2.Contribution of Professor SHI Xue-min's academic thoughts to treatment of stroke.

Based on the thought of Zhishen (a kind of mind regulation), Professor SHI Xue-min, academician of the China Academy of Engineering, found the Xingnao Kaiqiao (to refresh the mind and to cause resuscitation) acupuncture method, which still plays an important role in the acupuncture treatment of wind stroke nowadays. Meanwhile, great importance is attached to the comprehensive treatment of wind stroke. Danqi Piantan capsule (see text) is developed and "wind stroke unit" is set up. In recent years, Professor SHI shifts the center of research to the treatment of hypertension, the risk factor of wind stroke. Taking Renying (ST 9) as the major acupoint, acupuncture with standard measurement and manipulations is established. And good clinical effect has been obtained as well. Therefore, this article focuses on the introduction of Professor SHI Xue-min's contribution to wind stroke treatment.

6.Anterior cruciate ligament reconstruction with the remaining bundle and ruptured fiber

[Objective]To investigate the technique and significance of arthroscopic anterior cruciate ligament(ACL) reconstruction with the remaining bundle and ruptured fiber.[Method]In 47 cases of ACL rupture verified with arthroscopy,there were 15 cases of partial disruption,26 cases of complete disruption,and 6 cases of ligament flabby.ACLs were reconstructed with single bundle of autogenous quadrupled hamstring tendons under arthroscopy without remove of the remaining bundle or remnants of ACL.The femur tunnel was fixed by the RigidFix system,and the Intrafix system was used to fix the tibia tunnel.[Result]All of 47 cases were followed up for an average of 19 months(ranged from 12 to 24 months).The stabilization was strengthened after operation.Lysholm knee function score increased by 26.3 scores,from average 68.6 scores of preoperation to average 94.9 scores after operation.Lachman test was negative in 43 cases,weakly positive in 3 cases and positive in 1 case.According to Insall knee therapeutic effect evaluation criterion,38 cases achieved excellent results and 9 cases achieved good results.[Conclusion]Arthroscopic reconstruction of ACL with the remaining bundle preserving technique could facilitate revasculization and reinnervation of the reconstructed ligament and maintain stability of knee joint.
7.Inhibition of telomerase activity in colon cancer LS-174T cells with liposome-mediated cripto antisense oligodeoxynucleotide

AIM: To study the effects of cripto antisense oligodeoxynucleotide (ASODN) on telomerase activity in colon cancer LS-174T cells. METHODS: Colon cancer LS-174T cells were treated with cripto ASODN. The expression of cripto mRNA was studied using real-time PCR. The telomerase activity was detected by telomeric repeat amplification protocol enzyme-linked immunoabsordent assay (TRAP-ELISA). The growth of LS-174T cells was observed with soft agar. RESULTS: Cripto ASODN inhibited the growth of LS-174T cells in a dose-dependent manner in soft agar. Cripto ASODN decreased the telomerase activity in colon cancer LS-174T cells, and the effects of repression were time-and dose-dependent. CONCLUSION: Cripto may contribute to the activation of telomerase in colon cancer.
8.Effects of artemisunate on anoikis resistance in human breast cancer cell MCF-7

AIM: To study the effects of artemisunate on anoikis resistance in human breast cancer cells. METHODS: After breast cancer MCF-7 cells were treated with artemisunate, the growth of anchorage independence of breast cancer MCF-7 cells was examined in soft agar colony formation, and apoptosis of breast cancer were evaluated by terminal deoxynucleatidyl transferase-mediated deoxyuridine triphosphate nick end labeling (TUNEL) and DNA ladder. RESULTS: Artemisunate significantly inhibited the growth of anchorage independence of breast cancer MCF-7 cells in soft agar colon formation in a dose-dependent manner. Artemisunate induced apoptosis in breast cancer cells was conformed by DNA ladder and TUNEL assay, which was in a time-and dose-dependent manner. CONCLUSION: Artemisunate inhibits the growth of anchorage independence of breast cancer MCF-7 cells, which is related to anoikis.
9.Construction of Recombinant Adenovirus Expression Vector of Human Sema4C Gene and Its Expression in Mouse Myoblasts Cell Line C2C12

To generate recombinant adenovirus expression vector of human Sema4C gene and observe its expression in mouse myoblasts cell line C2C12 for ensuring easy access to investigate the role of Sema4C gene during myogenesis. The recombinant plasmid was packaged and amplified after being transfected in HEK293 cells through Lipofectamine. After infecting C2C12 myoblasts with recombinant adenovirus vector, the adenoviral infection efficiency was determined by confocal microscope which showed that the expression of green fluorescence could be detected at 12h and then reached peak at 24h after recombinant adenovirus infection. The infection efficiency was almost 100% confirmed by FACS examination. 10.Experimental study on the protective effect of anisodamine on flaps with ischemia reperfusion injury.

OBJECTIVETo observe the effect of anisodamine (Ani) injection on the survival rate and histologic result of flaps with ischemia-reperfusion injury, so as to demonstrate the protective effect of Ani on the flap survival.
METHODSA total of 48 healthy male Wistar rats were randomly divided into model control, normal saline(NS) and anisodamine groups, with 16 rats in each group. An 3 cm x 6 cm axial flap was formed at the right lower abdomen with abdominal superficial blood vessel as the pedicle. 0.5 cm x 0.5 cm skin tissue was taken from the middle part of flaps in each group, immediately after operation, 12, 18, 24 h after operation. The superoxide dismutase (SOD), nitric oxide (NO), nuclear factor-kappaB contents in the specimens were detected. The histologic study was also performed. The flap survival rate was recorded 7 days after operation.
RESULTSFlap survival rate was (78.6 +/- 7.3) % in Anisodamine group. 12, 18, 24 h after reperfusion injury, the SOD was (103.3 +/- 3.9), (82.6 +/- 3.8), (67.5 +/- 4.6) U/mg; the NO was (5.33 +/- 2.05), (4.75 +/- 1.68), (4.15 +/-1.59) nmol/mg; the NF-kappaB was 0.211 +/- 0.039, 0.313 +/- 0.033, 0.096 +/- 0.028. The contents of SOD, NO and NF-kappaB had the statistical difference of at different time. The skin pathological changes in Anisodamine group was obviously better than those in NS group. Flap survival rate in Anisodamine group was significantly higher than that in NS group.
CONCLUSIONSIn the flap with ischemia-reperfusion injury, Anisodamine can reduce the damage of free radical, increase the blood flow, reduce the production of NF-KB, decrease inflammatory reaction. So Anisodamine can increase the survival rate of flaps with ischemia reperfusion injury.
