1.Effects of Stilaox on Subjective Tinnitus Patients with Sleep Disorders

Objective To study whether stilnox intervention could prove the prognosis for subjective tinnitus patients combined with sleep disorders.Methods By random and double-blind method,68 tinnitus patients with sleep disorders were randomly divided into the treatment group and control group,with 34 cases in each group.The patients in the control group received ginaton and methycobal oral administration.On the basis of this,the patients in the treatment group received Stilnox oral administration before sleep.All objects of study were evaluated using Tinnitus handicap inventory (THI)and Pittsburgh sleep quality index (PSQI)two weeks before and weeks after treatment.Results The tinnitus disability in the treatment group showed statistical differences before and after treatment (t=8.810,<0.05).The levels of sleep disorders in the treatment group showed statistical differences after treatment (t=20.446,<0.05).The tinnitus disability in the control group showed statistical differences after treatment(t=6.900,<0.05).The sleep disorders in the control group showed statistical differences before and af-ter treatment(t=4.631,<0.05).The tinnitus disability (t=2.313,<0.05)and sleep disorders(t=7.906,<0. 05)after treatment showed statistical differences between the treatment group and control group.Conclusion Stil-nox intervention therapy could improve the prognosis of subjective tinnitus patients with sleep disorders.

Galanin is a peptide with wide-ranging effects,especially within the central,peripheral nervous system and endocrine system.Many tumors of neuroendocrine origin,and also an increasing number of non-neuroendocrine cancers,have been shown to express galanin and/or its receptors.Expression of galanin peptide has been detected in pheochromocytoma,squamous cell carcinoma,pituitary adenoma,gastrointestinal cancers and so on.Galanin peptide plays an important role in tumori-genesis and progression of many kinds of tumors.Therefore,studying the roles of galanin and its receptors are helpful for diagnosis and treatment of tumors.
4.Synonymous Codon Usage Bias and Overexpression of a Synthetic Gene Encoding Interferon α2b in Yeast

To achieve higher level expression of Interferon α2b (IFN-α2b) in methylotrophic yeast (Pichia pastoris), a cDNA fragment coding for the mature IFN-α2b was designed and synthesized based on the synonymous codon bias of P. pastoris and optimized G+C content. The synthetic IFN-α2b was inserted into the secreted expression vector pPICZαA, and then integrated into P. pastoris GS115 genome by electroporation. Multi-copy integrants in the Mut+ recombinant P. pastoris strain were screened by high concentrations of Zeocin. 120 hours culturing allowed expression of the IFN-α2b transformant up to 810 mg/L as detected by SDS-PAGE and quantitative methods. In addition, Western blot analysis showed that the recombinant proteins had immunogenicity. The significant antiviral activity of the recombinant IFN-α2b protein was verified by WISH/ VSV system, which was 3.3×105 IU/mL.
5.Proteins structure change of COL4A4 gene point mutation and its association with phenotype in thin basement membrane nephropathy

Objective To detect the proteins structure encoded by COL4A4 gene with different missense mutations of thin basement membrane nephropathy (TBMN) and to analyze the effect of gene mutation on the secondary structure of α4 (Ⅳ) chain and its association with phenotype. Methods A COL4A4-linked TBMN patient with FSGS by a missense mutation (g. 1214G>A resulting in p. G405E) diagnosed by clinical manifestations, family history and renal biopsy examination, as well as two controls (one healthy, one pure TBMN carrying a g. 1550G>A mutation resulting in p. G448S) were enrolled in this study. The fragments of cDNA with the two mutations and that of corresponding cDNA from the healthy control were expressed in E. coll. The secondary structures of recombinant polypeptides were analyzed by circular dichroism (CD) spectroscopy. Results CD spectra of healthy control exhibited a negative peak near 208 nm whereas that of TBMN patient with FSGS exhibited a negative peak near 220 nm. Furthermore, the magnitude of the negative peak of this patient decreased as compared with that of healthy control. CD spectra of pure TBMN control was slightly changed with the negative peak remaining near 208 run and the magnitude slightly decreased as compared with that of healthy control. In addition, the secondary structure of pelypeptide from healthy control was composed of about 1/4 α-helix and 1/4 β-sheet, whereas that from the patient presented about 1/3 α-helix without any β-sheet. The secondary structure of polypeptide from pure TBMN control was almost the same as the healthy control, except a shght reduction of α-helix and a slight increase of β-sheet. Conclusions Although the glycine substitutions exists in the nearby domain of α4 (Ⅳ)chain, the TBMN patient complicating FSGS with severe phenotype and g. 1214G>A mutation and the pure TBMN control with the mild phenotype and g. 1550G>A mutation are revealed with different secondary structures of α4 (Ⅳ)chain. Moreover, the secondary structure change of α4 (Ⅳ) chain is consistent with their corresponding phenotype severity.
6.Cost- effectiveness Analysis of Four Therapeutic Schemes for Hp Infection in Children

OBJECTIVE:To evaluate the economic effectiveness in different pharmacotherapeutic schemes for Hp infection in children.METHODS:To analyze four therapeutic schemes for Hp infection in children with cost - effectiveness analysis.RESULTS: The cost- effectiveness ratios of four therapeutic schemes were 14.92,8.85,8.37 and 8.58 respectively. CONCLUSIONS: scheme C(clarithromycin + bismuth potassium citrate + metronidazole)is the best one.
7.Effects of Artesunate on hepatic fibrosis and its mechanism.

OBJECTIVETo investigate the effects of Artesunate(Art) on the LX-2 cell.
METHODSThe cultured hepatic stellate cells were divided into control group and Art-treated groups with 250,350,450 µmol/L. The rate of cellular proliferation was detected by MIT assay, the content of ceramide (Cer)was determined by HPLC method, the content of hydroxyproline (Hyp) was determined by enzyme digestion method, the expressions of PPAR-γ, p53 and Caspase 3 were detected by Western blot.
RESULTSCompared with control group, IX-2 treated with Art were inhibited in a concentration-dependent manner(P < 0.01). Art could significantly increase the content of cerarnide in LX-2 ( P <0.01), and the content of Hyp was significantly decreased (P <0.05, P <0.01). The expressions of PPAR-γ, p53 and Caspase 3 were increased compared with that of control group(P < 0.01).
CONCLUSIONArtesunate could inhibit the proliferation and induce apoptosis of hepatic stellate cells through upregulating ceramide.

8.Feasibility of in situ eyelid reconstruction in guinea pigs using human umbilical cord jelly

BACKGROUND:Healthy human umbilical cord jel y is a mucous connective tissue without vessels and has been used in eye plastic operation because of its elasticity and toughness. It contains lysozyme, placental globulin, hyaluronic acid enzyme, AMP antibody and complement, and also contains a lot of mesenchymal stem cells, so it is not prone to infection and rejection. OBJECTIVE:To observe the histocompatibility and histopathological changes of human umbilical cord jel y as a tarsal substitute transplanted for eyelid reconstruction in guinea pigs. METHODS:The mucous connective tissue of healthy neonate umbilical cord jel y was made into tissue blocks at even thickness. Models of tarsal defects were established in guinea pigs, and then the mucous connective tissue of healthy neonate umbilical cord jel y was implanted. Samples of implanted materials were col ected for histological examination at 1, 2, 3 weeks postoperation under light microscope. RESULTS AND CONCLUSION:There were no obvious rejection, infection and eyelid deformation observed, the corneas of al the animals were clear, corneal epithelial shedding did not occur, and the eyelid could move normal y. One week after implantation, there was no infection and rejection on the conjunctiva and the incision of the eyelid, the eyelid could move normal y, and partial inflammatory cells were observed between the human umbilical cord jel y and the muscle of the eyelid with microscopy. Two weeks after implantation, there was no infection and rejection on the conjunctiva and the incision of the eyelid, the cornea was clear, the eyelid and eye could move normal y, and no symblepharon occurred;umbilical cord jel y showed the tendency of absorption, and the inflammatory cells were reduced at 2 weeks after implantation. Three weeks after implantation, the incision of the conjunctiva healed wel , the cornea was clear;there was no difference in the eyelid form and movement between the two eyes;the umbilical cord jel y was absorbed partial y, normal fibrous tissues formed and there were no inflammatory cells. With low immunogenicity, human umbilical cord jel y can guide the growth of new col agen and serve as an ideal tarsal substitute.
9.An Analysis of Use of the Antidiabetic Agents

Objective:To find out the current situation of and objectively evaluate the use of the antidiabetic agents so as to provide reference information to the departments of production, sales and consumption of the agents. Methods:A analytic review of 6,639 020 prescriptions during the period of 1995~1999 was made regarding the costs and DDDs. Results: In recent 5 years, the annual increase rate of the expenses for was(36.03±11.92)%; and that of DDDs was 20.05%. The order of expenses for various antidiabetic agents from high to low was as follows: glicalazide, acarbose, glipizide and metformin, while that of DDDs was glicalazide, glipizide, metformin and acarbose. The percentage of expenditure for oral versus the injection antidiabetic agents was(87.08±3.50)% to(12.92±3.50)%. Conclusion: The antidiabetic agents is a kind of common drugs with a bright future of development.
