3.Effects of epidural capsaicin on pain threshold, neurological function and spinal neurons in rabbits

Objective To evaluate the effects of different doses of epidural capsaicin on pain threshold, neurological function and spinal neurons.Methods Thirty-six male New Zealand white rabbits weighing 2.5-3.0 kg were randomly divided into 4 groups (n = 9 each): control group and 3 capsaicin groups (Cap 1, 2, 3). The animals were anesthetized with 10% chloral hydrate 50-75 mg?kg-1 i.v. . An intrathecal catheter was inserted at L6,7 interspace and correct placement was confirmed by outflow of CSF. 1 ml of 0.1 % ( Cap1) , 0.25 % ( Cap2 ) or 0.5% (Cap3 ) capsaicin was injected intrathecally 24 h after IT catheter was placed. Threshold to noxious thermal stimuli was measured and gait of the hind limbs were assessed using Johnson score (5 = normal, 0 = completely paralyzed), before and 1, 3, 7, 10, 15, 20, 25, 30 days after IT injection. On the 2nd day after IT injection 3 animals were killed in each group and the lumbar segment of spinal cord (L6,7) was immediately removed for light and electron microscopic examination. 4.Effects of tobacco extract on the proliferation and HSP70 expression of human periodontal ligament cells

Objective:To study the effects of smokeless tobacco extract(ST)on the proliferation and the heat shock protein 70 (HSP70)expression of human periodontal ligament cells(hPDLCs).Methods:hPDLCs were cultured in vitro and identified by im-munohistochemistry(IHC).The cells were stimulated with ST at 0.01 6 -50 g/L respectively for 24 h,the proliferation was examine by MTT assay,HSP70 expression was detected by immunehistochemical staining and Western Blot.Results:ST inhibited the prolifer-ation and increased HSP70 expression in cytoplasm and nucleus at 0.4 -50 g/L dose dependantly.Conclusion:ST may inhibit the proliferation and increase HSP70 expression of hPDLCs in a dose depandant manner.
5.Protective effects of total of flavone c on cerebral ischemia injury in mice

AIM: To investigate the protective effects of total of flavone C (TFC) on acute cerebral ischemia in mice and focal cerebral ischemia in rats. METHODS: The occlusion of bilateral common carotid arteries with vagus nerves in mice was used for make the acute cerebral ischemia models. The survival time and the death rate were observed. The permanent occlusion of the proximal of the right middle cerebral artery (MCA) was used for make the focal cerebral ischemia models. The extent of neurological deficits was observed, and the infarct area was measured by NBT staining technique. The activity of LDH and the content of MDA and NO in the ischemic cerebral cortex were determined. RESULTS: TFC of 80 and 40 mg?kg -1 prolonged the survival time and decreased the death rate of mice with acute cerebral ischemia injury. TFC of 60, 30, and 15 mg?kg -1 ameliorated neurologic deficits score and the infarct size of rats with MCAO. CONCLUSION: TFC provides significant protective effects against cerebral ischemia injury.
6.Clinical application of He-Ne laser combined with psychological intervention in the treatment of acute her-pes and zoster

Objective The medical code of biology-psychology-societyplays a vital role in modern treatment .The arti-cle aimed to investigate the clinical value of He-Ne laser combined with psychological intervention in the treatment of acute herpes and zoster. Methods Retrospective analysis was made on the clinical data of 81 patients with acute herpes and zoster in our hospital from Jan 2015 to Jan 2016 .The patients were divided into 3 groups randomly .Patients in group A were treated with antiviral and neu-rotrophic medication , psychological intervention and analgesic drugs . Patients in group B were treated with antiviral and neurotrophic medi-cation, psychological intervention and He-Ne laser.Patients in group C were treated with antiviral and neurotrophic medication , psychologi-cal intervention, analgesic drugs and He-Ne laser.Visual analogue scale ( VAS) was evaluated before treatment in hospital on the 3rd day, the 5th day of treatment, and telephone follow -up after 4 weeks of treatment.VAS and average length of stay were analyzed . Results No difference was found as to VAS before treatment in hospital among three groups (8.1 ±1.1, 8.1 ±0.9 vs 7.9 ±1.0, P<0.05).VAS was decreased in all groups after nursing propaganda , psychological intervention and placebo (P<0.05).Compared with the VAS before treatment, VAS in three groups decreased significantly on the 3rd day and the 5th day of treatment.From the re-sults on the 5th day and telephone follow -up after 4 weeks of treatment , VAS was still at a low level in group A and group B , on the contrary, VAS was decreased continuously in group C . Conclusion Combined treatment of antiviral , neurotrophic, analgesic drugs and He-Ne laser can relieve pain , decrease VAS and shorten the average length of stay for patients with acute herpes and zoster .
7.Expressions of progesterone receptor A and B in breast cancer and adjacent non-malignant tissues and significances

Objective To study the expressions of progesterone receptor A (PRA) and B (PRB) in breast cancer and adjacent non-malignant tissues and the correlations between their expressions and the clinical characteristics. Methods The expressions of PRA and PRB in 50 specimens of female human breast cancer and adjacent non-malignant tissues were detected by immunohistochemistry. The correlations between the expressions of PRA and PRB and the clinical characteristics were analyzed.Results PRA and PRB expressed in both the nuclei and the cytoplasma of tumor cells and epithelial cells of the acini and ducts.The percentages of PRA and PRB positive cells were 42%,42% and 52%,36% in the cancer and the adjacent non-malignant tissues,respectively,there was no significant difference.The expression of PRA was significantly correlated with the age of patient(r=-0.316 8,P
8.The Preparation and the Anti-Tumor Targeting Therapy of Adriamycin Long Circulating Temperature-Sensitive Liposome

Objective: To develop adriamycin liposome (AL) , adriamycin long circulating liposome (ALCL) and adriamycin long circulating temprerature-sensitive liposome ( ALTSL) and to study their anti-tumor effects on tumor-bearing mice. Methods: The antitumor activity was observed using the tumor weight as index. The life prolongation rate of mice was calculated according to the tumor-bearing mice survival time. The tissue distribution of adriamycin was determined by HPLC method. Tumor, heart, liver and kidney tissue of the tumor-bearing mice, were sliced and prepared to observe the tissue pathology differences. Results: Compared with free adriamycin, the anti-tumor effects of ALCL and ALTSL were remarkably increased. Their tumor growth inhibitory rates were 57. 8% and 67. 0% respectively. The study of pharmacoki-netics indicated that the adriamycin concentrations were remarkably higher in tumor tissue and blood,lower in heart and lung tissue of ALCL and ALTSL groups when compared with the free ADM group; The pathology slices indicated that tumor cells in the ALTSL group with hyperthermia were mostly destroyed; the cardiac muscle cells in the ALTSL group were similar to the normal cardiac muscle. Conclusion: ALCL and ALTSL remarkably increased the adriamycin concentration on the tumor site, significantly enhanced the anti-tumor effects, decreased the side-effects (such as cytotoxicity) when compared with free ADM, they also significantly prolonged the survival time of the tumor-bearing mice.
9.Protective Effect of Total Flavone of Camellia Against Cerebral Ischemic Injury

Objective To observe the protective effect of total flavone of Camellia (TFC)against cerebral ischemic injury.Methods Decapitation method and close hypoxia method were used to observe the effect of TFC on anoxic tolerance of mice and step down test was used to observe the effect of TFC on learning and memory; after ischemia, the content of malondialdehyde (MDA)and nitric oxide (NO)and the activity of lactate dehydrogenase (LDH)were detected. Rat models with acute incompletely cerebral ischemia were established by means of ligating right common carotid arteries and effect of TFC on cerebral water volume, permeability of cerebral vessels and cerebral histopathological changes were also observed.Results TFC prolonged the grasping time after decapitation and the survival time after anoxia in mice, and improve the learning and memory during the step down test. TFC decreased MDA and NO contents, counteracted the de creases of LDH activities in the mice cerebral cortex, reduced the water volume and permeability of cerebral vessels in ischemic rats and improve the cerebral hitstopathological changes. Conclusion TFC has protective effects against cerebral ischemic injury and the mechanism may be related to the inhibition of free radicals and NO production.

Objective To establish human lung adenocarcinoma multidrug resistance cell lines in vitro,observe their biological characteristics,and investigate the mRNA expressions of DNA pol?,mdr 1,mrp1,GST-?,lrp and topo Ⅱ genes.Methods Paclitaxel-resistant cell lines(A549/TXL20) were established in vitro by exposure to stepwise increased concentrations of the drug in a cell culture medium.Biological morphology and cell cycles were analyzed by morphometry and flow cytometry.The chemoresistance indexes of cells were measured by methyl tetrazolium assay.Evaluation of growth and in vitro drug sensitivity were performed.RT-PCR was employed to analyze the mRNA expressions of the DNA pol?,mdr 1,mrp1,GST-?,lrp,and topo Ⅱ genes.Results ① Compared with parent cells,the resistant sublines had a lower confluent density.They were smaller and mixed with giant cells in different sizes and with different numbers of nucleoli,and the growth property of A549/TXL20 did not change significantly compared with A549 cell lines.② The resistant cells,A549/TXL20,were 19.3 times more resistant to paclitaxel and 67.4 times more resistant to cisplatin than the parent cells,and also demonstrated cross-resistance to mitomycin,vinblastine,and 5-fluouracil(5-FU). ③ Compared with the A549 celllines,an unreasonably higher level of drug resistance and lower drug concentration was detected in A549/TXL20 cells after exposure to the drug in the culture medium.④ The mRNA expression level of DNA pol?,mdr1,GST-?,mrp1 andlrp genes in A549/TXL20 cells was significantly higher than that in A549 cell lines(P
