BACKGROUND: Placenta mesenchymal stem cells have become hot spots in stem cells study in recent years because of its advantages. OBJECTIVE:To analyze the biological characteristics of amniotic mesenchymal stem cells and amnion epithelial cells, and to explore the feasibility of amniotic mesenchymal stem cells and amnion epithelial cells applied as seed cells in three-dimensional liquid culture to construct the tissue-engineered skin. METHODS:The amniotic mesenchymal stem cells and amnion epithelial cells were obtained by using multi-step digestion with trypsin and col agenase;then the flow cytometry, reverse transcription-PCR and immunofluorescent staining techniques were adopted to identify the surface molecular markers, stem cellcharacteristics and keratinocytes similarity respectively. Based on these data, amniotic mesenchymal stem cells and amnion epithelial cells used as seed cells together with rat type Ⅰ col agen matrix were adopted for three-dimensional liquid culture. RESULTS AND CONCLUSION:Flow cytometry test showed that amniotic mesenchymal stem cells and amnion epithelial cells cultured in vitro could highly express CD90, CD73 and CD105, and could not express the hemopoietic stem cellmarker of CD34 and MHC-class Ⅱ molecular HLA-DR. Reverse transcription-PCR results detected that amniotic mesenchymal stem cells could express stem cellcharacteristic genes CMCY and NANOG;amnion epithelial cells could express stem cellcharacteristic genes CMCY and KLF4, showing that both amniotic mesenchymal stem cells and amnion epithelial cells have stem cellproperties. Reverse transcription-PCR results showed that amniotic mesenchymal stem cells could express keratinocytes characteristic genes K19,β1-integrin and K8;amnion epithelial cells could express K19,β1-integrin, K5 and K8. Immunofluorescence staining results showed amnion epithelial cells could express keratinocytes proliferation related protein K14, which revealed that there was certain similarity in the mRNA expression between keratinocytes and amnion epithelial cells, and indicating that it has the potential to differentiate into keratinocytes. Tissue-engineered skin was successful y constructed by using amniotic mesenchymal stem cells and amnion epithelial cells, hematoxylin-eosin staining section showed that it has certain skin structure, and amnion epithelial cells had a preliminary differentiation. Al these prove that it is feasible to construct human skin tissues with amniotic mesenchymal stem cells and amniotic epithelial cells through the three-dimensional culture.

Objective To analyze the influence of different recall time periods on epidemiological survey of injuries among children and adolescents. Methods A study on injuries for 12 months follow-up period was actively surveyed among 4523 pupils selected by cluster sampling from 12 kindergartens, 1 elementary school, 1 junior high school, 1 senior high school and 1 college in Maanshan city in November, 2007 to November, 2008. Injury events were actively surveyed by school doctors or class health workers simultaneously. Participants were divided into three groups, the first group including self-reported injuries by 3-months interval, the second group with self-recalled injuries by 6-months interval and the third group with self-reported injuries in the past 12 months. Validity and reliability of injuries between surveillance injuries and retrospective injuries were compared. Results The overall rate of injury events was 74.2% and injury events and rate was higher in boys than that in girls (P<0.01) , In total, 6350 injury events were registered for the survey. Compare with surveillance findings, the injury rate declined 0.02% and the number of injuries had a 19 person/time reduction from the retrospective injuries by 3-months interval. The injury rate declined 0.02% and number of injuries reduced 58 person/times from the survey of retrospective injuries by 6-month interval. The rate of injuries declined 8.8% and frequencies had a 505 reduction from data of self-reported injuries in the past 12 months. The largest declines were found for the 3 to 6 year-old age group and for minor injuries. Conclusion The 12-month recall period on underestimate injury rates was compared to the 3-month recall periods and 6-month recall periods. Recall periods of 6 months were recommended to be used in injury survey in children and adolescents.

To explore the method of explants directly induced bud and establish the tissue culture system of mutiple shoot by means of direct organogenesis, core bud and daughter bulbs (the top of bud stem expanded to form daughter bulb) of T. edulis were used as explants and treated with thidiazuron (TDZ) and 1-naphthlcetic acid (NAA). The results showed that the optimal medium for bud inducted form core bud and daughter bulb were MS + TDZ 2.0 mg x L(-1) + NAA 4.0 mg x L(-1) and MS +TDZ 2.0 mg x L(-1) + NAA 2.0 mg x L(-1) respectively, both of them had a bud induction rate of 72.92%, 79.22%. The optimal medium for cluster buds multiplication was MS + TDZ 0.2 mg x L(-1) + NAA 0.2 mg x L(-1), and proliferation coefficient was 2.23. After proliferation, cluster buds rooting occurred on MS medium with IBA 1.0 mg x L(-1) and the rooting rate was 52.6%, three to five seedlings in each plant. Using core bud and daughter bulb of T. edulis, the optimum medium for adventitious bud directly inducted from daughter bulb, core bud and cluster bud multiplication were screened out and the tissue culture system of multiple shoot by means of direct organogenesis was established.

Chinese Journal of Medical Genetics 2003;20(3):211-214
OBJECTIVETo study the association between the polymorphism of human monoamine oxidase type A (MAO-A) gene and Parkinson's disease(PD).
METHODSFnu4HI restriction fragment length polymorphism(RFLP) and PCR-RFLP were used to detect the mutation of MAO-A gene. The frequencies of alleles and genotypes at the MAO-A Fnu4HI locus on the X chromosome in different PD group were compared with those of the control group.
RESULTSIt was found that the frequencies of G allele in the patients with PD and controls were 0.613 and 0.527 respectively, P=0.039 "the frequencies of TT genotype were 0.303 and 0.415(P=0.014), and the frequencies of GG genotype were 0.564 and 0.451 respectively(P=0.021). When the patients were divided into two groups by age-onset, significant difference in the allelic and genotypic frequencies was observed only between early-onset PD group and control group. And when the PD patients were grouped by sex, significant difference was observed only between male PD group and male control group (the frequencies of G allele being 0.669 and 0.500 respectively, P=0.005).
CONCLUSIONThis study revealed significant differences between PD group and control group in allelic and genotypic frequencies. The findings supported the hypothesis about an association between MAO-A gene and PD, suggesting that age at onset of PD and gender predisposition might be related to the putative association, and Fnu4HI SNP be a risk factor for PD.

Objective To examine the epidemiological features and relative risk factors of repeated injuries among middle, high and college students in Ma' anshan city of Anhui province. Methods A prospective study on repeated injuries for 1-year follow-up period was carried out among 1494 students from 3 middle schools or colleges. Risk factors for repeated injuries were analyzed with multivariate logistic regression model. Results 799 students suffered 1639 episodes of injuries, with an incidence rate of 53.48%. The repeated group consisted of 200 students who suffered 854 injuries, accounted for 52.10% among the total injuries. Results from Multi-normial logistic regression analysis showed that male, number of family members of 4 or over, extraversion of character trait, and upper quartile of self-rating anxiety symptoms were the risk factors for the occurrence of repeated injuries. Moderate or well-of family income, mothers with senior high school education, and upper quartile of self-rating depression symptoms were protective factors for repeated injuries. Conclusion Socio-demographic factors and emotional status may predict the occurrence of repeated injuries that called for further study.
