OBJECTIVE:To get clear idea about the current situation and future trend of use of antidepressants METHODS:By using the methods of order of sum of money and DDDs,the antidepressants used in our hospital in years 1998~2001 were analysed RESULTS:The annual sum of money and DDDs of antidepressants were increasing and the sum of money and DDDs of antidepressants used in 2001 were 3 69 times and 2 14 times as much as those in 1998,respectively Although the proportion of sum of money of tricyclic antidepressants(TCAs) was lower than that of new type antidepressants(NTAs),Clomipramine hydrochloride and Doxepin belonging to TCAs were in the first five places in order of DDDs during the past four years Because of the low price and less side effect,the position of NTAs in therapy of depressions was increasing CONCLUSION:TCAs are still the first choice in therapy of depressions The prospects of NTAs will be wide and bright It is important to speed up the development of Chinese materia medica in therapy of depressions

OBJECTIVE :To study the preparation technique and clinical therapeutic effect of Tangan capsules METHODS : Tangan capsule was made of chrominum pyridinate ,crystalline cellulose ,starch and vehicle The content of chrominum pyridi natein product was detected with HPLC RESULTS :This preparation was rationalin design ,feasible in preparation technique , stable in quality,reliable in quality control and satisfactory in the rapeutic efficacy CONCLUSION :Tangan capsuleis of im portance as an accessory agent in treating type 2 diabetes.
3.Action Mechanism of Rukang Oral Liquid in Rats with Hyperplasia of Mammary Glands

Objective:To study the action mechanism of Rukang oral liquid in rats with hyperplasia of mammary glands. Meth-ods:The rats were randomly divided into six groups:the normal group, the model group, tamoxifen group, low, medium and high dose of Rukang oral liquid groups. The model of hyperplasia of mammary glands was induced by estradiol benzoate injection and progester-one injection. After the administration, ELISA was used to detect the serum concentrations of ER, PR, P53 and Oct4, Western blot was used to detect the expression of ER, PR, P53 and Oct4 in breast tissue, a fluorescence quantitative PCR method was used to de-tect the expression of GnRH, GnRHR, and the expression of telomerase in breast tissue was detected by in situ hybridization. Results:Rukang oral liquid could reduce the expression of ER and PR in serum and breast tissue(P<0. 01), reduce the expression of P53, te-lomerase and Oct4(P<0. 01), and decrease the expression of GnRH in hypothalamus and pituitary, which could reverse the hyperpla-sia of mammary glands and played the role effectively(P<0. 01). Conclusion: Rukang oral Liquid can reduce the expression of ER and PR in serum and breast tissue, and decrease the expression of GnRH in hypothalamus and pituitary to play the role in the treatment of hyperplasia of mammary glands. Rukang oral Liquid may prevent breast hyperplasia from developing into breast cancer through re-ducing the expression of three related markers P53, telomerase and Oct4.
4.Study on Relationship between Dopamine D1 Receptor Gene Polymorphisms and Clozapine Efficacy in Male Schizophrenic Patients

Objective:To investigate the relationship between dopamine D1 receptor ( DRD1 ) gene polymorphisms and clozapine efficacy in male schizophrenic patients. Methods:Totally 46 male schizophrenic patients were treated by clozapine for 6-8 weeks. The clinical response was determined by the Positive and Negative Symptom Scale (PANSS). DRD1 gene rs265981, rs5326, rs4532, rs1799914, rs686 and rs4867798 polymorphisms were detected by the gene sequencing, while plasma clozapine levels were monitored during the treatment. Results:The total clinical efficacy of clozapine response group and the non-response group was compared, the distribution of rs265981 genotype TT, TC and CC and allele T and C had statistically significant differences (P=0. 025;P=0. 005), and the distribution of rs686 genotype CC, CT and TC and allele C and T had statistically significant differences ( P=0. 044;P=0. 010). The negative symptom of the response group was compared with that of the non-response group, the distribution of rs4532 gen-otype GG, GA and AA and allele G and A had statistically significant differences (P=0. 034; P=0. 013). Conclusion: The poly-morphisms of DRD1 gene rs265981 and rs686 may have influence on the clinical efficacy of clozapine, and rs4532 may have influence on the negative symptom.
5.Regional differences of genotype frequency of 5-hydroxytryptamine 2A receptor gene T102C of schizophrenics patients

BACKGROUND:Now researches on psychiatric disease and molecular heredity are becoming more and more gradually because of the development of molecular genetics,but conclusions are controversial,and for the time being there are no reports about whether there are regional differences and ethnical differences in the distribution of genotype frequency of 5-hydroxytryptamine 2A receptor gene T102C.OBJECTIVE:Researches on the geographic distribution of genotype frequency of 5-hydroxytryptamine 2A receptor gene T102C of schizophrenics people.DESIGN:Sample survey and observation with schizophrenics selected as subjects.SETTING: Mental Department, Changzhou Peace Hospital.PARTICIPANTS: The experiment was completed in The No.102 Hospital of PLA from January 1999 to August 2003.Totally 177 people of Han nationality (Age: 18-45; Duration of illness: 1 month-20 years) from different provinces and autonomous regions of China in compliance with diagnostic standards of Psychiatric Disease Classification and Diagnosis Standards of China (The 3rd version) were selected as the subjects.METHODS:The blood samples from 117 schizophrenics were given test of polymerase chain reaction(PCR),and the distribution differences of genotype frequency of 5-hydroxytryptamine receptor gene of normal control population from different provinces and autonomous regions were compared. DNA were abstracted by means of phenol chloroform, amplification and cataphoresis test of polymerase chain reaction of target DNA,primer sequence of gene segment amplification of 5-hydroxytryptamine (5-HT) 2A receptor gene T102C: 5'-TCT GCT ACA AGT TCT GGC TT-3', 5 '-CTG CAG CTT TTT CTC TAG GG-3'; Reaction system of 50 μL, DNA 0.5 μg,Primer 50 pmol,TagDNA enzyme 2U were added with dNTP to the final concentration of 200 μmol/L.MAINOUTCOMEMEASURES: DNA cataphores is test of schiophrenicanddistributionofgenotypefrequencyof5-hydroxytryptamine 2A receptor gene.RESULTS: The distribution of genotype frequency of 5-hydroxytryptamine(5-HT) 2A receptor gene: A1A1, 0.07-0.03; A1A2, 0.50-0.72; A2A2,0.17-0.29. Correlation analysis of genotype frequency in different regions:A1A1, x2=4.44, P=0.617 1, P > 0.05;A1A2, x2=1.14, P=0.942 2, P > 0.05;A2A2, x2=0.93, P=0.985 7, P > 0.05.CONCLUSION: The geographic distributions of genotype frequency of 5-hydroxytryptamine 2A receptor gene T102C of schizophrenics of the Chinese Han nationality people were comparatively even.
6.Sedative and hypnotic effects and mechanism of Yening Capsules on mice

Objective To study the sedative and hypnotic effects of Yening Capsules and investigate its bioactive mechanism in mice. Methods The mice were randomly divided into control group, estazolam group (0.8 mg/kg), low, medium and high-dose Yening Capsules groups (400, 600 and 800 mg/kg). The locomotor activity, latency to persistent sleep, sleep duration and sleep rate were determined respectively in mice via the open field test and injection of pentobarbital sodium in subthreshold and suprathreshold doses. The content of GABA, 5-HT, DA and NE in brain tissue of mice were detected by enzyme-linked immunosorbent assay(ELISA). Results Compared with the control group, Yening Capsules medium and high dose group (P<0.05, P<0.01)significantly decreased the locomotor activity of mice.The sleep latency in Yening Capsules medium and high dose group were significantly shorten (P<0.05, P<0.05)and the sleep duration (P<0.05, P<0.01)were extended. The sleep rate of Yening Capsules medium and high dose groups (P<0.05, P<0.01) was significantly increased. Compared with the control group, high dose of Yening Capsules can significantly increase GABA (P<0.05), 5-HT (P<0.05), DA (P<0.05), NE (P<0.01) in mouse brain tissue. Conclusion Yening Capsules had obvious sedative and hypnotic effects, and its mechanism may be related to the increasement of GABA, 5-HT, DA and NE level in brain tissue of mice.
