Objective:To explore the anti-apoptostic effect of Ghrelin in cardiomyocytes after Hypoxia/Reoxygenation injury and manifest the specific cellular singaling transduction pathway.Methods:Newborn SD rats' hearts were isolated and cardiomyocytes were separated by trypsin digestion method .The primary cardiomyocytes were collected and incubated in vitro and randomly alloted to 4 groups(①Control;②H/R group;③H/R +Ghrelin group;④H/R+Ghrelin+Wortmannin group) after 5 days of cultivation.Hypoxia/ Reoxygenation injury was established by 4 hours of hyopxia and subsquently 1 hour of reoxygenation.Lactic dehydrogenase concentration,cell apoptotic rate and the expression of phosph-Akt were detected by Automatic Biochemistry Analytic Instrument ,Tunel method and Western blotting method respectively.Results:Compared with H/R group and H/R+Ghrelin+Wortmannin group,the LDH concentration in media was significantly reduced and the myocardium apoptotic rate was pretty low in H/R+Ghrelin group.Ghrelin also increased phosph-Akt activity but this effect was abolished by applying Wortmannin simultaneously.Conclusions:Ghrelin provides concrete protection against Hypoxia/ Reoxygenation injury in the primary cultivated rat cardiomyocytes,which manifest dereasing LDH leaking,inhibitting cell apoptosis.Theantiapoptotic effect is partly mediated by the activation of the PI3K /Akt(Phosphatidylinositol-3 kinase/ Akt) pathway.

Objective To study the viability of S.aureus in common cosmetics and the sensitivities of different detection methods adopted in detecting faecal coliform. Methods A standard method stated in "Hygienic Standard for Cosmetics" and lactose-bile salt medium improved by adding neutralizers were adopted in detecting Faecal coliform. Results The amounts of S.aureus dropped gradually as the prolongation of preservation period and in cosmetics containing or not containing antiseptics were different too. In detecting Faecal coliform, the sensitivity of the improved method was higher than that of the standard method. Conclusion Some ingredients of common cosmetics, especially antiseptics, obviously affected the viability of S.aureus. The lactose-bile salt broths containing Lecithin and Tween 80 were much better than those not containing Lecithin and Tween 80 in detecting faecal coliform.

Objective To investigate the synergistic effects of pirarubicin (THP) combined with hyperthermia on proliferation and invasive potential of gastrointestinal cancer cells in vitro and to discuss the underlying mechanisms. OBJECTIVE: To establish a HPLC method for the determination of content of entecavir and its related substances. METHODS: The HPLC condition was consisted of a Luna C18 column with a column temperature at 25 ℃, using gradient eluate method, the mobile phase was acetonitrile,with a flow rate of 1.0 mL?min-1 and detection at 254 nm. RESULTS: The calibrated linear curve of entecavir was within 0.033 45～0.167 2 mg?mL-1(r=0.999 98). The average recovery was 100.09%,RSD=0.20%, The content of related substance was 0.54%～1.48%. CONCLUSION: This accurate and reliable HPLC method is applicable for the quality control of entecavir and its related substances.

AIM To investigate the effects of Tongluo Xingnao Effervescent Tablets (Chuanxiong Rhizoma,Angelicae sinensis Radix and Scutellariae Radix) on the learning and memory function in SAMP8 mice,and improvements in cognitive function.METHODS Morris water maze was used to evaluate the change of cognitive function in SAMP8 mice after they were treated with Tongluo Xingnao Effervescent Tablets for 60 d,and the activity of superoxide dismutase (SOD),the concentration of protein carbonyls (PC),the ratio of NAD +/NADH in brain tissue were detected by ELISA,the expression of Nampt,SIRT1 and FOXO3 in hippocampus were measured with immunohistochemical methods.RESULTS Tongluo Xingnao Effervescent Tablets shortened escape latency and obviously increased percentage of time in target quadrant in hidden platform test and increased the times entering the target quadrant in spatial probe test in SAMP8 mice.It evidently enhanced the activity of SOD,the ratio of NAD +/NADH and distinctly decreased the protein carbonyls level.Moreover,Tongluo Xingnao Effervescent Tablets could markedly up-regulate the expression of Nampt and SIRT1,but evidently down-regulate FOXO3 protein expression.CONCLUSION The experimental data show that Tongluo Xingnao Effervescent Tablets can enhance the cognitive function of SAMP8 through regulating Nampt/SIRT1/FOXO3 signal pathway.

OBJECTIVE:To study the pharmacokinetics and relative bioavailability of two kinds of preparations of valaciclovir hydrochloride.METHODS:This was a randomized two-way cross-over study.A total of 18 healthy volunteers were randomly assigned to receive single oral dose of test valaciclovir tablets(trial group) or reference valaciclovir tablets(control group),with the plasma aciclovir concentrations determined by HPLC,the pharmacokinetics parameters calculated and the relative bioavailability evaluated using 3p97 program.RESULTS:The main pharmacokinetic parameters test and reference valaciclovir hydrochloride tablets were as follows:AUC0～ 24:(12.85? 4.32) vs.(12.19? 4.63) ?g? h? mL-1;AUC0～ ∞:(14.65? 5.75) vs.(13.27? 5.03) ?g? h? mL-1;Cmax:(3.55? 0.92) vs.(3.71? 0.97) ?g? mL-1;tmax:(1.44? 0.43) h vs.(1.33? 0.37) h;t1/2:(6.23? 2.81) h and(4.55? 1.84) h.The relative bioavailability of the test valaciclovir tablets were(111.01? 23.52) %.CONCLUSION:The reference valaciclovir tablets and the test valaciclovir tablets were bioequivalent.
