1. Expression purification and biological activity of a fusion protein IVT

OBJECTIVE: To clone, express and purify a fusion protein IVT, and detect its biological activity. METHODS: A fusion gene IVT with immunal function, anti-angiogenesis and apoptosis-inducing activities was constructed by genetic engineering means. pPIC9-IVT was constructed and transformed into Pichia pastoris GS115(his4). The recombinant fusion protein IVT was expressed in yeast engineering strain and purified from the culture supernatant. MTT assay was used to observe the effect of the IVT on the proliferation of CTLL-2 cells, human umbilical vein endothelial cells(HUVECs)and NCI-H446 cells. Transwell migration assay was used to observe the impact of the IVT on cell migration ability of HUVECs. The apoptosis of NCI-H446 cells was examined by Hoechst staining. RESULTS: The IVT promoted CTLL-2 cells proliferation, inhibited HUVECs and NCI-H446 cells proliferation, decreased HUVECs migration and induced apoptosis of NCI-H446 cells. CONCLUSION: The IVT has a variety of biological activities. This study laid a foundation for the development of novel multi-functional fusion proteins.
2.Celecoxib suppresses xenograft tumor of colon cancer in nude mice

Objective To evaluate the effects of celecoxib on tumor growth,COX-2 and survivin expressions and angiogenesis in nude mice. Methods Xenograft animal model was established by injecting human colon cancer HT-29 cells into the BALB/c nude mice subcutaneously. Fifty mice were randomly divided into 4 groups 16 d after injection:control group,celecoxib group(receiving 25,50,75,100 mg?kg-1?d-1 for 35 d). Tumor volumes were measured every week. The expression level of COX-2,survivin and the microvessel density (MVD) of the xenograft tumor tissues were measured by immunohistochemistry,and mRNA level of VEGF by RT-PCR. Vectors used to carry foreign genes play an important role in gene therapy, among which, the adeno-associated virus (AAV) has many advantages, such as nonpathogenicity, low immunogenicity, stable and long-term expression and multiple-tissue-type infection, etc. These advantages have made AAV one of the most potential vectors in gene therapy, and widely used in many clinical researches, for example, Parkinson's disease. This paper introduces the biological characteristics of AAV and the latest research progress of AAV carrying neurotrophic factor, dopamine synthesis related enzymes and glutamic acid decarboxylase gene in the gene therapy of Parkinson's disease.

4.Long-term efficacy of surgical treatment of slow transit constipation with pelvic floor hernia and rectal mucosal prolapse

Objective To evaluate the long-term therapeutic efficacy of subtotal colectomy,ascending colon-rectum anastomosis, pelvic floor hernia repair, functional rectal suspension, and uterine suspension surgery for slow transit constipation with pelvic floor hernia and rectal mucosal prolapse.Methods From June 2007 to May 2008, 35 patients with intractable constipation caused by slow colonic transit combined with pelvic floor hernia and rectal mucosal prolapse underwent subtotal colectomy and ascending colon-rectum anastomosis, pelvic floor hernia repair, functional rectal suspension, and uterine suspension surgery. Postoperative defecation, complications, quality of life, and degree of satisfaction were followed-up. Results The average follow-up period was two years. At one month after the operation, the average defecation frequency was five times (2 -8 times) a day, with a semi-liquid stool consistency; After two years the frequency was twice ( 1 -3 times) a day, with solid stool consistency. Of the 35 patients, 19 were satisfied with the surgical efficacy, and 16 were very satisfied. All the patients' quality of life improved significantly. Conclusions For patients suffering from slow transit constipation with pelvic floor hernia and rectal mucosal prolapse subtotal colectomy, ascending colon-rectum anastomosis, pelvic floor hernia repair,functional rectal suspension, and uterine suspension surgery has satisfactory results.

Objective To study the antagonistic effect of N-acetylcysteine(NAC)on the damage of hepatic mitochondria of rats induced by cadmium in vitro.Methods The mitochondria were prepared from the clean Wistar rats' whole liver by using differ ential centfifugation.The mitochondria were incubated in the assay buffer containing different concentration of CdCl_2 (10,100,1 000,10 000 ?mol/L)at 37 ℃ for 1 h.The effect of NAC(500 ?mol/L)was studied at a CdCl_2 concentration of 1 000 ?mol/L.The incubation buffer was collected and the level of GSH,cytochrome C and the activity of Mn-SOD were determined. 7.Effect of preoperative cyclooxygenase-2 inhibitor for postoperative pain in patients after total knee arthroplasty: a meta-analysis.

OBJECTIVETo systematically evaluate the efficacy and safety of preoperative administration of cyclooxygenase-2 (COX-2) inhibitor on pain occurring with total knee arthroplasty (TKA).
METHODSWe electronically searched PubMed, Cochrane Library, EMBASE, CNKI, CBM, Wanfang data from inception to March 15, 2014 and manual searched journal of library collection to identify randomized controlled trials (RCTs) about preoperative administration of COX-2 inhibitor on pain occurring with TKA. The methodological quality of the included RCTs was assessed and the data were extracted according to the Cochrane Handbook 5.1.0. Meta-analysis was performed by using RevMan 5.2 software.
RESULTSA total of 6 RCTs involving 228 patients were included. The results of meta-analyses showed that: (1) Efficacy: The visual analog scale (VAS) of post-operation at 12-hour (WMD = -0.60, 95% CI -0.83 to -0.37, P < 0.000 01) and 24-hour (WMD = -0.74, 95% CI -1.29 to - 0.19, P = 0.008) was decreased when COX-2 inhibitor was used before operation. And compared with control group, experimental group decreased the modified numerical pain rating scale (MNPRS) at 24-hour (WMD = -0.50, 95% CI -0.70 to -0.30, P < 0.000 01), 48-hour (WMD = -0.55,95% CI -0.65 to -0.45,P < 0.000 01) under quiescent conditions, and the same result at 24-hour (WMD = -0.82, 95% CI -1.26 to -0.38, P <0.000 01), 48-hour (WMD = -0.71, 95% CI -0.82 to -0.60, P < 0.000 01) under active conditions. The morphine consumption postoperatively were fewer in experimental group at the first day (WMD = - 1.35, 95% CI -1.92 to -0.79, P < 0.000 01) and the second day (WMD = -1.60, 95% CI -2.68 to -0.52, P = 0.004). (2) Safety: COX-2 inhibitor could lessen the incidence of postoperative pruritus (RR = 0.35, 95% CI 0.15 to 0.84, P = 0.02), but not statistically decrease of nausea and vomiting (RR = 0.83, 95% CI 0.54 to 1.28, P = 0.40) and exhaustion (RR = 0.63, 95% CI 0.05 to 7.67, P = 0.72).
CONCLUSIONThe current evidence indicated that preoperative administration of COX-2inhibitor can effectively improve the effect of postoperative analgesia, reduce the consumption of morphine and lessen the incidence of pruritus. Due to the limited quantity of the included studies and the evidence with limited strength,further high-quality RCTs are needed to verify the aforementioned conclusion.

8.The correlation study about quality of life and depression in opioid dependent patients receiving methadone maintenance treatment

Objective To explore the depression status and its con-elation with quality of life(QOL) in patients receiving methadone maintenance treatment (MMT). Methods 120 patients treated with MMT were assessed with Quality of Life Scale for Drug Addicts (QOL-DA) and Beck Depression Inventory (BDI-21). Results More than 41 years old group's QOL-DA physical function domain(28.79 ±8.53) was significantly lower than 31~40 years old group (33.52 ± 6. 73) (P= 0.003). According to BDI-21 score ,55% patients receiving MMT were classified as moderate or severe depression. QOL-DA scores decreased in order of the depression severity classified as no depression group,mild depression group,moderate depression group and severe depression group according to the BDI-21 score,and the difference between the groups were significant statistically (P<0.01). There was significantly negative correlation between BDI-21 score and QOL-DA score (P<0.01). Conclusion Depression is very common in opioid dependent patients receiving MMT and decreases their quality of life,so it is important to enhance depression intervention or treatment in MMT clinics.
9.Estimation of typical value and confidence interval of gamma-radiation dose rate from tank-transported copper ore by means of bootstrap resampling method

Objective To investigate the typical value of gamma-radiation dose rate and its confidence interval in tank-transported copper ore by using bootstrap resampling techniques. Methods Bootstrap resampling method, coupled with kernel density estimation, introduced to acquire the typical value of gamma-radiation dose rate in copper ore. Results The typical value of gamma-radiation dose rate in copper ore was expressed as the central tendency of the means of resampling, and two kinds of confidence interval, empirical percentile and bias-corrected accelerated confidence interval, were provided as standard error. Conclusion It is clearly demonstrated that this method has an advantage to give a robust description in explanation of central tendency and variation range of gamma-radiation dose rate data profiles.
