2.Effects of berberine hydrochloride on secretion of interleukin-6 from human periodontal ligament cells cultured in vitro

		                        			Objective To observe the effects of berberine hydrochloride on the secretion of interleukin-6(IL-6)from human periodontal ligament cells(PDLCs)cultured in vitro.Methods Periodontal ligament tissues were isolated from extracted human premolars,and PDLCs were cultured in vitro.After identification by immunohistochemistry technique,human PDLCs were divided into LPS group and NLPS group according to whether lipopolysaccharide(LPS,50 ?g/mL)was contained in cell culture media.Then in accordance with the final concentrations of berberine hydrochloride in cell culture media(0,0.010,0.020,and 0.030 g/L),the groups were subdivided into LPS and NLPS control group,LPS1 and NLPS1 group,LPS2 and NLPS2 group,LPS3 and NLPS3 group.IL-6 contents in supernatants of each group were determined by enzyme-linked immunosorbent assay,and statistical analysis and comparison were conducted as well.Results Compared with corresponding LPS and NLPS groups,IL-6 contents in supernatants of LPS3 group and NLPS3 group reduced markedly(P
		                        		
		                        		
		                        		
		                        	
3.Fanconi anemia protein and acute myeloid leukemia

		                        			Fanconi anemia (FA) is an rare autosomal recessive inherited disease which manifests progressive marrow failure, congenital bone malformation, high risk to cancers and so on. Chromatin of FA cells display auto-instability and high hypersensitivity to interstrand DNA cross-links such as mitomycin C. As normally FA develop into acute myeloid leukemia easily, it has been regarded as pre-leukemia state. Till now 11 FA genes have been found and play a role in sustaining stability of gene groups through the same mechanism. As an active connecting protein, FANCF protein play an important part in correct FA complex formation. Which makes FANCD2 single ubiquitin. Ubiquitin FANCD2 induces chromatin and BRCA1 interact, and repair injured DNA. 4.Effect observation on heat-sensitive moxibustion for cervical spondylosis of vertebral artery type

		                        			Objective:To observe the difference of the therapeutic effect between heat-sensitive moxibustion and warm needling therapy for cervical spondylosis of vertebral artery type (CSA). 
 Methods:A total of 60 cases with CSA who met the inclusion criteria were randomly divided into a heat-sensitive moxibustion (HSM) group and a warm needling therapy (WNT) group according to the visiting sequences, with 30 cases in each group. Patients in the former group were treated by heat-sensitive moxibustion, while patients in the latter group were treated by warm needling therapy. Both groups were treated once every other day, with 10 times as 1 course of treatment, and the therapeutic effects were evaluated after 1 course of treatment. 
 Results:After 1 course of treatment, the total effective rate of the HSM group was 93.3%, versus 83.3% in the WNT group. There was a statistically significant difference between the two groups (P<0.05). 
 Conclusion:Heat-sensitive moxibustion has better therapeutic effect for CSA than warm needling therapy.
		                        		
		                        		
		                        		
		                        	
5.Operation parameters of ultrafiltration separation of protein from earthworm homogenate

		                        			AIM: The aim of this investigation was to confirm the best operation conditions in the membrane separation of protein from the homogenate of red earthworm, Eisenia faefida. METHODS: Ultrafiltration membrane of fresh red earthworm with the cut-off of 10 000 was used as the target process, with the homogenate flux as index, the best value of operation parameters such as pressure、 temperature, surface rotation speed and feed concentration were fixed on. RESULTS: (1) The best suitable operation conditions were 0.25 MPa, 30?C , 200 r/min, pH=6.7, 33.3% (w:w) concentration and not a bit of NaCl. (2) The molecular weight distribution of protein in the homogenate of red earthworm mainly lay between 50 000~100 000. 		                        			Epidermal growth factor receptor (EGFR) is found to express at high levels in a variety of solid tumors including gliomas. This study was to examine the effect of an EGFR-tyrosine kinase inhibitor (AG1478) alone or in combination with cisplatin (CDDP) on the growth of glioma cells (U87). U87 glioma cells were treated with AG1478 (10 μmol/L) or CDDP (25 μmol/L) as a single agent or in combination for 24 or 48 h. The expression of EGFR and the components in its downstream signaling pathway [extracellular signal-regulated kinase (ERK), protein kinase B (AKT)] in U87 glioma cells was detected by Western blotting. Cell growth, cell cycle distribution and cell apoptosis were determined by MTT method and flow cytometry, respectively. The results showed that CDDP could induce the activation of EGFR and the components in its downstream signaling pathways in a concentration-dependent manner. The combined treatment of AG1478 with CDDP could inhibit the proliferation of U87 glioma cells, arrest the cell cycle and promote cell apoptosis. In the EGFR signaling pathway, AG1478 decreased the phosphorylation of ERK, AKT and EGFR in U87 glioma cells. It was concluded that the combined treatment of AG1478 and CDDP may exert synergistic inhibitory effects on the growth of glioma cells by suppressing the activities of EGFR, AKT and ERK.
		                        		
		                        		
		                        		
		                        	
8.Construction of genetic linkage map of Bupleurum chinense DC. using ISSR and SSR markers.

		                        			Abstract: Molecular genetic map is a fundamental organizational tool for genomic research. However, a genetic linkage map for Bupleurum chinense DC. has not been developed. In this study, with the theory of pseudo-testcross, 96 F1 plants from an intraspecific cross of B. chinense were used as mapping populations. Twenty eight ISSR (inter-simple sequence repeat) primers and 44 SSR (simple sequence repeat) primers were used to detect the polymorphisms between the parental plants, and of them, 28 ISSRs and 14 SSRs were selected to analyze the F1 populations. The map consisted of 13 linkage groups which included 80 (72 ISSRs and 8 SSRs) loci, and covered 2 633.9 cM with an average density of 33.4 cM. All 13 linkage groups consisted of 2-31 loci ranging in length from 15.4-1295.7 cM. This map will provide a basis for studies on gene mapping, map-based cloning and maker-assisted selection of important traits in B. chinense.
		                        		
		                        		
		                        		
		                        	
9.Time-dependent changes of facet joint asymmetry in patients with lumbar disc herniation

		                        			Objective To discuss the time-dependent changes of facet joint asymmetry in patients with lumbar disc herniation (LDH). Methods A total of 54 patients with LDH were prospectively analyzed. CT was used to judge whether there existed small joint facet asymmetry and to measure the facet joints. The present results were compared with the results that would be investigated two years later. Results The sagittal process of small facet plane accounted the majority in patients with LDH after two years, with no statistical difference compared with the results before two years (P > 0.05). During 2-year period, the facet joint asymmetry disappeared in seven patients, while the facet joint asymmetry occurred in other seven patients, with no statistical difference (P > 0.05). Conclusions Facet plane and the asymmetry will change with time. Observation of the relationship between facet joint asymmetry and LDH should be carried out in a period of time rather than at a time point.
		                        		
		                        		
		                        		
		                        	
10.Application of magnetic resonance spectroscopy in diagnosing DNS

		                        			Objkctive To determine the potential ability of the area of each peak for the proton magnetic resonance spectroscopy(1 H-MRS) in the diagnosis of DNS(Delayed neuropsychological sequelae after carbon monoxide poisoning).Methods MRI and STEAM(stimulated-echo-acquisition mode)were performed in 38 DNS patients and 18 healthy volunteers.The area of the peak of each organic compound were obtained in each volume of interest.Results The data spectra abtained from patients shows NAA(881±76),Cho(2 352 ±86),β、γGlx(3 024 ± 187).Compared with the control group,theβ、γGlx ratio and Cho area increased,but the NAA ratio decreased.The area of NAA,Cho,β、γGlx between DNS and control group were compared,the results showed significant differences (P <0.05).In spectroscopy,the Lip evaluated peak in 0.9~1.40 ppm were detected.Conclusion 1H-MRS may do some contribution to the diagnosis of DNS.
