To prepare monoclonal antibodies for blocking acute hemorrhagic conjunctivitis (AHC)viruses,BALB/C mice were immunized with Hela cells. The spleen of immunemouse was re-moved,and spleen cells were fused with SP2/0 cells. The antibodies against cell receptors wereselected by cell protection assay.Four cell strains secreted McAbs designated R7.R8.R16 andR24 were screened to be able to block cellular receptors and inhibit the infection of Ev70 as wellas CA24v on Hela cell. These results demonstrate that the receptors for Ev70 and CA24v are si-miliar.Indirect immunofluorescence test with the four McAbs showed definite fluorescence onliving Hela cells, which the intensity was concordance with the mean intensity resulted fromflowcytometer.Positive reaction on fixed Hela cells were only observed with R7 and R8.

China Oncology 2001;0(05):-
Radiotherapy is one of the main treatment options for patients with advanced limited non small cell lung cancer. From the viewpoint of radiobiology, the change of fractionation schedule in radiotherapy might improve the therapeutic effect. There are many different methods of fractionated irradiation: from conventional radiation including hyperfractionated radiotherapy, accelerated radiotherapy and hypofractionated radiotherapy, etc. In this paper,we summarized the definition of the different types of fractionated irradiation, and its clinical application.

Objective:To establish a rapid method to detect drug-resistance genotypes of extended spectrum-β-Lactamases (ESBLs) produced by gram negative bacillus using the real -time fluorescence quantitative PCR. Methods: According to clinical common genotypes of ESBLs, SHV, TEM.CTX-M.OXA and their homology, 9 pairs of specific primers were designed including SHV, TEM, CTX-M-1, CTX-M-2, CTX-M-8, CTX-M-9, OXA-1, OXA-2 and OXA-10. To extract DNA template by boiling assay, and then establish and grade up SYBR GREEN I real-time fluorescence quantitative PCR reaction system, finally definite real-time fluorescence quantitative PCR method. Its precision and range of linearity were tested. With established assay 51 multi- drug resistant ESBLs- E. coli K. pneumoniae were detected and compared with improved three dimensional extract tests. Results: Except OXA-2, 8 genotypes SHV, TEM, CTX-M-1, CTX-M-2, CTX-M-8, CTX-M-9, OXA-1 and OXA-10 were amplified by quantitative PCR from 39 ESBLs+ and 51 multi-drug resistant ESBLs-E. coli K. pneumoniae and confirmed by sequence testing. The range of linearity was 3×10~3-3×10~8 copies/mL, r =-0.994 7. Repetitive experiments showed that the average coefficient of variation between -runs was 9.6%. Comparing with three dimensional extract test, there was no significant difference (χ2 = 1.125,P> 0.05). Conclusion: Testing drug-resistance genotypes of ESBLs with SYBR GREEN I real-time fluorescence quantitative PCR is a rapid, specific and sensitive method, which is capable of inspecting genotypes of ESBLs from clinical strains.

Objective With the isocentric C-arm (Iso-C) three-dimensional computerized navigation system, cadaveric pelvic specimens were used to imitate double screw fixation of the symphysis pubic.Practicability and safety of the screw trajectory were examined postoperatively by local cadaveric dissections and imaging tests. Methods Pelvic specimens were harvested from 8 male and 7 female adult cadavers.Double screw placement in symphysis pubic was performed using the Iso-C three-dimensional navigation and entry point and safety trajectory was achieved. With the detailed local dissection postoperatively, distances from screw entry and exit points to unilateral structures (spermatic cord, femoral artery and vein, femoral nerve, obturator artery and vein, obtrurator nerve, and so on) were measured respectively. After complete removal of surrounding soft tissues of the specimen with only bony structure kept, the entry angle and length were calculated.Accuracy of double screw fixation of symphysis pubic was further checked using X-ray and CT.Results Entry point of the first screw was at the junction of unilateral pubic tubercle and transitional site of superior pubic ramus.Mean angle of the first screw with the horizontal plane was (7.7 ± 1.9) ° in men and (8.1 ± 1.7) ° in women.Mean angle between the first screw and coronal plane was (7.8 ± 1.8)° in men and (7.7 ±2.0)° in women.Entry point of the second screw was in the same place in the contralateral pubic tubercle.Mean angle between the second screw and horizontal plane was (30.6 ± 4.0) ° in men and (30.8 ± 3.4) ° in women.Mean angle between the second screw and coronal plane was (9.1 ± 3.0)°in men and (9.2 ± 3.3)°in women. Conclusions With the three-dimensional computerized navigation system, the bony channels of double screws implanted in the symphysis pubic are achieved and reliable.Percutaneous double screw fixation is feasible to treat the pubic symphysis diastasis.

Objective To investigate the causes of an outbreak of healthcare-associated infection with methicillin-resist-ant Staphylococcus aureus (MRSA)in a neurosurgical intensive care unit(NSICU).Methods Epidemiological investigation on 8 patients with lower respiratory tract infection (LRTI)in a NSICU between June 15 and June 28,2104 were performed by combination methods of prospective and retrospective survey. Results The attack rate of MRSA LRTI in NSICU patients was 22.86%,a total of 16 MRSA isolates were detected from patients’clinical specimens, nasal vestibule,as well as hospital surroundings during the period,pulsed-field gel electrophoresis (PFGE)result revealed that infection outbreak was caused by two subtypes of MRSA;risk factors analysis showed that long length of stay in ICU and aspiration of spu-tum through bronchoscopy were risk factors for MRSA LRTI.Conclusion Contamination of bronchoscope was the key factor for this epidemic spread of healthcare-associated MRSA infection.

Objective To screen and isolate the radioresistance related genes of IRM-2 mice. Methods cDNA library of IRM-2 mice was constructed by SMART technique.Total RNA was isolated from spleens of IRM-2 male mice. The first-strand cDNA was synthesized by using PowerScript reverse transeriptase, and double-strand cDNA was synthesized and amplified by long PCR.The PCR products were purified, digested with restriction enzyme Sfi I.The ds-cDNA fragment lessthan 500 bp was fractionated and ligated to the Sfi I-digested pDNR-LIB vector. The ligation mixture was transformed into E.coil DH5α by electroporution transformation to generate the unamplified cDNA library. The quality of cDNA library was identified by PCR technique.130 clones from cDNA library were sequenced and compared with GenBank database. Results The cDNA library contained 2.25 x 106 independent clones with an average insert size of 1.2 kb.The ratio of recombination and full-length was 95% and 55%,respectively.21 pieces of EST sequences from cDNA library were not the same as the known mice genes and registered into GenBank EST database, with registered number DW474856-DW474876.Conclusions cDNA library of IRM-2 mice has been constructed successfully.21 pieces of EST implies that radioresistance correlative genes may be in IRM-2 mice, which will lay a foundation for isolating and identifying radioresistance related genes in further study.
