BACKGROUND: Pathological changes of form and structure are induced in some tissues during repairing albuginea penis trauma, while structure and component of collagen fiber will be correspondingly changed necessarily.OBJECTIVE: To find a convenient, shortcut, and sensitive method to observe the dynamic change of type and distribution of collagen fiber during albuginea penis healing. DESIGN, TIME AND SETTING: A contrast study was performed at the Laboratory of Tissue Engineering, Guiyang Medical College between June 2005 and October 2007. MATERIALS: Fifteen male New Zealand rabbits were used in this study. METHODS: All rabbits underwent an excision of albuginea sizing 5 mm×5 mm at dorsal penis to establish models of albuginea penis trauma, and then the excision was sutured in situ. Thereafter, albuginea penis was obtained from five rabbits once after 2, 6, and 12 weeks. The samples were treated with picric-sirius red staining and observed under polarizing microscope. MAIN OUTCOME MEASURES: Dynamic changes of area of collagen fiber during albuginea penis healing.RESULTS: There were significant differences in content of the same collagen fiber at different time points (P<0.05). At 2 weeks post-trauma, both type Ⅰ and type Ⅲ collagen fibers were observed, and the ratio was equal, but the arrangement was confused and disorder. At 6 weeks post-trauma, red and yellow type Ⅰ collagen fiber was apparently increased, but type Ⅲ collagen fiber was decreased, and the arrangement was still disorder. At 12 weeks post-trauma, thick, bright red and yellow type Ⅰ collagen fiber was rapidly increased, but thin and green type Ⅲ collagen fiber was rapidly decreased.CONCLUSION: Picric-sirius red polarized light method can not only differentiate types of collagen fiber, but also clearly display the distribution, form and proportion of type Ⅰ and type Ⅲ collagen fiber, characterizing by simple, convenient, specific, and highly sensitive.

PurposeTo study the effect of gasoline on the extracellular matrix of dermis in rat. Methods45 male Wistar rats were divided into control group, 3 different exposure level groups, which were exposed to gasoline on 2 cm× 3 cm skin area with the dose of 250 mg/cm2 for 1 (group A) ,4(group B)and 8 days (group C) respectively, and an intervention group exposed to 250 mg/cm2 × 8 days gasoline after the application of protective agent on skin. After the treatment, collagen, elastin and glycosaminoglycan (GAG) were assayed in skin departed from intoxicated area.ResultsCompared with control group, collagen was decreased in group C(P<0.05); elastin was decreased in group B and C (P<0.05); glycosaminoglycan was decreased in all exposure groups and intervention group ( P < 0.05 or P < 0.01 )。 Collagen, elastin and glycosaminoglycan(GAG) were significantly lower in group C than in intervention group ( P < 0.05 ). ConclusionsThe extracellular matrix of dermis, including collagen, elastin and GAG were decreased in rat dermal exposed to gasoline.

Objective To study the anti-fatigue effect of Cynomorium songaricum flavone(CSF)on old rats.Methods50 old male Wistar rats were randomly divided into the control group(no swimming,no drugs),single swimming group(swimming, given saline by gavage),and three swimming plus CSF groups(swimming, given CSF 20 g/kg weight,10 g/kg weight and 5 g/kg weight respectively),total five groups with 10 animals in each group.Animals swam with free swimming in endless cycles water 30 minutes once every day for 10 days. The level of monoamine oxidase(MAO) and glutathione peroxidase(GSH-px) in blood of animals were examined by MAO kit,GSH-px kit and ultraviolet spectrophotometer at 11th day.ResultsCompared with single swimming group, the body weights of swimming plus CSF groups increased significantly by 12.9%,80.6% and 200% respectively(P<0.01);swimming times before sinking increased by 19.4%,29% and 48.4%;sinking times of each rat(time/10 min) decreased by 17.8%,43.2% and 72.9%;total swimming times increased by 2.8%,7% and 29.6% respectively(P<0.01);MAO decreased by 60%,69.8% and 80.5%,GSH-px increased by 465.3%,563.8% and 635% respectively(P<0.01).ConclusionCSF shows anti-oxidative and anti-fatigue properties and can be given as prophylactic/therapeutic supplements for increasing antioxidant enzyme activities and preventing lipid peroxidation during strenuous exercise.

OBJECTIVETo develop the chromatographic fingerprints of composite Folii Isatidis injection by HPLC.
METHODThe separation was performed on a Diamonsil C18 column with a mobile phase consisting of methanol-water-phosphoric acid as gradient eluent at the flow rate of 1.0 mL x min(-1). The UV detection was set at 254 nm.
RESULTA standard HPLC fingerprint procedure was developed for composite Folii Isatidis injection, with 20 common peaks and a similarity threshold of 9.0 established.
CONCLUSIONThis method was accurate, repeatable and useful for the quality control of composite Folii Isatidis injection.

To investigate the upregulated genes associated with cold acclimation, a cold acclimation model was established based on Balb/C mouse. mRNA of muscle and liver were isolated, and the upregulated genes of these tissues were studied by representational differential analysis (RDA). The upregulated genes then were sequenced and searched by Blast software in GenBank database. The results showed that some genes were upregulated and possibly associated with cold acclimation. Three of these genes, transferrin, fibrinogen B-beta-chains and a new gene fragment (Genbank ID: AF454762), were confirmed to be upregulated by RNA slot-blot analysis. The finding of these genes might contribute to further understanding of the molecular mechanisms of cold acclimation.

OBJECTIVETo investigate the effects of catecholamine hormone on the blood and brain of heroin addicts.
METHODSRats were divided into three groups and treated with the glucose (control group), the heroin (im) (heroin group), and the combination of the intramuscular injection of reserpine and heroin (reserpine group). Changes in the levels of the dopamine (DA), cAMP, and cGMP were detected by the radioimmunoassay (RIA) method in the blood and brain tissue.
RESULTSNo significant withdrawal symptoms were observed in the reserpine group. Compared with the control and heroin groups, the blood cAMP levels were increased by 35.36% and 15.53% in the reserpine group, respectively; the cAMP levels in the midbrain ventral tegmental area (VTA), prefrontal cortex (PFC), and hippocampus (Hipp) were increased by 24.08% & 8.53%, 15.66% & 8.13%, and 21.95% & 8.40%, respectively. While compared to the control and heroin groups, the DA levels of the PFC, Hipp, striatum, and nucleus accumbens (NAc) were significantly reduced in the reserpine group, decreasing by 74.09% & 82.86%, 81.06% & 82.23%, 91.62% & 86.55% and 84.35% & 90.63%, respectively. The concentrations of cGMP of the brain tissues in the reserpine group were lower than those in the control group. In addition, the neural electrophysiological testing showed that the electroencephalogram (EEG), electrocardiogram (ECG), and muscle spindle discharge diagram of rats in both the reserpine and heroin groups were apparently changed.
CONCLUSIONCatecholamine hormone plays an important role in heroin addiction.
