reliability of father and mother questionnaire was satisfactory, with Cronbach ? coefficient 0.91; and the test-retest reliability was also satisfactory, with test-retest correlation coefficient 0.88 and 0.89, which showed good content validity and high construct validity. Conclusion: The questionnaire has good reliability and validity, and meets the need of psychometrics.

Objective To investigate oxidative damage effect of the serum of severe preeclamptic patients on human umbilical vein endothelial cells (HUVEC).Methods (1) HUVEC were randomly divided into 4 groups according to the following:blank group as control,normal group added 20％ normal sera of pregnant women,group PE added 20％ sera of severe preeclamptic patients,and group PE + Cat added 20％ sera of severe preeclamptic patients plus 3 x 103 U/ml catalase.After cultured for 24 hours,the injury morphology and APO2.7 expression of HUVEC were detected by transmission electron microscopy and flow cytometry respectively.(2) Under the real-time scanning by laser scanning confocal microscopy,HUVEC were randomly divided into 4 groups according to the following:control group added 100 μmol/L H2O2 as positive control,normal group,group PE,and group PE + Cat.HUVEC of each group was scanned for 120 seconds to determine levels of reactive oxidative species (ROS),calcium homeostasis,and mitochondria membrane potential.Results (1) Obvious injury morphology of HUVEC was observed in group PE,and it was obviously improved by catalase in group PE + Cat.Percentage of HUVEC expressed APO2.7 was (37.8 ± 1.1) ％ in group PE,which was significantly higher than (13.4 ± 1.1) ％ in blank group or (13.5 ± 1.5) ％ in normal group,but significantly lower than (19.2 ± 1.6) ％ in group PE + Cat (all P ＜ 0.01).(2) The fluorescence intensity curves of intracellular ROS and calcium showed slowly rising in group PE,but no obvious changes in normal group and PE + Cat.The values of ROS and calcium in group PE (12.0±1.3,4.1 ±0.7) were higher than those in normal group (1.1 ±0.4,0.6 ±0.4),but lower than those in group PE + Cat (1.5 ± 0.5,0.9 ± 0.5 ; all P ＜ 0.01).Conclusion The serum of severe preeclamptic patients caused oxidative damage on HUVEC by increasing intracellular ROS generation,calcium overload,and decreasing mitochondrial membrane potential.

Objective To evaluate the relationship between the ratio of the maximal value of the H-reflex to the maximal value of the M response (Hmax/Mmax), the ratio of the developmental slope of the H-reflex to the slope of the M response (Hslp/Mslp), and the ratio of the threshold of the H-reflex to the threshold of the M response (Hth/Mth) and upper-extremity spasticity in stroke patients. Methods Upper-extremity spasticity in 34 stroke patients was assessed by modified Ashworth Scale (MAS). H-reflex was recorded in flexor carpi radialis muscle on both sides. Hmax/Mmax, Hslp/Mslp and Hth/Mth in the spastic side were compared to the unaffected side. Results The mean of Hmax/Mmax (spastic side, 0.36±0.17; unaffected side, 0.14±0.07) and Hslp/Mslp (spastic side, 0.72±0.40; unaffected side, 0.26±0.22) showed significant difference between 2 sides (P<0.05). The mean of Hth/Mth (spastic side, 0.93±0.14; unaffected side, 0.98±0.12) showed no significant difference (P>0.05). Hmax/Mmax and Hslp/Mslp were correlated positively with score of MAS in the stroke patients (r=0.608, P<0.05; r=0.917, P<0.05), while there was no correlation between Hth/Mth and score of MAS (r=-0.128, P>0.05). Conclusion Hmax/Mmax and Hslp/Mslp could be sensitive electrophysiologic parameters to evaluate upper-extremity spasticity after stroke.
