1.Analysis of level change on peripheral blood lymphocyte subgroup,erythrocyte Zinc and hs-CRP concentrations in children with hand-foot-mouth disease

Objective To explore the level change of peripheral blood lymphocyte subgroup ,the erythrocyte zinc and hs-CRP in children with hand-foot-mouth disease (HFMD) .Methods Children with severe HFMD in this hospital were selected as experi-mental group ,and children in health that received physical examination in this hospital were selected as control group .Level of the peripheral blood lymphocyte subsets ,erythrocyte zine and hs-CRP were detected .Results In the HFMD group ,total T cell percent-age(CD3 + ) ,auxiliary/ induced T cells (CD3 + CD4 + ) percentage ,inhibit/cytotoxic T cells(CD3 + CD8 + ) percentage ,and the eryth-rocyte zine levels were all significantly lower than that of the control group(P< 0 .05) ,while the hs-CRP determination level ,B cells (CD19 + ) percentage were significantly higher than those of the control group(P< 0 .05) .Conclusion Children with severe HFMD have cellular immunity and hormone immunity function disorder and are lack of erythrocyte zinc .And their body cannot effectively resist infection virus ,which can lead to secondary bacterial infection .
2.Construction of pMSV-Slfn5-GFP plasmid and analysis of gene structure in mice

Objective To perform mouse pMSV-Slfn5-GFP gene recombinant expression plasmid construction and gene structure analysis.Methods Total RNA was extracted from mouse liver and turned into cDNA by reverse transcription.Mouse Slfn5 coding sequence (CDS) fragment was amplified by PCR and connected with the pGEM-T Easy vector.The connected product was transferred the E.coli DH5a.The positive clones were selected for extracting plasmid,which was identified by double enzyme of restriction endonuclease Hpa Ⅰ and Xho Ⅰ.Then correct plasmid identified by enzyme digestion was sequenced by Macrogen USA.Then correct plasmid by sequencing was connected with pMSV-GFP by HindⅢ and Xbo Ⅰ,which was named as pMSV-Slfn5-GF.UCSC (http://genome.ucsc.Edu/) was used to analyze mouse Slfn5 and its family genomic structure.Slfn5 protein structural domain was determined by NCBI.Results Slfn5 full-length gene sequence was cloned into the expression vector pMSV-GFP,the fragment size was about 2.65 kb by enzyme digestion identification.The conservatism of AAA_4 protein domain in Slfn5 protein family was determined by phylogeny.fr.Conclusion Mouse full-length gene pMSV-Slfn5-GFP expression vector is successfully constructed.
3.Diagnostic value of serum thyroglobulin autoantibody on recurrence and/or metastasis following surgery in patients with differentiated thyroid carcinoma

Objective To evaluate the clinical significance of serum thyroglobulin autoantibody(TGAb) in thyroglobulin(TG)-negative and TGAb-positive patients with differentiated thyroid carcinoma(DTC)after thyroid ablation and ascertain the optimal operating point(OOP)of TGAb.Methods A total of 169 patients with histologically confirmed DTC and thyroid remnant ablation showed TG-negative and TGAb-positive results which were assessed by electrochemiluminescence immunoassay(ECLIA).The cases were divided into group A(59 cases)and group B(110 cases)with or without evidence of recurrence or metastasis,respectively.The receiver operating characteristic(ROC)curve and positive likelihood ratio of different threshold values were analysed according to their serum TGAb level.Results Serum TGAb level(1368?1343)IU/ml in group A was significantly higher than that(154?539)IU/ml in group B(P1000 IU/ml was 1.12 times that of 204 IU/ml≤TGAb≤1000 IU/ml,4.03 times that of 100 IU/ml≤TGAb
4.Evaluation of thyroglobulin mRNA in peripheral blood of patients with differentiated thyroid carcinoma after remnant ablation

Objective The aim of this study was to evaluate the diagnostic value of thyroglobulin(Tg)mRNA in peripheral blood samples of patients with differentiated thyroid carcinoma(DTC)after remnant ablation.Methods Tg mRNA Was detected by reverse transcription-polymerase chain reaction(RT-PCR)in peripheral blood of 162 patients.SPSS 13.0 was used for date analysis.Results The Tg mRNA assay had higher sensitivity than the conventional serum Tg measurement[86.61%(97/112)vs 53.57%(60/112),X2=29.153,P<0.001]which Was the"gold standard"for the identification of recurrence or metastases.In the anti-Tg antibodies(TgAb)positive DTC patients,the sensitivity of Tg mRNA was higher than that of serum Tg[86.54%(45/52)vs 0]in identifying recurrent or metastatic thyroid disease(X2=79.322,P<0.001).There was a significantly positive correlation between Tg mRNA expression and the clinical stages(Kendall correlation coefficient=0.515,P<0.001).There was no difference of Tg mRNA expression in peripheral blood among ages,sex,pathological types and location of metastases,respectively(Kendall correlation coefficient=0.020,0.069,0.050 and 0.028;all P>0.05).Conclusions Circulating Tg mRNA is a more sensitive marker than serum Tg in identifying recurrent or metastatic DTC,particulady in patients during levo-thyroxine(l-T4)therapy and with TgAb-positive.The DTC patients who have positive expression of Tg mRNA indicates poor prognosis.

6.Clinical Observation of Acupuncture by Stages for Post-stroke Swallowing Dysfunction

Objective To observe the clinical efficacy of acupuncture by stages in treating post-stroke swallowing dysfunction. Method Sixty eligible patients were randomized into a staging-acupuncture group and an ordinary-acupuncture group, to respectively receive acupuncture by the stage of swallowing and ordinary acupuncture in addition to the basic treatments. Function estimation was conducted before and after intervention in both groups to compare the therapeutic efficacies.Result According to the water drinking test, the total effective rate was 83.3% in the staging-acupuncture group versus 53.3% in the ordinary-acupuncture group, and the difference was statistically significant by rank sum test (P<0.05), and the staging-acupuncture group was superior to the ordinary-acupuncture group especially in the pharyngeal stage (P<0.05); the differences were statistically insignificant in the oral stage and esophageal stage (P>0.05); regarding the video fluoroscopic swallowing study (VFSS), the staging-acupuncture group was significantly better than ordinary-acupuncture group in improving swallowing function (P<0.05).Conclusion Acupuncture by swallowing stages is effective in treating post-stroke swallowing dysfunction, especially in improving the swallowing function in pharyngeal stage.
7.Submucosal myoma resection by guidance of esophageal endoscopic ultrasound

Objective To investigate the effects of submucosal resection by guidance under esophageal endoscopic ultrasound. Methods 79 cases ofsuspected upper gastrointestinal submucosal tumors were found by endoscopic ultrasonography,transesophageal endoscopic ultra-sound showed lesions under muscularis mucosa,after informed consent,they were underwent endoscopic mucosal resection. Pathological diag-nose of the lesions were found out after resection. Results Compared with pathological results showed that endoscopic ultrasound accurately determine tumor location in the hierarchy;through endoscopic mucosal resection to remove all lesions,5 patients were found bleeding postop-erative,6 patients were found esophageal stricture,the prognosis was good after treatment. Conclusion Endoscopic ultrasonography can accu-rately determine submucosal myoma,it can be used to guide resection of submucosal myoma.

New products of ethnic medicine are important application form to play therapeutic advantages of ethnic medicine, whose research and development should focus on the maintenance of characteristics of ethnic medicine. From the perspective of the research and development of ethnic medicine, this article discussed research ideas in the development of new products of ethnic medicine and put forward that attention should be paid to the experience on using the ethnic medicine, related literature studies, and ethnic medicine resource protection and utilization as well. In addition, this article put forward the research tactics in new products of ethnic medicine, such as medicinal materials, effective parts, active ingredients, the secondary development research and so on, with a purpose of improving the level of researching and developing new products of ethnic medicine.
9.Impacting factors on the professional identity of railway young workers and the relationship with the professional identity and subjective well-being

Objective To investigate the impacting factors on the professional identity of railway young workers and the relationship of the professional identity and subjective well-being.Methods The sample consisting of 5 762 young workers was selected from railway enterprises by stratified and random sampling according to the proportion.Occupational professional identity Scale and Index of Well-being,Index of General Affect Scale were used.The affect of the variable,such as sex,age,working years,educational background and income on professional identity were studied.Moreover,the relationship between professional identity and subjective well-being was reported.Results (1) Professional identity ascended witb the increase of ages,and there was significance between three groups(F=6.667,P<0.05).(2)Professional identity ascended with the increase of length of service (F=5.147,P<0.01),but it dropped at the point of 9-12 years.(3) The professional identity of workers with bachelor degree was higher than that with junior college education background.and there was significance between three groups (F=3.538,P<0.05).(4)The workers who had more than 3 500 RMB monthly salary was regarded on the top in professional identity,while those whose monthly salary were under 2 500 RMB was in the lowest level (F=42.275,P<0.01).(5)The result of linear regression indicated that the degree of professional identity can forecast the degree of subjective well-being(R =0.534,P<0.01).Conclusion The professional identity of railway young works was affected by their ages,the length of service,the educational degree and salary,The professional identity can forecast subjective well-being.

Aim To explore the effect of Procyanidin from Vaccinium(PC) on the myocardial fibrosis and its mechanism.Methods The model of myocardial fibrosis in rats was built by subcutaneous injection of isoprenaline(Iso) for 5 mg?kg-1?d-1 in vivo.Meanwhile,rats were treated with PC 100,200 and 400 mg?kg-1?d-1 by gastrointestinal administration.The contents of hydroxyproline(Hyp),malondialdehyde(MDA) and the activity of superoxide dismutase(SOD) in left ventricle were assayed with spectrophotometry respectively.Changes of myocardial ultrastructure were observed by electronic microscope.The cultured neonatal rat CFb was isolated by trypsin digestion method.The proliferation of CFb was induced by Angiotensin Ⅱ and assessed by thiazolyl blue(MTT) assay.PC was added in different dosage(25,50 and 100 mmol?L-1).Ⅰ,Ⅲ collagen quantification and TGF-?1 levels were determined by the ELISA.Results In vivo,compared with model group,the contents of hydroxyproline and malondialdehyde of left ventricle were significantly reduced and the activity of superoxide dismutasein left ventricle was markedly enhanced in PC three groups(P
