2.Enhancement of the epitope vaccines from new gene wx2b4a of Toxoplasma gondii potency using pidotimod in mice

Objective To observe the effect of pidotimod to enhance the immune response and protective immunity induced by the epitope vaccines from new gene wx2b4a of Toxoplasma gondii in mice.Methods Mice were divided into four groups,each group was inoculated intramuscularly three times with pcDNA3-W2b4a,pcDNA3-W2b4a and pidotimod,pcDNA3,natural saline (NS),respectively.The induced immune responses were tested by ELISA detecting IgG,and flow cytometry sorting the subsets of T lymphocyte.All mice were challenged with highly virulent RH tachyzoites to observe the survival time.Results After the immunization,the level of IgG in sera of mice inoculated with wx2b4a and pidotimod,wx2b4a were significantly higher than those control group(all P<0.05).CD4+ and CD8+T cell counts in immunized groups were higher than those control group(P<0.05).After the immunization,CD4+T cell counts and CD4+/CD8+T cell proportion in wx2b4a and pidotimod groups were higher than those in wx2b4a group (all P<0.05).After challenged with highly virulent tachyzoites,the mean survival time of wx2b4a and pidotimod groups was significantly longer than control group and wx2b4a group (all P<0.05).Conclusion Pidotimod can increase protective immunity of epitope vaccines from new gene wx2b4a of Toxoplasma gondii in mice.
3.Therapeutic Observation of Acupuncture-moxibustion for Chronic Simple Rhinitis Due to Yang Deficiency

Objective To observe the clinical efficacy of XIE’s Wen Du Qu Mai (Warming the Governor Vessel to expel smog) needling method plus heat-sensitive moxibustion in treating chronic simple rhinitis due to yang deficiency. Method Eighty patients with chronic simple rhinitis due to yang deficiency were randomized into a treatment group and a control group, 40 in each group. The treatment group was intervened by acupuncture plus heat-sensitive moxibustion, while the control group was by medication. The symptom and sign score was observed before and after 1-week treatment, and the clinical efficacy was compared. Result The total effective rate was 85.0%in the treatment group versus 55.0% in the control group, and the difference was statistically significant (P<0.05). The symptom and sign scores were changed significantly after treatment in both groups (P<0.01,P<0.05). After treatment, the symptom and sign score in the treatment group was significantly different from that in the control group (P<0.05). Conclusion XIE’s Wen Du Qu Mai needling method is effective in treating chronic simple rhinitis due to yang deficiency.
4.Meta-analysis on the efficacy of nebulized budesonide and salbutamol in the treatment of asthma

Objective To assess the efficacy of nebulized budesonide and salbutamol in the treatment of asthma by Meta‐analy‐sis .Methods According to the requirements of Cochrane systematic review ,a through literature search was performed among Wan‐fang database ,CNKI database ,VIP database of Chinese Periodicals and Pubmed database .Meta‐analysis was conducted on a total of 4 649 patients involved in 51 papers that met the inclusion criteria by RevMan4 .3 software .Results Compared with control group (RR=1 .27 ,95% CI=1 .23-1 .32) ,budesonide group which was treated by atomization inhales of nebulized budesonide based on routine treatment(RR=1 .31 ,95% CI=1 .16-1 .49) and salbutamol group which was treated by atomization inhales of salbutamol based on routine treatment(RR=1 .23 ,95% CI= 1 .17 -1 .28) ,the combined group which was treated by atomization inhales of nebulized budesonide and salbutamol based on routine treatment exhibited higher rates of total effectiveness with significant inter‐group difference when taking rates of total effectiveness as index(P<0 .01) .When taking hospitalization rate as index ,the result of the combined group was lower ,compared with the salbutamol group (RR=0 .34 ,95% CI=0 .13-0 .90) ,and the intergroup differ‐ence was significant(P<0 .05) .Conclusion The effect of nebulized budesonide and salbutamol in the treatment of asthma is cer‐tain .
5.Systematic Evaluation on Olmesartan Medoxomil Pharmacoeconomics Studies

OBJECTIVE:To evaluate the pharmacoeconomics characteristic of olmesartan medoxomil in ARB anti-hypertension drugs,and to provide reference for clinical drug use. METHODS:The literatures on the pharmacoeconomics study of olmesartan medoxomil published from Jan. 2002 to Dec. 2013 were searched with assigned search strategy from domestic and international data-bases like CNKI,PubMed,etc.,and comprehensive comparison and consistency analysis were conducted. RESULTS:4 literatures about pharmacoeconomics study of olmesartan medoxomil in different countries were screened,with essential hypertension patients as target population cost-minimization analysis or cost-effectiveness analysis as method,and 9 months-5 years as study course. The results consistency was well and showed the treatment cost of olmesartan medoxomil was usually less or better cost-effectiveness. CONCLUSIONS:Olmesartan medoxomil shows pharmacoeconomics advantage among ARBs drugs based on existed literature re-view.
8.Effect of Early Rehabilitation Nursing on Secondary Disturbance in Hemiplegics after Stroke

ObjectiveTo study the effect of early rehabilitation nursing on secondary disturbance in hemiplegics after stroke.Methods140 patients were randomly divided into two groups , rehabilitation nursing group (70 cases) and control group (70 cases). Patients in the rehabilitation nursing group were given clinical treatment and regularly convalescent nursing, while those in control group were given clinical treatment and unguided self-training. Evaluation was performed in pre-treatment and 12 weeks post-treatment respectively.ResultsSecondary disturbance rate in the rehabilitation nursing group was significant lower than that in the control group (P<0.01). The motor scores and ADL scores measured by Barthel Index in both groups raised after 12 weeks treatment, but the scores in the early rehabilitation nursing group were obviously superior to those in the control group (P<0.01).ConclusionEarly rehabilitation nursing for hemiplegics afte sroke may prevent or lessen the secondary disturbance, as well as obviously improve motor function of the limbs and ADL.
9.Role of TAK1 mediated cell autophagy after global cerebral ischemia-reperfusion in rats

Objective To study the effects of TGFβ-activated kinase-1 (TAK1)mediated cell autophagy after global cerebral ischemia-reperfusion (IR ) in rats.Methods Seventy-two male Kunming rats were randomly divided into six groups:control group (group C),sham operation group (group S),ischemia-reperfusion group (group IR),TAK1 shRNA lentivirus group (group T),nega-tive lentivirus group (group Y)and normal saline group (group NS)(n = 12 each).The rats in groups T,Y and NS received cerebral ventricles injection of TAK1 shRNA lentivirus,negative lenti-virus and normal saline 10 μl two weeks before preparing animal model.Using thread embolism of the right middle cerebral artery occlusion (MCAO)to cause focal ischemia for 2 h and released for 24 h for reperfusion in groups IR,T,Y and NS.The common carotid arteries were separated but not liga-ted in group S,the rest of the procedure as the same as group IR.The rats of each group were evalua-ted by neurological severity scores (NSS)24 h after reperfusion,the cerebral infarct volume was measured with the method of TTC and the expression of TAK1,LC3Ⅱ/LC3Ⅰ,Beclin1 and p62 pro-tein in rat hippocampus were determined by using Western blot.Results The infarct volume and NSS in groups IR,T,Y and NS were significantly higher than those in group C (P <0.05).The infarct volume and NSS in group T were significantly lower than those in group IR (P <0.05).TAK1 pro-tein of hippocampus in groups IR,Y and NS was significantly higher than that in group C (P <0.05).TAK1 protein of hippocampus in group T were significantly lower than that in group IR (P <0.05).LC3Ⅱ/LC3 Ⅰand Beclin1 protein of hippocampus in groups IR,T,Y and NS were signifi-cantly higher than those in group C,and the p62 protein of hippocampus in groups IR,T,Y and NS was significantly lower than that in group C (P <0.05).The LC3 Ⅱ/LC3 Ⅰand Beclin1 in group T were significantly lower than those in group IR,and the p62 protein of hippocampus in group T was significantly higher than that in group C (P < 0.05 ).Conclusion TAK1 mediated cell autophagy takes part in the mechanism of brain ischemia-reperfusion injury in rats.
10.Simultaneous Determination of Lysophophatidyl Choline and Lysophosphatidyl Ethanolamine in Nimodipine Fat Emulsion by HPLC

OBJECTIVE:To establish a method for simultaneous determination of lysophosphatidyl choline(LPC)and lysophos-phatidyl ethanolamine(LPE)in Nimodipine fat emulsion. METHODS:HPLC was performed on the column of Lichrosher Diol with detector of evaporative light scattering detector with mobile phase A of heptane- isopropyl alcohol solution(43∶57,V/V),mobile phase B n-heptane-isopropyl alcohol-water(29.5∶59∶11.5,V/V/V)(gradient elution)at a flow rate of 1.5 ml/min,column tempera-ture was 40℃,and the injection volume was 20 μl. RESULTS:The linear range was 0.020 0-0.400 0 mg/ml for LPC(r=0.999 0)and 0.010 0-0.200 0 mg/ml for LPE(r=0.999 6),and the logarithm value of concentration and peak area showed good linear relationship;the limit of quantitation was 0.013 4 mg/ml for LPC and 0.013 0 mg/ml for LPE;the limit of detection was 0.007 8 mg/ml for LPC and 0.007 6 mg/ml for LPE;RSDs of precision,stability and reproducibility tests were lower than 2%;recoveries were 95.96%-100.63%(RSD=1.83%,n=9)and 99.22%-101.76%(RSD=0.80%,n=9). CONCLUSIONS:The method is simple,and can be used for the determination of related substance in Nimodipine fat emulsion.
