1.Influence of position change on objective indexes and subjective symptoms of patients with single side pulmonary disease

Objective Three common positions have been observed for its influence on the objective indicators of the patients with unilateral lung disease,and the change of subjective symptoms in these patients were collected.Methods The study group consisted of 93 patients who had unilateral lung diseases,each of which was randomly observed for three common positions (healthy side position,affected side position and the horizontal position) for 30 minutes respectively.In these patients,the changes of the objective indictors (heart rate,breathing,blood oxygen saturation,the mean arterial pressure) and subjective symptoms (chest stuffiness,breath is aggravating,etc.) were recorded.Results When using different positions,no statistical difference was observed with the heart rate,breathing,oxygen saturation and mean arterial pressure,whereas when affected side position was taken,the occurrence of worrisome symptoms was obviously higher than the condition when using healthy side position and the horizontal position.There was no statistical difference between the two groups of healthy side position and the horizontal position.Conclusions The occurrence of worrisome symptoms in the case of affected side po-sition is significantly more than the other two positions,indicating that patients with unilateral pulmonary disease are more willing to adopt the healthy side position and the horizontal position.
2.Treatment and prognosis of 70 nasal non-Hodgkin lymphoma patients

Objective To analyze the initial response rate of radiotherapy and chemotherapy,and the influence of different treatment modalities on pmgnnsis of primary non-Hodgkin lymphoma(NHL)of the nascl cavity.Methods From January 1993 to December 2002,70 patients were confirmed by histopathology as NHL,including 52 T cell origin,2 B cell and 16 NK cell.The main radiotherapy portal was set in front of the nose with a spade like protrusion,supplement with a portal next to the ear on one side or both sides.Combined portal in facial cervical area was first used when there was invasion of the oropharynx.The median dose to the nasal cavity wag 54.0 Gy with a range of 336 to 66 Gy,given before or during after radiotherapy or alone,consisted of 2-6 cycles chemotherapy of COP,CHOP or COBDP.Prognostic factors were analyzed with Cox model. Results The local control rates was 12.5%, 66.7% and 74.0% in chemotherapy alone,radiotherapy alone,and chemotherapy plus radiotherapy.The 5-year survival rate Wag 12.5%,50.0% and 62.0%,respectively(P<0.05).Multifaetor analysis showed that tumor extension and treatment methods were the most important prognostic factors besides Ann-Arbor stage,but gender,pathology,age and symptoms had little effect on prognosis.Conclusion Chemotherapy plus radiotherapy group achieved a better survival rate than radiotherapy alone.For some NHL patients with good financial condition,APBSCT is a good choice.
3.c-CSN:Single-cell RNA Sequencing Data Analysis by Conditional Cell-specific Network

The rapid advancement of single-cell technologies has shed new light on the complex mechanisms of cellular heterogeneity.However,compared to bulk RNA sequencing (RNA-seq),single-cell RNA-seq (scRNA-seq) suffers from higher noise and lower coverage,which brings new computational difficulties.Based on statistical independence,cell-specific network (CSN) is able to quantify the overall associations between genes for each cell,yet suffering from a problem of overestimation related to indirect effects.To overcome this problem,we propose the c-CSN method,which can construct the conditional cell-specific network (CCSN) for each cell.c-CSN method can measure the direct associations between genes by eliminating the indirect associations.c-CSN can be used for cell clustering and dimension reduction on a network basis of single cells.Intuitively,each CCSN can be viewed as the transformation from less "reliable" gene expression to more "reliable" gene-gene associations in a cell.Based on CCSN,we further design network flow entropy (NFE) to estimate the differentiation potency of a single cell.A number of scRNA-seq data-sets were used to demonstrate the advantages of our approach.1) One direct association network is generated for one cell.2) Most existing scRNA-seq methods designed for gene expression matrices are also applicable to c-CSN-transformed degree matrices.3) CCSN-based NFE helps resolving the direction of differentiation trajectories by quantifying the potency of each cell.c-CSN is publicly available at https://github.com/LinLi-0909/c-CSN.
4.Intervention effect of apocynin on silicosis induced by silica in rats

Objective:To investigate the intervention effect and its mechanism of apocynin, an inhibitor of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase (NOX) on silicosis induced by silica (SiO 2) in rats. Methods:In October 2021, 24 SPF SD male rats were divided into control group, silicosis model group and apocynin intervention group according to random number table method, with 8 rats in each group. SiO 2 was exposed by one-time intratracheal instillation. The rats in the apocynin intervention group were intraperitoneally injected with apocynin 50 mg/kg, 3 times a week, on the second day after treatment. The rats were sacrificed 28 days later, and lung coefficients were calculated after lung tissues were weighed. Hematoxylin-eosin staining and Masson staining were used to observe the lung histopathological changes in each group, respectively. The levels of NOX, reactive oxygen species (ROS), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) in lung tissue were detected. The expressions of interleukin-1 beta (IL-1β), interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α) were determined by Enzyme-Linked Immunosorbent Assay (ELISA). The level of hydroxyproline (HYP) was detected by alkaline hydrolysate. The expressions of transforming growth factor beta 1 (TGF-β1), E-cadherin (E-cad) and α-smooth muscle actin (α-SMA) in lung tissue were detected by Western blotting. Results:Compared with the control group, the body weight of silicosis model group was decreased, the lung tissue showed obvious inflammatory infiltration and fibrosis, and the levels of lung coefficient, IL-1β, IL-6, TNF-α and TGF-β1 were significantly increased ( P<0.05). Compared with the silicosis model group, the lung tissue injury in the apocynin intervention group was significantly improved, the lung coefficient, NOX, ROS, MDA, IL-1β, IL-6, TNF-α and TGF-β1 levels were decreased, and the activity of GSH-Px was increased ( P<0.05). Compared with the silicosis model group, the expressions of HYP and α-SMA were decreased and the level of E-cad was increased in the apocynin intervention group ( P<0.05) . Conclusion:Apocynin may alleviate SiO 2-induced fibrosis in silicosis rats by reducing oxidative stress, the release of inflammatory factors and inhibiting the process of epithelial-mesenchymal transition.
